The construction and operation of a hand-operated box sampler is described. The sampler covers an area of 25 × 25 cm and penetrates into the sediment to the depth of 15 cm. This paper is concerned with modeling the architecture of colonic crypts and the implications of this modeling for understanding possible coordinated response of carcinogen-induced DNA damage between various regions of the colon. The methods we develop to address these two issues are applied to a particular important example in colon carcinogenesis. We cast the problem as an unusual and not previously studied hierarchical mixed-effects model characterized by completely missing covariates in units at a structurally base level, except for some randomly selected units. Information concerning the missing covariates is available through certain known ordering constraints and surrogate measures. Our methods use Bayesian machinery. We exploit the biological structure of this problem to generate the missing covariates simultaneously and efficiently at the base levels, as opposed to the naive practice of generating units at the base levels one-at-a-time with Metropolis-Hastings steps. We apply our methods to show that different regions of the colon have different architectures, and to estimate an important but non-standard function that measures the interrelationship of DNA damage mechanisms in different regions of the colon. 相似文献

A volumetric aeropalynological sampling was carried out for the first time in MontevideoUruguay, from October 2000 to September 2001, using a Rotorod sampler Model 40. During the year 76 pollen types were identified. Airborne pollen was recorded over the year but a maximum pollen period was observed from August to April. For the rest of the months, pollen concentration was below 1% of the total annual pollen (TP). The pollen spectrum was characterized by the dominance of herbaceous pollen (NAP), which represented 68% of the TP and dominates the spectrum from November to March. Poaceae was the most frequent and abundant pollen type accounting for 45% of TP. The pollen spectrum reflected the floristic diversity of the city and most of the sources of airborne pollen are present in local and regional flora. Fourteen pollen types reach more than 1% of the TP and most of them are cited as allergenic pollen in other regions. These results may prove important for future medical research. 相似文献

AIMS: To investigate methods for the recovery of airborne bacteria within pig sheds and to then use the appropriate methods to determine the levels of heterotrophs and Escherichia coli in the air within sheds. METHODS AND RESULTS: AGI-30 impingers and a six-stage Andersen multi-stage sampler (AMS) were used for the collection of aerosols. Betaine and catalase were added to impinger collection fluid and the agar plates used in the AMS. Suitable media for enumerating E. coli with the Andersen sampler were also evaluated. The addition of betaine and catalase gave no marked increase in the recovery of heterotrophs or E. coli. No marked differences were found in the media used for enumeration of E. coli. The levels of heterotrophs and E. coli in three piggeries, during normal pig activities, were 2.2 x 10(5) and 21 CFU m(-3) respectively. CONCLUSIONS: The failure of the additives to improve the recovery of either heterotrophs or E. coli suggests that these organisms are not stressed in the piggery environment. The levels of heterotrophs in the air inside the three Queensland piggeries investigated are consistent with those previously reported in other studies. Flushing with ponded effluent had no marked or consistent effect on the heterotroph or E. coli levels. SIGNIFICANCE AND IMPACT OF THE STUDY: Our work suggests that levels of airborne heterotrophs and E. coli inside pig sheds have no strong link with effluent flushing. It would seem unlikely that any single management activity within a pig shed has a dominant influence on levels of airborne heterotrophs and E. coli. 相似文献

In regression applications with categorical predictors, interest often focuses on comparing the null hypothesis of homogeneity to an ordered alternative. This article proposes a Bayesian approach for addressing this problem in the setting of normal linear and probit regression models. The regression coefficients are assigned a conditionally conjugate prior density consisting of mixtures of point masses at 0 and truncated normal densities, with a (possibly unknown) changepoint parameter included to accommodate umbrella ordering. Two strategies of prior elicitation are considered: (1) a Bayesian Bonferroni approach in which the probability of the global null hypothesis is specified and local hypotheses are considered independent; and (2) an approach which treats these probabilities as random. A single Gibbs sampling chain can be used to obtain posterior probabilities for the different hypotheses and to estimate regression coefficients and predictive quantities either by model averaging or under the preferred hypothesis. A Bayesian approach to the direct mapping of a quantitative trait locus (QTL), fully utilizing information from multiple linked gene markers, is presented in this paper. The joint posterior distribution (a mixture distribution modeling the linkage between a biallelic QTL and N gene markers) is computationally challenging and invites exploration via Markov chain Monte Carlo methods. The parameter's complete marginal posterior densities are obtained, allowing a diverse range of inferences. Parameters estimated include the QTL genotype probabilities for the sires and the offspring, the allele frequencies for the QTL, and the position and additive and dominance effects of the QTL. The methodology is applied through simulation to a half-sib design to form an outbred pedigree structure where there is an entire class of missing information. The capacity of the technique to accurately estimate parameters is examined for a range of scenarios. 相似文献

A series of inexpensive, pneumatic thin layer water samplers is described. They can be operated from small boats, and permit sampling at 2.5 cm intervals with little or no disturbance of stratified systems such as oxyclines or redoxclines of meromictic lakes, or microstratification of flagellates in sheltered epilimnia. A new hand-operated sampler was developed consisting of a perspex cylinder (thickness 0.5 cm, length 32 cm, diameter 13 cm) cut in half lengthwise. The valves are joined together by means of a piano hinge for opening and closing of the sampler. In each of the sites there are openings covered with a net of 0.18×0.18 mm mesh. The external free edges of the halves are covered with weather stripping. The apparatus allows sampling of epiphytic fauna, or animals swimming around various macrophyte structures, including those trailing on the bottom, and those with floating leaves. 相似文献
