Hybrids have been obtained by protoplast fusion of penicillin-producing strains of Aspergillus nidulans with strains of A. rugulosus which do not produce the antibiotic at detectable levels. Induced haploidization of the hybrids allowed the recovery of stable segregants, which were screened for penicillin production. Approximately half of the segregants from each cross produced penicillin, indicating segregation of one or several linked genes which are essential for penicillin biosynthesis. These essential genes were found to be located on linkage-group VI of A. nidulans by observing segregation in multi-marked strains. The penicillin-producing segregants showed a wide range of titres. Some segregants, which had recombinant phenotypes, produced significantly more penicillin than the A. nidulans parent.  相似文献

Synteny-mapping horse microsatellite markers using a heterohybridoma panel   总被引:1,自引:0,他引:1  
A panel of horse-mouse heterohybridoma cells was tested for genetic markers using biochemical and polymerase chain reaction-(PCR-) based tests. Biochemical markers included phospho-glucomutase ( PGM ), glucose phosphate isomerase ( GPI ) and 6-phosphogluconate dehydrogenase ( PGD ). Markers detected using PCR-based tests included microsatellite markers HTG2–15, HMS 1–3, 5–8, VHL20, ECA2 and genes for equine major histocompatibility gene ELA-DRA , tumour necrosis factor alpha (TNFA) and transferrin. The results were analysed for correlation and concordance. Based on the results, five synteny groups were identified, specifically between ELA-DRA, TNFA, HMS5 and HTG5 ; between HTG3 and HTG13 ; between HTG4, HTG8 and HMS3 ; between HTG6 and HMS1 ; and between HTG7, HTG9 and HMS6. Evidence was also found for synteny between HTG12, HMS7 and ECA2 , however, confirmation requires further testing. Cytogenetic evaluation of the cell lines making up the panel indicated that large metacentric chromosomes were preferentially lost or tended to break at the centromere. Consequently, the results from this analysis can be used to identify synteny, but not to exclude synteny.  相似文献

A rare phenomenon of the occurrence of novel non-parental chloroplast DNA (cpDNA) variants in natural sexual interspecific hybrids between Populus deltoides var deltoides and P. nigra, P. x canadensis is described. Restriction fragment variation of cpDNA in 17 P. x canadensis cultivars was examined and compared with that of representative samples of P. deltoides and P. nigra using 83 combinations of 16 restriction enzymes and six Petunia hybrida cpDNA probes. Twelve cultivars had one to five novel non-parental cpDNA fragments in the chloroplast genome region homologous to the 9.0-kb PstI cpDNA fragment of Petunia from the large single-copy region.  相似文献

The expression of the adenylate kinase isozymes and of phosphopyruvate hydratase was studied in man-mouse and man-hamster hybrid clones. Concordant segregation of the loci coding for AK-2 and PPH was observed in 54 of 55 primary hybrid clones, and these loci were demonstrated to be synthetic with the loci specifying PGM-1 and PGD. The pattern of expression of the four enzymes in discordant clones suggests the gene order 1pter-(PGD,PPH)-AK-2-PGM-1-centromere. Leaf surfaces of the native Heracleum sphondyliutn , the alien Heracleum mantegazzianum and the interspecific hybrid were examined using SEM and light microscopy. The surfaces differed in hairiness, especially in the length of the hairs, although the hairs did not differ in general form, or distribution on the leaf surface. Stomata differed little in length, width or frequency, but there were differences in the sort of stomata in relation to the surrounding epidermal cells. The hybrid material was intermediate between the parental species, and sometimes displayed aberrant trichomes that were contorted in contiguous pairs.
The results are discussed in relation to other work on the epidermal features of the Umbelliferae: as a means of discriminating the three taxa, and in relation to the ecological distributions of the species.  相似文献

A series of kaempferol derivatives have been identified in fronds of three parental species of the Appalachian Asplenium complex. Asplenium platyneuron is characterised by the presence of the 7-glucoside of kaempferol 3,4′-dimethyl ether and also contains kaempferol 3,7-diglucoside, free and with an aliphatic acyl attachment. By contrast, A. rhizophyllum contains a remarkable caffeoyl complex of kaempferol glycosides, which appears to be chromatographically homogenous. However, on deacylation, the complex yields caffeic acid and the 7-glucoside, 3,7-diglucoside, 3-sophoroside-7-glucoside and 7,4′-diglucoside of kaempferol. Asplenium montanum, in addition to having previously characterised glycosylxanthones, has two further kaempferol derivatives. It has been confirmed that these various species specific flavonoids are inherited in an additive fashion in three interspecific hybrids.  相似文献

Summary We tested the effect of a large number of diazotrophic strains from the genera Azotobacter, Klebsiella, Escherichia, Derxia, Beijerinckia, and Azospirillum, on nitrogen concentration in four corn hybrids. The range obtained, from highly significant positive to highly significant negative effects, indicates that the strains studied differed in their characters, including the capacity to form useful associations with certain hybrids. The forming of positive associations depended on the specificity of diazotrophic strains and corn hybrids.  相似文献

As whole-genome sequencing has become pervasive, some have suggested that reduced genomic representation approaches, for example, sequence capture, are becoming obsolete. In the present study, we argue that these techniques still provide excellent tools in terms of price and quality of data as well as in their ability to provide markers with specific features, as required, for example, in phylogenomics. A potential drawback of the wide-scale application of reduced representation approaches could be their drop in efficiency with increasing phylogenetic distance from the reference species. While some studies have focused on the degree and performance of reduced representation techniques in such situations, to our knowledge, none of them evaluated their applicability to inter-specific hybrids and polyploids. This highlights a significant gap in current knowledge since there is increasing evidence for the frequent occurrence of natural hybrids and polyploids, as well as for the major importance of both phenomena in evolution. The main aim of the present study was to carry out a thorough validation of SEQcap applicability to (1) a set of non-model taxa with a wide range of phylogenetic relatedness and (2) inter-specific hybrids of various ploidies and genomic compositions. Considering the latter point, we especially focused on mechanisms causing allelic bias and consequent allelic dropout, as these could have confounding effects with respect to the evolutionary genomic dynamics of hybrids, especially in asexuals, which virtually reproduce as a frozen F1 generation.  相似文献

Summary In poeciliid fish, melanoma of different degrees of malignancy can be produced by crossing specific genotypes. For a detailed investigation of the processes leading to proliferation or differentiation of the melanoma cells, it is necessary to establish cell cultures. The aim of the present study was to find out the optimal conditions for initiating and culturing poeciliid fish cells for the purpose of establishing cell cultures of melanoma. The optimal method was developed by using small pieces of late embryos as starting material and includes: (a) dispersion of tissue by mild stepwise treatment with a trypsin-EDTA mixture at low temperature; (b) culture of cells in the complex medium 199; (c) supplementation of medium with high percentage (20%) of fetal bovine serum; and (d) stabilization of pH by buffering the medium with HEPES. Under these conditions, primary and secondary cultures of embryonic cells have been initiated. An epithelial-like cell line has been subcultured for more than 80 passages. The method developed for embryonic tissues was used to start cell cultures from melanoma of platyfish-swordtail hybrids. Until now, only cells of rapidly growing malignant albino melanoma could be maintained in primary cultures. Secondary cultures could not be initiated since the melanoma cells tended to differentiate and stopped growing before a confluent monolayer was formed. Dedicated to Professor Dr. C.
