Immunogold labeling was used to study the distribution of (1 → 3)-β-glucans and (1 → 3, 1 → 4)-β-glucans in the rice grain during cellularization of the endosperm. At approximately 3–5 d after pollination the syncytial endosperm is converted into a cellular tissue by three developmentally distinct types of wall. The initial free-growing anticlinal walls, which compartmentalize the syncytium into open-ended alveoli, are formed in the absence of mitosis and phragmoplasts. This stage is followed by unidirectional (centripetal) growth of the anticlinal walls mediated by adventitious phragmoplasts that form between adjacent interphase nuclei. Finally, the periclinal walls that divide the alveoli are formed in association with centripetally expanding interzonal phragmoplasts following karyokinesis. The second and third types of wall are formed alternately until the endosperm is cellular throughout. All three types of wall that cellularize the endosperm contain (1 → 3)-β-glucans but not (1 → 3, 1 → 4)-β-glucans, whereas cell walls in the surrounding maternal tissues contain considerable amounts of (1 → 3, 1 → 4)-β-glucans with (1 → 3)-β-glucans present only around plasmodesmata. The callosic endosperm walls remain thin and cell plate-like throughout the cellularization process, appearing to exhibit a prolonged juvenile state. 对近20年来黄淮冬麦育成推广的53个小麦品种的测定结果表明,参试品种容重平均值776.1kg/h,硬度平均值47%,出粉率平均值60.3%。多数品种测定值 于国外对照品种,但亘个别品种优于最好的对照品种,如N82-10,小偃6号,D10-2和冀84-5103等;出粉率和容重有极显著正相关性、出粉率为HMW-谷蛋白亚基组成的回归方程显著,利用该方程预测出粉率效果良好。  相似文献

The cellular pathway of sucrose transfer from the endosperm cavity to the starchy endosperm of developing grains of wheat (Triticum turgidum) has been elucidated. The modified aleurone and sub-aleurone cells exhibit a dense cytoplasm enriched in mitochondria and endoplasmic relicilium. Significantly, the sub-aleurone cells are characterized by secondary wall ingrowths. Numerous plasmodesmata interconnect all cells between the modified aleurone and starchy endosperm. The pro-tonophore carbonylcyanide-m-chlorophenyl hydrazone (CCCP) slowed [14C]sucrose uptake by grain tissue slices enriched in modified aleurone and sub-aleurone cells but had no effect on uptake by the starchy endosperm. The fluorescent weak acid sulphorhodamine G (SRG) was preferentially accumulated by the modified aleurone and sub-aleurone cells, and this uptake was sensitive to CCCP. The combined plasma membrane surface areas of the modified aleurone and sub-aleurone cells appeared to be sufficient to support the in vivo rates of sucrose transfer to the starchy endosperm. Plasmolysis of intact excised grain inhibited [14C]sucrose transfer from the endosperm cavity to the starchy endosperm. The sulphydryl group modifier p-chloromercuribenzenesulphonie acid (PCMBS) decreased [14C]sucrose uptake by the modified aleurone and sub-aleurone cells but had little effect on uptake by the starchy endosperm. In contrast, when PCMBS and [14C]sucrose were supplied to the endosperm cavity of intact excised grain, PCMBS slowed accumulation by all tissues equally. Estimates of potential sucrose fluxes through the interconnecting plasmodesmata were found to be within the published range. It is concluded that the bulk of sucrose is accumulated from the endosperm cavity by the modified aleurone and sub-aleurone cells and subsequently transferred through the symplast to the starchy endosperm.  相似文献

The hypothesis we propose is that during photosynthesis the balance between potentially detrimental and beneficial photochemically induced events can be tipped beneficially toward increased photosynthesis and toward increased crop yield. To test this hypothesis a procedure has been devised with the rice plant, Oryza sativa, that has resulted in increasing both canopy photosynthesis and rice grain yield. Two elite rice varieties selected independently in the contrasting environments of either South China or Texas, each with distinct photosynthetic traits, were crossed to produce a hybrid with an increased canopy photosynthesis and grain yield that is regularly 20 to 22% higher than the mid-yields of the parents. The photosynthetic and mechanisms which may contribute to these beneficial results in the hybrid rice are: a reduction of the midday depression of photosynthesis; a rapid development of the canopy for photosynthetic light interception and an increased canopy photosynthesis; increased amounts of carotenoids for the xanthophyll cycle; an increased protection against free radicals induced by paraquat treatment; a 6 to 12 day shorter plant reproductive life cycle; and a 8 to 10 day increase in the longevity of the flag leaf over the parents. While the hybrid rice has successfully integrated these and likely other unknown characteristics to increase both crop photosynthesis and grain yield, we propose that understanding the underlying beneficial photosynthetic mechanisms supporting these crop plant traits is worthy of thorough investigation and application in crop production.Dedicated to the memory of Professor D.I. The maize weevil, Sitophilus zeamais (Motchulsky) (Coleoptera: Curculionidae), and the Angoumois grain moth, Sitotroga cerealella (Olivier) (Lepidoptera: Gelechiidae), are important insect pests of stored products. The coexistence in nature of the two species on maize has been difficult to explain from laboratory set‐ups, as the moth is inevitably eliminated by S. zeamais. However, early laboratory experiments ignored the spatial dimension, while several studies have lately revealed that two competing species may coexist in a spatially divided environment even though one of the species is competitively superior in a simpler universe. Two hypotheses for coexistence were examined by studying the interactions in a system where the resources were spatially divided. It was found that spatial subdivision in the current experiment was insufficient to ensure coexistence between the two species, so neither of the hypotheses could be confirmed. A simple model describing the population dynamics of S. zeamais was fitted to the data. Its parameters provide important information about how resource depletion influences the per capita growth rate. Finally, the study indicates that the mechanism responsible for S. zeamais's rapid elimination of S. cerealella is the superiority of the former species to colonize and monopolize new patches. The design of this study may serve as a template for further laboratory experiments revealing the effect of a spatially divided environment on competitive interactions. However, the present study also indicates that choosing the appropriate spatial scale may be critical for relating the results to natural environments.  相似文献

ABSTRACT. The characteristic grain size distributions observed in the test walls of agglutinated foraminifera can be replicated by numerical simulations. the logistics employed by the model parallel those seen in foraminifera during culturing experiments of test construction. Both fractal and log-normal grain distributions are generated by a simple space-filling algorithm. However, to generate the specific grain distributions observed in foramimferan tests a strong preference for selecting larger grains from the available sediment must be effected. the exclusion of smaller grains, during selection, produces log-normal grain distributions similar to those previously observed. These and similar modelling techniques may provide valuable insights into the mechanisms of shell construction employed by protists.  相似文献

The microclimate in facilities for studying effects of elevated CO2 on crops differs from ambient conditions. Open-top chambers (OTCs) increase temperature by 1–3 °C. If temperature and CO2 interact in their effect on crops, this would limit the value of OTC experiments. Furthermore, interaction of CO2 and temperature deserves study because increases in atmospheric CO2 concentration are expected to cause global warming. This paper describes two experiments in which a recently developed cooling system for OTCs was used to analyse the effects of temperature on photosynthesis, growth and yield of spring wheat (Triticum aestivum L., cv. Minaret). Two levels of CO2 were used (350 and 700 ppm), and two levels of temperature, with cooled OTCs being 1.6–2.4 °C colder than noncooled OTCs. Photosynthetic rates were increased by elevated CO2, but no effect of temperature was found. Cross-switching CO2 concentrations as well as determination of A–Ci curves showed that plant photosynthetic capacity after anthesis acclimated to elevated CO2. The acclimation may be related to the effects of CO2 on tissue composition: elevated CO2 decreased leaf nitrogen concentrations and increased sugar content. Calculations of the seasonal mean crop light-use efficiency (LUE) were consistent with the photosynthesis data in that CO2 increased LUE by 20% on average whereas temperature had no effect. Both elevating CO2 and cooling increased grain yield, by an average of 11% and 23%, respectively. CO2 and temperature stimulated yield via different mechanisms: CO2 increased photosynthetic rate, but decreased crop light interception capacity (LAI), whereas cooling increased grain yield by increasing LAI and extending the growing season with 10 days. The effects of CO2 and temperature were not additive: the CO2 effect was about doubled in the noncooled open-top chambers. In most cases, effects on yield were mediated through increased grain density rather than increased individual grain weights. The higher growth response to elevated CO2 in noncooled vs. cooled OTCs shows that a cooling system may remove a bias towards overestimating crop growth response to CO2 in open-top chambers.  相似文献

A previous study of Campanula punctata pollinated by Bombus diversus showed greater male reproductive success in plants with wider corollas when male reproductive success was measured by paternity analysis. Pollen removal alone was not a good measure of male reproductive success. The aim of the current study is to elucidate the mechanism underlying the selection on corolla width in C. punctata. Multiple regression analysis of male reproductive success in experimental populations revealed that the advantage of a wide corolla to the male is independent of the corolla width of pollen recipients, indicating that the position of pollen deposited on Bombus may not be an important factor for selection. However, the number of pollen grains that fell on the petal or off the flower and were lost at the first visit of Bombus (pollen fall) decreased with increasing corolla width, so that most pollen grains removed from a wide flower were deposited onto the pollinator. In a narrow flower, a greater proportion of the pollen removed was wasted through pollen fall. Pollen removal does not reflect male reproductive success, at least in part because of the loss of pollen in some flowers through pollen fall. The positive relationship between wider corollas and reduced loss of pollen through pollen fall is likely to lead to greater male reproductive success of wider flowers under repeated visits of pollinators. Decreased pollen fall, probably due to more gentle contact between pollen and bees in flowers with wider corollas, may be one of the mechanisms underlying the selection on corolla width of C. punctata.  相似文献

Stability of grain yield performance is an important characteristic in the selection of new crop cultivars. Information from cultivar trials, however, is seldom fully analysed for genotype by environment interactions and, therefore, information on stability of current crop cultivars is lacking. The objectives of this study were to investigate the stability of agronomic traits among genotypes of barley (Hordeum vulgare) across 17 environments (location‐years) in Maryland (USA) from 1994 through 1997 and to examine the effect of locations and years of testing on grain yield performance in this region. Significant differences were observed among barley cultivars and experimental lines for grain yield, plant height, and heading date. Grain yield was positively correlated with plant height and negatively correlated with heading date. Genotype x environment interactions measured through regression analysis were significant for grain yield, heading date and plant height, with the environmental component having the largest effect. Most barley genotypes tested (90%) had regression slopes for grain yield that did not differ from 1.0, indicating good potential for yield response under improving environmental conditions. The most widely grown cultivar in the mid‐Atlantic region, ‘Nomini’, had a regression slope that was higher than 1.0 for grain yield. This indicates that it tends to respond with increasingly higher yields under favorable conditions. In this study, the slope and the standard error of the slope were moderately correlated with grain yield. The genotype's coefficient of variation was not a good indicator of stability for this region. Grain yields of genotype entries common to all years and locations were correlated with corresponding yields at each of the locations and years to assess the relative performance of each location and year. Correlation coefficients across locations were relatively high (r=0.64) within each year of testing. Correlations between years for the same and across locations were generally much lower. The data presented here supports a testing program over more years rather than increased locations to fully characterise the performance of new cultivars.  相似文献

Northern hybridizations were used to study the site of synthesis of three carboxypeptidases (Cpases I-III) which occur in the starchy endosperm of germinating barley grain ( Hordeum vulgare L.). Further evidence was obtained by studying secretion of these enzymes from scutella or aleurone layers separated from germinating grains. Messenger RNA for Cpase II was detected only in developing grain, and the bulk of the mRNA was localized in the starchy endosperm. This suggests that Cpase II is synthesized at the site of its accumulation, the starchy endosperm. In contrast, Cpase I is expressed during germination and the predominant site of synthesis is the scutellum, from which it is secreted into the starchy endosperm. Cpase III is also synthesized during germination, but the bulk of it is synthesized in and secreted from the aleurone layer. Thus, the three carboxypeptidases, all of which seem to play a role in hydrolysis of the reserve proteins in the starchy endosperm during germination, have different sites of synthesis.  相似文献
