Gradual transfer to sea water of rainbow trout: effects on liver carbohydrate metabolism 总被引:2,自引:0,他引:2
The levels of glycogen and glucose, as well as of the activities of several key enzymes of glycogenolysis, glycolysis, gluconeogenesis and the pentose phosphate shunt were assessed in livers of small (97 g) and large (166 g) rainbow trout Oncorhynchus mykiss during gradual transfer to sea water (salinities of 0, 9, 18 and 28%). Plasma glucose and protein levels were also evaluated. Liver carbohydrate metabolism changed during gradual adaptation of rainbow trout to sea water. Glucose increased with salinity in livers of both sizes of animals, as did glycogenolysis and gluconeogenesis, but this latter only in large animals, and glycolytic rates in small animals. The different responses in metabolic parameters between sizes of animals may reflect a higher stress of the small animals and/or a better adaptation of large animals to increased salinities, in a way similar to that previously suggested by other authors after evaluation of non-metabolic parameters. Endosperms of quiescent barley grains contained, on average, 54.5 μg of neutral glyceride-glycerol, equivalent to ca 480 μg glyceride. Of this probably 90% was located in the aleurone layer. During germination the level of glyceride-glycerol declined. It also declined in degermed grains and aleurone layers incubated in vitro. The fall was accelerated by GA3, but indoleacetic acid, kinetin and glutamine were without effect. Increases in the levels of malate synthase and isocitrate lyase from very low initial values, and the results of incorporation studies with [14C]-labelled substrates, indicate that the glyoxylate cycle functions to convert glycerides to sucrose in germinating grains and degermed grains incubated with GA3, but not in degermed grains without the hormone. In the absence of GA3 the glyceride could be a respiratory substrate in degermed grains. The aleurone layers converted exogenous glucose to sucrose. Pi depletion of proximal tubule cells isolated from mouse kidney results in a decrease in the cell content of fructose-2,6-bisphosphate and an increase in the rate of gluconeogenesis from pyruvate, malate and succinate. Gluconeogenesis from glycerol is unaffected by Pi depletion. Introduction of fructose-2,6-bisphosphate into the cytosol of ATP-permeabilized cells is accompanied by a fall in gluconeogenesis. The presence of external Ca2+ stimulates gluconeogenesis. When cytosolic Ca2+ is raised to 1.8 microM by permeabilization, the resealed cells still require 2.5 mM Ca2+ in the bathing medium in order to perform gluconeogenesis at the maximum rate. Cells permeabilized in the presence of cAMP show a decreased rate of glucose production. Phorbol ester stimulates gluconeogenesis provided that the phorbol treatment is performed in the absence of Ca2+ ions. It is suggested that Pi depletion may stimulate pyruvate carboxylase activity and facilitate the entry of certain gluconeogenic substrates into mitochondria.
