Two methods of aerobiological collection, the Burkard(Hirst-based) and the Cour, were compared using datafrom pollen collected at Bellaterra (Barcelona, Spain)between 1994 and 1996. Results for three pollen taxaof allergenic relevance (Urticaceae, Poaceae andOlea) are presented. This study confirmed thedifficulty of finding a reliable conversion formulabetween these sampling methods, as has been found byseveral other authors.An alternative statistical analysis was carried outusing categorical data, the pollen concentration databeing first converted into an ordinal scale with fivelevels based on local records. We present observations on the variability of sediment penetration depth by the Ponar grab sampler, which lead us to question the reliability of grab samples in the quantitative study of freshwater benthos. Penetration depth of the Petite Ponar grab depends on substrate type, and correlates with the amount of organic carbon, the water content and the granulometry of the sediment. Since these factors can also influence faunal composition and vertical distribution in the sediment, it is important to study the performance of the sampler before a biological explanation for the observed pattern is given. We discuss here the influence of sample size (number of replicates) on the accuracy and precision of the results when sampling profundal benthos with an Ekman grab according to the Finnish standard, SFS 5076, which is equivalent to the Swedish and Norwegian standards. The aim was to find criteria for choosing a sample size which would avoid any powerful influence of chance on the results without entailing an unreasonable amount of work for monitoring purposes.Lake Haukivesi (area 620 km2, total phosphorus 13 µg l–1 and colour 35 Pt mg l–1), Lake Paasivesi (116 km2, 5 µg l–1 and 35 Pt mg l–1) and Lake Puruvesi (322 km2, 4 µg l–1 and 5 Pt mg l–1) were sampled randomly in June and October 1991. 25 Replicate samples were taken on each occasion from the deep profundal area of each lake, defined here as 60-100% of the maximum depth. The sedimentation areas studied were fairly homogeneous, since the animal communities were not markedly affected by the variations in depth. Distribution estimates for the statistics studied, such as number of individuals, expected number of species, diversity and benthic quality indices, were calculated for a large set of random samples taken from the empirical data by computer (bootstrap sampling). The sample variance, s 2, correlated with the mean animal density, m (ind. m–2), according to the equation s 2 = 31.77 m 1.247. The sample size required to achieve the desired precision in mean animal density (D, expressed as the ratio standard error/mean) can thus be estimated as n = 31.77 m –0.753 D –2. The number of replicate samples needed to achieve a standard error of 20% of the mean density was 10 in Lake Haukivesi, seven in Lake Paasivesi and 11 in Lake Puruvesi. The accuracy and precision of the estimated number of species, Shannon's diversity and Benthic Quality Index improved markedly as the sample size was increased to 10 replicates. As a compromise between work load and statistical reliability, a figure of 10 replicate Ekman samples is proposed here for the monitoring of profundal benthos. The proposed sample size usually produces individual numbers which are high enough for practical purposes, probably at least 100 individuals, which is recommended as a minimum in the standard. The lower number of replicate samples recommended in recent Finnish handbook, 3–5, usually produces inadequate data, and this may detract from the comparability of the results and leave the changes in profundal communities undetected.  相似文献

There are practical and valid reasons topresent biological field trial referenceresults as agent containing particles per literof air (ACPLA). However, workers in biologicalaerosol research have a need to know how manyviable individual organisms are contained in asingle particle of a given diameter. Anecdotalevidence may exist suggesting that the task hasbeen accomplished but without a way toreplicate the measurements, it is difficult toaccept unsubstantiated claims. It is verydifficult to declare a finite number thatsatisfies all the experimental requirements, asthe problem is a statistical probability issue. This paper describes a method for estimatingthe number with practical instructions forreplicating the work in other laboratories.The test aerosol was contained in a 90 m3chamber at concentrations as low as 5 ACPLA. Amodified version of slit sampler collectedviable particles. A statistical method was usedto demonstrate sampling predictability at 95%confidence level. By using glass fiber filtersmounted in a dichotomous sampler, samplingefficiencies were estimated for a variety ofcommon aerosol collectors. The accumulated datapermitted the estimation of the number ofviable spores per particle. For a 2.5 to4 µm particle, arguments have beenpresented for considering 4.5 as the mostprobable ACPLA value.  相似文献

Habitat selection models are used in ecology to link the spatial distribution of animals to environmental covariates and identify preferred habitats. The most widely used models of this type, resource selection functions, aim to capture the steady-state distribution of space use of the animal, but they assume independence between the observed locations of an animal. This is unrealistic when location data display temporal autocorrelation. The alternative approach of step selection functions embed habitat selection in a model of animal movement, to account for the autocorrelation. However, inferences from step selection functions depend on the underlying movement model, and they do not readily predict steady-state space use. We suggest an analogy between parameter updates and target distributions in Markov chain Monte Carlo (MCMC) algorithms, and step selection and steady-state distributions in movement ecology, leading to a step selection model with an explicit steady-state distribution. In this framework, we explain how maximum likelihood estimation can be used for simultaneous inference about movement and habitat selection. We describe the local Gibbs sampler, a novel rejection-free MCMC scheme, use it as the basis of a flexible class of animal movement models, and derive its likelihood function for several important special cases. In a simulation study, we verify that maximum likelihood estimation can recover all model parameters. We illustrate the application of the method with data from a zebra.  相似文献

Powdery mildew on roses gives the impression of being an insignificant disease, but for several reasons more attention to the disease is justifiable. Apart from the economic value of the attacked host plant, we have to do with a very interesting combination of host and parasite. Viruses that affect humans, animals and plants are often dispersed and transmitted through airborne routes of infection. Due to current technological deficiencies, accurate determination of the presence of airborne viruses is challenging. This shortcoming limits our ability to evaluate the actual threat arising from inhalation or other relevant contact with aerosolized viruses. To improve our understanding of the mechanisms of airborne transmission of viruses, air sampling technologies that can detect the presence of aerosolized viruses, effectively collect them and maintain their viability, and determine their distribution in aerosol particles, are needed. The latest developments in sampling and detection methodologies for airborne viruses, their limitations, factors that can affect their performance and current research needs, are discussed in this review. Much more work is needed on the establishment of standard air sampling methods and their performance requirements. Sampling devices that can collect a wide size range of virus-containing aerosols and maintain the viability of the collected viruses are needed. Ideally, the devices would be portable and technology-enabled for on-the-spot detection and rapid identification of the viruses. Broad understanding of the airborne transmission of viruses is of seminal importance for the establishment of better infection control strategies.  相似文献

In this paper a comparative study of different adhesive media used with the impaction suction sampler have been done. The sampling was carried out by means of a Burkard spore trap and a double-blind comparison of vaseline, petrolatum white and silicone fluid (silicone/carbon tetrachloride) was done. With the aim to compare the adhesive efficiency with different pollen grain sizes and shapes, some different pollen types were used. Moreover, in order to compare the media under different weather conditions the sampling was done outdoors, in different seasons. The differences in the efficiency of these media where small, but occasionally significant. These differences depend on the climatic conditions as temperature influences vaseline adhesivity. In conclusion, we suggest the utilization of silicone fluid because of its stability to temperature and its easy application.  相似文献
