Ligation of sphingosine 1-phosphate (S1P) to a set of specific receptors named S1P receptors (S1PRs) regulates important biological processes. Although the ability of S1P to increase cytosolic Ca2+ in various cell types is well known, the role of the individual S1PRs has not been fully characterized. Here, we provide a complete analysis of S1P-dependent intracellular Ca2+ homeostasis in HeLa cells. Overexpression of S1P2, or S1P3, but not S1P1, leads to a significant increase in cytosolic and mitochondrial [Ca2+] in response to S1P challenge. Moreover, cells ectopically expressing S1P2, or S1P3 exhibited an appreciable decrease of the free Ca2+ concentration in the endoplasmic reticulum, dependent on stimulation of receptors by S1P endogenously present in the culture medium which was accompanied by a reduced susceptibility to C2-ceramide-induced cell death. These results demonstrate a differential contribution of individual S1PRs to Ca2+ homeostasis and its possible implication in the regulation of cell survival.  相似文献

城市公园绿地水、热与CO2通量观测与分析   总被引:5,自引:1,他引:5  
水、热和CO2通量是评价城市绿地生态效益的重要指标。在北京海淀公园中部和边缘架设涡度相关系统,连续观测和定量研究城市公园绿地影响下的通量变化特征。结果表明,公园绿地所获得的净辐射在植被生长季节大部分用于植被的蒸散作用,潜热大于显热;而在植被非生长季节大部分用于显热,潜热数值非常小。晴天显热与潜热的比值在春季随着植被枝叶的生长逐渐减少,到夏季达到最小值约1/3,在秋季随着植被叶片枯黄逐渐变大,冬季达到数倍。公园绿地能量平衡率在52%~83%,普遍存在能量不平衡。公园中部的潜热观测值大于公园边缘,而显热小于公园边缘;公园中部CO2通量日均值为负,公园绿地是CO2汇。公园绿地在植被生长季节具有明显的降温增湿、吸收CO2等生态效应。  相似文献

We report large induction (>65fold increases) of volatile organic compounds (VOCs) emitted from a single leaf of the invasive weed mossy sorrel, Rumex confertus Willd. (Polygonaceae), by herbivory of the dock leaf beetle, Gastrophysa polygoni L. (Coleoptera: Chrysomelidae). The R. confertus VOC blend induced by G. polygoni herbivory included two green leaf volatiles ((Z)-3-hexenal, (Z)-3-hexen-1-yl acetate) and three terpenes (linalool, ß-caryophyllene, (E)-ß-farnesene). Uninjured leaves produced small constitutive amounts of the GLVs and barely detectable amounts of the terpenes. A Y-tube olfactometer bioassay revealed that both sexes of adult G. polygoni were attracted to (Z)-3-hexenal and (Z)-3-hexen-1-yl acetate at a concentration of 300 ng h−1. No significant G. polygoni attraction or repellence was detected for any VOC at other concentrations (60 and 1500 ng h−1). Yet, G. polygoni males and females were significantly repelled by (or avoided) at the highest test concentration (7500 ng h−1) of both GLVs and (E)-ß-farnesene. Mated male and female G. polygoni might be attracted to injured R. confertus leaves, but might avoid R. confertus when VOC concentrations (especially the terpene (E)-ß-farnesene) suggest high overall plant injury from conspecifics, G. viridula, or high infestations of other herbivores that release (E)-ß-farnesene (e.g., aphids). Tests in the future will need to examine G. polygoni responses to VOCs emitted directly from uninjured (constitutive) and injured (induced) R. confertus, and examine whether R. confertus VOC induction concentrations increase with greater tissue removal on a single leaf and/or the number of leaves with feeding injury.  相似文献

Recent data have provided evidence for an unrecognised ancient lineage of green plants that persists in marine deep-water environments. The green plants are a major group of photosynthetic eukaryotes that have played a prominent role in the global ecosystem for millions of years. A schism early in their evolution gave rise to two major lineages, one of which diversified in the world's oceans and gave rise to a large diversity of marine and freshwater green algae (Chlorophyta) while the other gave rise to a diverse array of freshwater green algae and the land plants (Streptophyta). It is generally believed that the earliest-diverging Chlorophyta were motile planktonic unicellular organisms, but the discovery of an ancient group of deep-water seaweeds has challenged our understanding of the basal branches of the green plant phylogeny. In this review, we discuss current insights into the origin and diversification of the green plant lineage.  相似文献

Zanthoxylum armatum and Zanthoxylum bungeanum, known as ‘Chinese pepper’, are distinguished by their extraordinary complex genomes, phenotypic innovation of adaptive evolution and species-special metabolites. Here, we report reference-grade genomes of Z. armatum and Z. bungeanum. Using high coverage sequence data and comprehensive assembly strategies, we derived 66 pseudochromosomes comprising 33 homologous phased groups of two subgenomes, including autotetraploid Z. armatum. The genomic rearrangements and two whole-genome duplications created large (~4.5 Gb) complex genomes with a high ratio of repetitive sequences (>82%) and high chromosome number (2n = 4x = 132). Further analysis of the high-quality genomes shed lights on the genomic basis of involutional reproduction, allomones biosynthesis and adaptive evolution in Chinese pepper, revealing a high consistent relationship between genomic evolution, environmental factors and phenotypic innovation. Our study provides genomic resources and new insights for investigating diversification and phenotypic innovation in Chinese pepper, with broader implications for the protection of plants under severe environmental changes.  相似文献

【背景】在农田生态系统中,土壤微生物与植物互作的机制仍不清楚。【目的】进一步加强对植物-微生物互作的认识,筛选出引起不同反馈作用的关键微生物或微生物类群。【方法】采集豆科绿肥救荒野豌豆(Vicia sativa, V)、十字花科绿肥油菜(Brassica napus, N)和荒坡土壤(remnant prairie, R)驯化的田块土壤0-20 cm作为菌剂在温室进行植物-土壤反馈(plant-soil feedback,PSF)试验。土壤菌剂的接种量为10%,即有90%理化性质一致的灭菌土壤作为背景土,同时设置灭菌土壤菌剂作为对照(CK),种植玉米。每组土壤菌剂处理均分为50 mg/kg高磷(high phosphorus,HP)和5 mg/kg低磷(low phosphorus, LP)两个磷浓度处理。玉米收获后,测定产量和植株地上部磷含量,并取土壤样品进行高通量测序,解析不同养分供给情况下微生物对作物生长的反馈效应。【结果】高磷和土壤反馈效应均促进了玉米的生长。在低磷水平下,V、N和R处理的玉米地上部生物量均高于CK处理,但N处理的玉米地上部生物量增加最多(38%),且增幅显著高...  相似文献

利用多分辨率分割生成3种尺度的城市绿地景观.以50像元和300像元的目标分割值生成小尺度和大尺度的景观影像目标层,利用最近邻法分类两个目标层.对小尺度景观分类后的结果进行分割生成中间尺度的景观影像目标层并进行分类.从3个景观尺度的分类目标层中矢量化提取出绿地信息并计算6个景观指数:多样性、优势度、均匀度和分维数、破碎度和内缘比.多样性、优势度、均匀度和分维数的最大值分别为2.2、0.681、0.948和0.326,最小分别为1.641、0.122、0.707和0.113.表明多样性、均匀度和破碎度随着景观尺度的增大而减小,优势度随着景观尺度的增大而增大.基于分割产生多尺度景观的方法可以满足城市绿地景观研究的需求.  相似文献

Green fluorescent protein (GFP) is the most commonly used reporter of expression in cell biology despite evidence that it affects the cell physiology. The molecular mechanism of GFP-associated modifications has been largely unexplored. In this paper we investigated the proteome modifications following stable expression of GFP in breast cancer cells (MDA-MB-231). A combination of three different proteome analysis methods (2-DE, iTRAQ, label-free) was used to maximise proteome coverage. We found that GFP expression induces changes in expression of proteins that are associated with protein folding, cytoskeletal organisation and cellular immune response. In view of these findings, the use of GFP as a cell reporter should be carefully monitored.  相似文献
