The effects of theophylline (a phosphodiesterase inhibitor) and cAMP on 17α, 20ß-dihydroxy-4-pregnen-3-one-induced germinal vesicle breakdown was investigatedin vitro in catfish (Clarias batrachus) oocytes. Folliculated oocytes incubated with 17α, 20ß-dihydroxy-4-pregnen-3-one at the concentration of 1 μg/ml induced 93.2 ± 2.23% germinal vesicle breakdown. When the oocytes were prestimulated with 17α,20ß-dihydroxy-4-pregnen-3-one for 6 h and then treated with different concentrations of theophylline, there was a significant drop in the frequency of germinal vesicle breakdown at the concentrations 2.0, 1.5 and 1.0 mM. However, theophylline was found to be incapable of inhibiting germinal vesicle breakdown at its lowest concentration (0.5 inM). In the time course study, significant inhibition of germinal vesicle breakdown was recorded when 1 mM theophylline was added up to 30 h of 17α,20ß-dihydroxy-4-pregnen-3-one Stimulation but the inhibitory effect of theophylline gradually (time dependent manner) declined if the stimulatory time of 17α,20ß-dihydroxy-4-pregnen-3-one was increased. A similar inhibition of germinal vesicle breakdown was also recorded with various concentrations of cAMP. Except 0.5 mM, all the higher concentrations of cAMP significantly inhibited 17α,20ß-dihydroxy-4-pregnen-3-one induced germinal vesicle breakdown.  相似文献

Studies were carried out in the laboratory on the influences of time of day, temperature, relative humidity and starvation on flight initiation byProstephanus truncatus (Horn) (Coleoptera: Bostrichidae). Flight occurred throughout the 12 h photophase and at the beginning of the scotophase but peaked at 2–0 h before darkness. Temperature exerted a significant effect on flight. The frequency of flight take-off increased with temperature over the range 20–30°C but declined sharply at 35°C. Flight activity increased with starvation up to a maximum at 2 days after which it began to decline.  相似文献

Distribution and properties of pea (Pisum sativum L.) cells, competent forAgrobacterium-mediated transformation were analysed byin situ histochemical detection of GUS (-glucuronidase) activity, 4 d after inoculation with engineeredAgrobacterium tumefaciens. The vector system consisted of the hypervirulent disarmed strain EHA101 and the binary plasmid pIBGUS, carrying an intron-containing, 35S-promotor drivengusA (oruidA) gene and two selectable marker genes. Cells competent for transformation were mainly restricted to the dedifferentiating cells neighbouring the vascular system of cotyledon and epicotyl explants. A standardized assay was developed, allowing determination and quantification of factors influencing number and distribution of competent cells. In etiolated seedlings, competence for transformation decreased with the distance of the epicotyl explant from the shoot apex and was specifically induced by the exogenous application of auxins. Transient expression ofgusA afterAgrobacterium-mediated DNA transfer was dramatically reduced upon application of cell-cycle and DNA replication inhibitors aphidicolin, colchicine and nalidixic acid. GUS expression after direct DNA transfer of double-stranded plasmid DNA (via PEG into protoplasts or via particle bombardment of epicotyl segments) was independent of cell-division/DNA replication.A GUS-positive mutant of EHA101 was constructed to allowin situ analysis of attaching bacteria within the plant tissue. Attachment and invasion was inhibited by well-developed cuticula but was restored after chloroform treatment of the tissue surface. Moreover, no correlation was found between distribution of attaching bacteria and the pattern of transformation-competent cells.  相似文献

Summary As part of an effort to optimize conditions required for the complete maturation of muscle cells in vitro, we have investigated the effects of the antibiotics penicillin, streptomycin, and Fungizone (amphotericin B) on the development of cultured chick embryo skeletal muscle. It is shown that even low dosages of streptomycin, but not penicillin or Fungizone, retard protein synthesis and accumulation in these cultures. Myosin accumulation was also reduced and the appearance of striations in fused cells was delayed in myotubes formed in medium containing streptomycin. Additional data suggest that this overall retardation of myogenesis is due to the influence of streptomycin on maturing myotubes rather than early proliferation and cell fusion. These results are discussed with regard to recent efforts to promote the full maturation of muscle cells grown in culture. Structural development of grain tissues of maternal origin in normal and seg1 barley (Hordeum vulgare L. cv. Betzes) was examined using light and electron microscopy. Chalaza and seedcoat cells of normal grains developed prominent tannin vacuoles which persisted throughout the grain-filling period. Tannins were present in the same tissues of seg1, but no large central vacuoles developed. Instead, the chalaza and nucellar projection degenerated and were crushed, presumably terminating sugar flow and causing formation of shrunken grains (35–55% normal dry weight). Tannins were localized using various histochemical stains. Extracts of chalaza and adjacent tissues contained proanthocyanidins which yielded delphinidin and cyanidin upon hydrolysis in boiling HCl. We suggest that the basis of the seg1 phenotype may be abnormal compartmentation of tannins causing precipitation of cytoplasmic proteins and early death of chalazal cells.Abbreviations FAA Formalin-acetic acid-ethanol - PAS periodic acid Schiffs reagent  相似文献

This paper considers projections made about world population levels and world food production from 1945 to 1970, attempting to evaluate them in terms of actual levels of 1975. Most underestimated the population explosion of the past two decades; short-term estimates were more reliable than long-term ones; there was little variation in population estimates at any one time; and U. N. figures were generally the most useful. Projections of world food production were less satisfactory than those of population levels, as both data and measures were debatable. The research suggests some of the problems in evaluating the danger of world famine by examining projections of population and food supply mechanically and in isolation from the complex processes which affect these factors.  相似文献

Summary In sexually mature male sticklebacks, the renal tubular cells are transformed from ion reabsorbing to mucus secreting cells and in these fish concomitant changes take place in the glomeruli.The present study compares glomerular structure of immature males in fresh water (controls) to those of mature males in fresh water and to immature male sticklebacks in seawater. Glomerular structure is markedly altered in the latter two groups and the changes are similar to a large extent. In these two groups the renal capsules and glomeruli are smaller and the lumina of the glomerular capillaries decrease in diameter, while the number and size of the endothelial fenestrations are reduced. Mesangial cells proliferate and the mesangial matrix greatly expands in both the centrolobular region and the subendothelial space around the capillaries. The secretory activity of the podocytes is enhanced and is responsible for the observed increase in thickness of the outer layer of the basal lamina, the lamina rara externa. The area covered by the filtration slit membranes is reduced, probably as a consequence of fusion of the pedicels of the podocytes. The permeability characteristics of the glomerular filtration barrier for macromolecules, as studied with ferritin injections, remain unaltered. However, the observed differences point to a reduction of the glomerular filtration rate (GFR) during maturation in male sticklebacks, as well as during adaptation of sticklebacks to seawater. Chronological ages of Alouattaon Barro Colorado Island (BCI) were estimated from longitudinal dental wear. Combining these data with visual censuses, we approximated the study population’s age profile. A stable model was then constructed; from it we derived agespecific mortality rates. Mortality of immature animals is high, with 88% of the males and 65% of the females dying before 5 years. Adult mortality is low until 11 years, when it accelerates. The average adult life span is 16.6 years for males and 15.5 for females. The maximum life span is over 20 years. A pubertal male growth spurt occurs from 3 to 5 years, at which time females are primiparous. Sexual size differences develop primarily during this time. The 1976 age profile had anomalously few animals aged 7 years and males aged 8–9 and 15–16 years. With corroborating evidence, we hypothesize that these deficits resulted from excessive rainfall in 1963 and 1971, which reduced the fall fruit crop and led to a high juvenile mortality. Within half of our study troops, some adult males have nearly identical ages, suggesting an active process of agemate coalition. We hypothesize a form of kin selection, wherein peripheral male cohorts from the same natal troop have greater survival and social success than solitary animals. The stable model suggests an annual growth rate of 1.5% during the 1970s. Estimates of 16.7 and 4% for the previous two decades, following a yellow-fever epidemic, imply that the BCI population is becoming stationary. A very different age structure existed on neighboring Orchid Island in 1976, suggesting general food limitation. By contrast, the lowered, but continuing growth on BCI suggests a less drastic limiting mechanism. We hypothesize that intratroop social competition limits population growth by regulating subadult survival rates during food scarcity cycles.  相似文献

The activities of adenosylhomocysteinase (EC 3.3.1.1) and adenosine nucleosidase (EC 3.2.2.7) were assayed in extracts from yellow lupin (Lupinus luteus L.) cotyledons at different stages of seed formation and seedling development. Adenosylhomocysteinase activity was demonstrated in all the cotyledon extracts examined. Its lowest level was found in the dry seeds and the highest, in 4-day-old seedling cotyledons. Extracts from the cotyledons of maturating seeds, dry seeds, and seedlings up to the second day of growth exhibited no adenosine nucleosidase activity. Adenosine nucleosidase activity appeared in the cotyledons of 2-day-old seedlings and its highest level was reached in 4-to 5-day-old seedlings. There is no inhibitor of adenosine nucleosidase in the maturating and dry yellow lupin seeds. No activator of a possible zymogen form of adenosine nucleosidase from maturating or dry seeds occurs in the growing seedlings.  相似文献
