Two Indian water buffalo (Bubalus bubalis), experimentally infected with Theileria lawrencei (Serengeti) by inoculation of stabilate material, died from acute theileriosis. Macroschizonts were first detected in both buffalo in the peripheral lymph node regional to the site of inoculation 8 days later. No microschizonts were seen in either animal and intraerythrocytic theilerial piroplasms were only detected in one in very low numbers. Both buffalo had marked febrile responses and died on Days 15 and 16 after exposure, when only low numbers of lymphoid cells were parasitized with macroschizonts, the vast majority of which contained few nuclei. Both buffalo showed evidence of terminal leucopenia and thrombocytopenia. On post mortem both animals showed generalised hyperplasia and edema of all lymphoid tissues, gross edema of the lungs, and hemorrhages in the heart and kidneys. T. lawrencei (Serengeti) infections of water buffalo and of cattle are compared and shown to be very similar.  相似文献

A mass spectrometric 16O2/18O2-isotope technique was used to analyse the rates of gross O2 evolution, net O2 evolution and gross O2 uptake in relation to photon fluence rate by Dunaliella tertiolecta adapted to 0.5, 1.0, 1.5, 2.0 and 2.5 M NaCl at 25°C and pH 7.0.At concentrations of dissolved inorganic carbon saturating for photosynthesis (200 M) gross O2 evolution and net O2 evolution increased with increasing salinity as well as with photon fluence rate. Light compensation was also enhanced with increased salinities. Light saturation of net O2 evolution was reached at about 1000 mol m-2s-1 for all salt concentrations tested. Gross O2 uptake in the light was increased in relation to the NaCl concentration but it was decreased with increasing photon fluence rate for almost all salinities, although an enhanced flow of light generated electrons was simultaneously observed. In addition, a comparison between gross O2 uptake at 1000 mol photons m-2s-1, dark respiration before illumination and immediately after darkening of each experiment showed that gross O2 uptake in the light paralleled but was lower than mitochondrial O2 consumption in the dark.From these results it is suggested that O2 uptake by Dunaliella tertiolecta in the light is mainly influenced by mitochondrial O2 uptake. The host response to infection and tissue susceptibility of larval and adult Japanese beetles,Popillia japonica Newman, to the microsporidium,Ovavesicula popilliae Andreadis & Hanula, are reported. The normally transparent Malpighian tubules of Japanese beetle larvae, were hypertrophied and white in color when infected withO. popilliae, a microsporidian which also infects larval fat body, epidermis and pericardial cells. In addition to these tissues, œnocytes and tracheal epithelial cells were also infected in adults. Adult and larval reactions to infection included hypertrophied cells and melanization of the pericardium, but only larvae exhibited an intense inflammatory response. The discoloration of the pericardium most likely resulted from an accumulation of melanin.   相似文献

Abstract 1. The use of flowering vegetation has been widely advocated as a strategy for providing parasitoids and predators with nectar and pollen. However, their herbivorous hosts and prey may exploit floral food sources as well. 2. Previous laboratory studies have shown that not all flower species are equally suitable in providing accessible nectar. Relatively little is known about actual nectar exploitation under field conditions. 3. The present study investigates nectar exploitation by the pest, Plutella xylostella, and its parasitoid, Diadegma semiclausum, under field conditions and examines whether floral nectar exploitation in the field can be predicted based on controlled laboratory studies. 4. Insects were collected from fields bordered by flowering margins containing Fagopyrum esculentum, Lobularia maritima, Anethum graveolens, Centaurea jacea or the grass Lolium perenne (control). Whole insect bodies were individually assayed by HPLC to establish their sugar profile as a measure of the level of energy reserves and the degree of food source use. 5. The average overall sugar content of P. xylostella and D. semiclausum collected in fields bordered by flowering margins was significantly higher than those of individuals collected from grass‐bordered control plots. To the authors’ knowledge, this represents the first demonstration that nectar‐providing plants enhance the energetic state of herbivores under field conditions. 6. In contrast to earlier laboratory studies, the present study did not find elevated sugar contents in P. xylostella and D. semiclausum individuals collected from fields bordered by buckwheat (F. esculentum). 7. The present study shows widespread sugar feeding by both the herbivore and its parasitoid. It also shows that laboratory studies establishing nectar exploitation under controlled conditions can not always be extrapolated to actual exploitation under field conditions. This emphasises the importance of studying field‐collected insects with regard to food source use and nutritional status.  相似文献

Bacterial diversity in sediments obtained along the Chilean margin from areas containing methane seeps, and a hydrate mound were explored by cloning and sequencing and multitag pyrosequencing (MTPS). These libraries were statistically compared to determine the robustness of taxonomic assignment derived from multiplexed pyrosequencing strategies targeting variable regions V1 and V2 of the small subunit rRNA gene for environmental studies. There was no statistical difference in the composition of the libraries, thus, MTPS was utilized to describe diversity in three geochemical zones in these environments. Unidentified Cyanobacteria isolates were abundant in the sulfate reduction zone (SRZ), Deltaproteobacteria were concentrated at the sulfate methane transition zone (SMTZ) and Chloroflexi/GNS dominated methanogenesis zone (MGZ). Although there was variation among specific groups, communities in the SRZ and MGZ did not differ significantly. However, the community dominated by Deltaproteobacteria differentiates the SMTZ from the other zones. Supplemental materials are available for this article. The traditional “safety factor”; method has been used for years to establish occupational exposure limits (OELs) for active ingredients used in drugs. In the past, a single safety factor was used to address all sources of uncertainty in the limit setting process. The traditional 100‐fold safety factor commonly used to derive an acceptable daily intake value incorporates a default factor of 10 each to account for interindividual variability and interspecies extrapolation. Use of these defaults can lead to overly conservative health‐based limits, especially when they are combined with other (up to 10‐fold) factors to adjust for inadequacies in the available database. In recent years, attempts have been made to quantitate individual sources of uncertainty and variability to improve the scientific basis for OELs. In this paper we discuss the science supporting reductions in the traditional default uncertainty factors. A number of workplace‐specific factors also support reductions in these factors. Recently proposed alternative methodologies provide a framework to make maximum use of preclinical and clinical information, e.g., toxicokinetic and toxicodynamic data, to reduce uncertainties when establishing OELs for pharmaceutical active ingredients.  相似文献

The common nettle (Urtica dioica L.) is a perennial and cosmopolitan plant species and is known to be the source of food for a great diversity of insects. To understand the importance of the nettle in agro-ecosystems, a field experiment was carried out in an experimental farm at Gembloux (Belgium) to study the effect of nettle margin strips on aphid and aphidophagous populations in close field crops, namely wheat (Triticum aestivum L.), green pea (Pisum sativum L.) and rape (Brassicae napus L.). The aphids and related beneficial populations were weekly assessed, from March to August 2005, by visual observations in two plots per field crop. A higher abundance of aphidophagous beneficials was collected in nettle strips when compared to the field crops. Particularly, the presence of predatory anthocorids, mirids and green lacewings was observed on nettle only. Nevertheless, the most abundant aphid predatory family, the Coccinellidae, was distributed in both environments, in nettle strips and in crop fields. The field margin supported a significantly higher density ofHarmonia axyridis than the field crops. In contrast, the field crops, green pea particularly, supported a higher density of Coccinella septempunctata. The distribution of the aphidophagous species, mainly the ladybirds, was discussed in relation to the host plant and related aphid species and their potential effect on integrated pest management.  相似文献

Water motion drives the flux of suspended and dissolved material (e.g., nutrients, gametes, and dissolved oxygen) to and from macrophyte canopies, and is one of the most important mechanisms that can regulate the growth, survival, and persistence of marine macrophytes populations. At small spatial scales (e.g., lamina or leaves and individuals), increasing flow‐rates have been demonstrated to enhance physiological processes, especially photosynthesis rates, and we expected a similar response at the canopy scale. We conducted seven experiments over 25 days using a pair of open‐air flow‐chambers under natural light, temperature, and seawater conditions. In the four marine macrophyte (Sargassum piluliferum, S. siliquastrum, S. thunbergii, and Zostera marina) canopies examined, an increase in flow‐rate did not enhance photosynthesis rates. The odds that daily gross photosynthesis rates increase with a decrease in flow‐rates was 1.77 to 1. We also examined if two non‐linear equations and one linear equation, often used to describe the relationship between photosynthesis to photosynthetic photon flux density (PPFD), biased estimates of the daily rates of photosynthesis and respiration. It was revealed that the functional form of the equation strongly influenced photosynthesis and respiration rate estimates at short time scales (i.e., minutes), however, daily rates were insensitive to the type of equation used to model the relationship between photosynthesis and PPFD. We suggest that the predominance of photosynthesis rates occurring in under‐saturating PPFD conditions (> 40 % of daylight hours) may be one of the reasons for this insensitivity.  相似文献

Precipitation pulses play an important role in regulating ecosystem carbon exchange and balance of semiarid steppe ecosystems. It has been predicted that the frequency of extreme rain events will increase in the future, especially in the arid and semiarid regions. We hypothesize that large rain pulses favor carbon sequestration, while small ones cause more carbon release in the semiarid steppes. To understand the potential response in carbon sequestration capacity of semiarid steppes to the changes in rain pulse size, we conducted a manipulative experiment with five simulated rain pulse sizes (0, 5, 10, 25, and 75 mm) in Inner Mongolia steppe. Our results showed that both gross ecosystem productivity (GEP) and ecosystem respiration (Re) responded rapidly (within 24 h) to rain pulses and the initial response time was independent of pulse size. However, the time of peak GEP was 1–3 days later than that of Re, which depended on pulse size. Larger pulses caused greater magnitude and longer duration of variations in GEP and Re. Differences in the response time of microbes and plants to wetting events constrained the response pattern of heterotrophic (Rh) and autotrophic (Ra) components of Re following a rain event. Rh contributed more to the increase of Re in the early stage of rain pulse response, while Ra played an more important role later, and determined the duration of pulse response, especially for large rain events of >10 mm. The distinct responses of ecosystem photosynthesis and respiration to increasing pulse sizes led to a threshold in rain pulse size between 10 and 25 mm, above which post wetting responses favored carbon sequestration. The disproportionate increase of the primary productivity of higher plants, compared with those in the activities of microbial decomposers to larger pulse events suggests that the carbon sequestration capacity of Inner Mongolia steppes will be sensitive to changes in precipitation size distribution rather than just precipitation amount.  相似文献

In desert ecosystems, microbial activity and associated nutrient cycles are driven primarily by water availability and secondarily by nutrient availability. This is especially apparent in the extremely low productivity cold deserts of the McMurdo Dry Valleys, Antarctica. In this region, sediments near streams and lakes provide the seasonally wet conditions necessary for microbial activity and nutrient cycling and thus transfer energy to higher organisms. However, aside from a few studies of soil respiration, rates of microbial activity throughout the region remain unexplored. We measured extracellular enzyme activity potentials (alkaline phosphatase, leucine-aminopeptidase, beta-glucosidase, phenol oxidase, and peroxidase) in soils adjacent to lakes and streams, expecting activity to be primarily related to soil water content, as well as time of season and organic matter supply. Phosphatase and beta-glucosidase activities were higher in shoreline than upland soils; however, potential rates were not correlated with soil water content. Instead, soil organic matter, salinity, and pH were the best predictors of microbial activity. Microbial nutrient limitation metrics estimated from extracellular enzyme activity were correlated with pH and salinity and exhibited similar patterns to previously published trends in soil P and N content. Compared to other terrestrial ecosystems, organic matter specific rates for leucine-aminopeptidase and oxidative enzyme activities were high, typical of alkaline desert soils. Phosphatase activity was close to the global mean whereas beta-glucosidase activity was extremely low, which may reflect the lack of vascular plant derived organic matter in the Dry Valleys. In this cold desert ecosystem, water availability promotes microbial activity, and microbial nutrient cycling potentials are related to soil geochemistry.
