Green fluorescent protein (GFP) possesses a unique folding landscape with a dual basin leading to the hysteretic folding behavior observed in experiment. While theoretical data do not have the resolution necessary to observe details of the chromophore during refolding, experimental results point to the chromophore as the cause of the observed hysteresis. With the use of NMR spectroscopy, which probes at the level of the individual residue, the hysteretic intermediate state is further characterized in the context of the loosely folded isomerized native-like state {Niso} predicted in simulation. In the present study, several residues located in the lid of GFP indicate heterogeneity of the native states. Some of these residues show chemical shifts when the native-like intermediate {Niso} responsible for GFP's hysteretic folding behavior is trapped. Observed changes in the chromophore are consistent with increased flexibility or isomerization in {Niso} as predicted in recent theoretical work. Here, we observed that multiple chromophore environments within the native state are averaged in the trapped intermediate, linking chromophore flexibility to mispacking in the trapped intermediate. The present work is experimental evidence for the proposed final “locking” mechanism in GFP folding forming an incorrectly or loosely packed barrel under intermediate (hysteretic) folding conditions. 相似文献

The integrin family of heterodimeric cell adhesion molecules exists in both low- and high-affinity states, and integrin activation requires binding of the talin FERM (four-point-one, ezrin, radixin, moesin) domain to membrane-proximal sequences in the β-integrin cytoplasmic domain. However, it has recently become apparent that the kindlin family of FERM domain proteins is also essential for talin-induced integrin activation. FERM domains are typically composed of F1, F2, and F3 domains, but the talin FERM domain is atypical in that it contains a large insert in F1 and is preceded by a previously unrecognized domain, F0. Initial sequence alignments showed that the kindlin FERM domain was most similar to the talin FERM domain, but the homology appeared to be restricted to the F2 and F3 domains. Based on a detailed characterization of the talin FERM domain, we have reinvestigated the sequence relationship with kindlins and now show that kindlins do indeed contain the same domain structure as the talin FERM domain. However, the kindlin F1 domain contains an even larger insert than that in talin F1 that disrupts the sequence alignment. The insert, which varies in length between different kindlins, is not conserved and, as in talin, is largely unstructured. We have determined the structure of the kindlin-1 F0 domain by NMR, which shows that it adopts the same ubiquitin-like fold as the talin F0 and F1 domains. Comparison of the kindlin-1 and talin F0 domains identifies the probable interface with the kindlin-1 F1 domain. Potential sites of interaction of kindlin F0 with other proteins are discussed, including sites that differ between kindlin-1, kindlin-2, and kindlin-3. We also demonstrate that F0 is required for the ability of kindlin-1 to support talin-induced αIIbβ3 integrin activation and for the localization of kindlin-1 to focal adhesions. 相似文献

Background information. The Plasmodium parasite, during its life cycle, undergoes three phases of asexual reproduction, these being repeated rounds of erythrocytic schizogony, sporogony within oocysts on the mosquito midgut wall and exo‐erythrocytic schizogony within the hepatocyte. During each phase of asexual reproduction, the parasite must ensure that every new daughter cell contains an apicoplast, as this organelle cannot be formed de novo and is essential for parasite survival. To date, studies visualizing the apicoplast in live Plasmodium parasites have been restricted to the blood stages of Plasmodium falciparum. Results. In the present study, we have generated Plasmodium berghei parasites in which GFP (green fluorescent protein) is targeted to the apicoplast using the specific targeting sequence of ACP (acyl carrier protein), which has allowed us to visualize this organelle in live Plasmodium parasites. During each phase of asexual reproduction, the apicoplast becomes highly branched, but remains as a single organelle until the completion of nuclear division, whereupon it divides and is rapidly segregated into newly forming daughter cells. We have shown that the antimicrobial agents azithromycin, clindamycin and doxycycline block development of the apicoplast during exo‐erythrocytic schizogony in vitro, leading to impaired parasite maturation. Conclusions. Using a range of powerful live microscopy techniques, we show for the first time the development of a Plasmodium organelle through the entire life cycle of the parasite. Evidence is provided that interference with the development of the Plasmodium apicoplast results in the failure to produce red‐blood‐cell‐infective merozoites. 相似文献

Cancer metastasis is the life‐threatening aspect of cancer and is usually resistant to standard treatment. We report here a targeted therapy strategy for cancer metastasis using a genetically‐modified strain of Salmonella typhimurium. The genetically‐modified strain of S. typhimurium is auxotrophic for the amino acids arginine and leucine. These mutations preclude growth in normal tissue but do not reduce bacterial virulence in cancer cells. The tumor‐targeting strain of S. typhimurium, termed A1‐R, and expressing green fluorescent protein (GFP), was administered to both axillary lymph and popliteal lymph node metastasis of human pancreatic cancer and fibrosarcoma, respectively, as well as lung metastasis of the fibrosarcoma in nude mice. The bacteria were delivered via a lymphatic channel to target the lymph node metastases and systemically via the tail vein to target the lung metastasis. The cancer cells expressed red fluorescent protein (RFP) in the cytoplasm and GFP in the nucleus linked to histone H2B, enabling color‐coded real‐time imaging of the bacteria targeting the metastatic tumors. After 7–21 days of treatment, the metastases were eradicated without the need of chemotherapy or any other treatment. No adverse effects were observed. This new strategy demonstrates the clinical potential of targeting and curing cancer metastasis with engineered bacteria without the need of toxic chemotherapy. J. Cell. “Environmentally superior technology” (EST) represents a research initiative in North Carolina (NC) to develop alternatives to lagoon treatment and land application of swine manure. EST performance standards included impacts of animal waste to surface and groundwater, emission of ammonia and odor, release of disease-transmitting pathogens, and heavy metal contamination of soil and groundwater. Five technologies were shown to meet these standards: a solids separation/nitrification–denitrification/soluble phosphorus removal system; a thermophilic anaerobic digester system; a centralized composting system; a gasification system; and a fluidized bed combustion system. Economic data compiled for all EST showed annualized (10-year) costs of retrofitting existing swine farms with the technologies ranged between $90 and over $400 per 1000 lbs. steady state live-weight. Value-engineering to reduce the costs of targeted EST has been successful. Policy providing institutional incentives to incorporate EST has been enacted as a result of this study. 相似文献

A new method for suspending a fine, activated carbon catalyst in a biomass feedstock used for a supercritical water gasification (SCWG) continuous reactor is proposed. In a previous study, the organic matter in poultry manure was shown to be completely converted into gases such as H2, CO2 and CH4 using SCWG. In practice, however, since the feedstock is not only composed of organic matter, but also contains inorganic material, water and catalyst, products such as gas, solid and liquid are produced during SCWG. The aim of this work was to investigate SCWG by-product utilization. This paper describes fundamental studies on the recovery of materials from SCWG products to develop a novel and simple recycling process that utilizes the by-products. A major portion of the activated carbon, monetite and ammonium sulfate can be isolated from the SCWG effluent. 相似文献

Slow pyrolysis or carbonization promotes the conversion of animal manures such as swine manure into charcoal. In this paper, the carbonizing kinetics of swine solids taken from different treatment stages were investigated with a thermogravimetric analyzer. Compared to their biologically stabilized counterpart (lagoon sludge) with an activation energy of 160 kJ mol−1, the activation energies for fresh swine solid samples such as homogenized flushed manure and dewatered solids were much lower between 92 and 95 kJ mol−1. Compared to the kinetics of first order decomposition of cellulose, the pyrolytic decomposition of the swine manures were more complex with the reaction orders varying at 3.7 and 5.0. The two different mathematical methods employed in this paper yielded the similar values of activation energy (E) and pre-exponential factor (A), confirming the validity of these methods. The results of this study provide useful information for development of farm-scale swine solid carbonization process. 相似文献
