The fate and availability of P derived from granular fertilisers in an alkaline Calcarosol soil were examined in a 65-year field trial in a semi-arid environment (annual rainfall 325 mm). Sequential P fractionation was conducted in the soils collected from the trial plots receiving 0–12 kg P ha−1crop−1, and the rhizosphere soil after growing wheat (Triticum aestivum L. cv. Yitpi) and chickpea (Cicer arietinum L. cv. Genesis 836) for one or two 60-day cycles in the glasshouse. Increasing long-term P application rate over 65 years significantly increased all inorganic P (Pi) fractions except HCl–Pi. By contrast, P application did not affect or tended to decrease organic P (Po) fractions. Increasing P application also increased Olsen-P and resin-P but decreased the P buffer capacity and sorption maxima. Residual P, Pi and Po fractions accounted for an average of 32, 16 and 52% of total P, respectively. All soil P fractions including residual P in the rhizosphere soil declined following 60-day growth of either wheat or chickpea. The decreases were greater in soils with a history of high P application than low P. An exception was water-extractable Po, which increased following plant growth. Changes in various P fractions in the rhizosphere followed the same pattern for both plant species. Biomass production and P uptake of the plants grown in the glasshouse correlated positively with the residual P and inorganic fractions (except HCl–Pi) but negatively with Po in the H2O-, NaOH- and H2SO4-fractions of the original soils. The results suggest that the long-term application of fertiliser P to the calcareous sandy soil built up residual P and non-labile Pi fractions, but these P fractions are potentially available to crops.  相似文献

基于我国南方双季稻区20年长期田间定位施肥试验,研究了不同氮磷钾施肥方式对水稻碳、氮积累与分配的影响.结果表明:偏施氮肥处理水稻籽实的碳、氮含量最高,分别达到433和18.9 g·kg-1.水稻植株的碳、氮储量以氮磷钾平衡施肥(NPK)及氮磷钾基础上有机物料循环施肥处理(NPKC)最高,其中NPKC和NPK处理籽实碳储量分别为2015和1960kg hm-2,茎叶碳储量分别为2048和2002 kg·hm-2;籽实氮储量分别为80.6和80.5kg·hm-2,茎叶氮储量则以NPK处理最高,为59.3 kg·hm-2.有机无机肥的配合施用显著增加了水稻植株体内碳和氮的累积;与偏施氮肥处理相比,氮磷钾的综合施用更利于水稻生长过程中碳、氮的累积与分配.  相似文献

Preliminary gut analysis of a recent Great Lakes invader, the round goby, Neogobius melanostomus (7.0–8.4 cm), collected from the Detroit River, showed that they ate zebra mussels (58%), snails (6%), and other invertebrates (36%), including aquatic insects (Hexagenia), softshelled crayfish, and zooplankton. Because zebra mussels, Dreissena polymorpha, predominated as prey, we investigated the ability of round gobies to consume different size classes of zebra mussels. In laboratory experiments, we examined feeding preferences of three size classes of round gobies (5.5–6.9 cm; 7.0–8.4 cm; 8.5–10.3 cm standard length) on four different size classes of zebra mussels (6.0–9.9 mm, 10.0–12.9 mm, 13.0–15.9 mm, 16.0–18.9 mm). All sizes of round gobies ate zebra mussels < 10.0 mm. Only the largest size class of round gobies ate larger zebra mussels (10.0–12.9 mm) when all prey sizes were presented. The association between the total mass of zebra mussels available and the amount consumed by round gobies increased positively up to about 6.5 g of available mussels and then levelled off. Round gobies consumed an average of 1.0 g of mussels in 24 h. There was a significant positive relationship between gape size and standard length of round gobies. Although larger round gobies (over the size range of fish in our study) are able to consume larger zebra mussels, small mussels were preferred. Our findings suggest that the preference of small zebra mussels by round gobies has the potential to alter the size structure of zebra mussel populations. 以陈年小麦离体胚为外植体,MS培养基为基本培养基,附加5种植物生长物质2,4-D、NAA、KT、6-BA、GA3进行单因子和正交试验,研究植物生长物质在陈年小麦离体胚萌发中的作用。单因子试验结果表明:单独使用6-BA 1.0mg/L、GA3 2.5mg/L均能在一定程度上促进陈年小麦离体胚出芽和生根,出芽率分别为70.0%和77.5%,比对照分别提高了21.7%和29.2%;生根率分别为63.3%和72.2%,比对照分别提高了21.3%和30.2%。正交试验结果显示GA3对陈年小麦离体胚的萌发影响显著,最佳植物生长物质组合为2,4-D2.0mg/L NAA 1.1mg/L 6-BA0.5mg/L GA3 4.0mg/L,出芽率为82.5%,生根率为60.0%,比对照分别提高了32.5%和17.5%。  相似文献

目的通过建立一个适应于尖吻蝮仔蛇生长发育的稳定的多功能生态系统的人工模拟生态养殖蛇场,以最大限度地降低仔蛇的饲料成本,从而达到降低人工养殖尖吻蝮蛇的成本。方法选取一个室外围墙式养蛇场作为试验基地。设计为适应于蛇、蛙、昆虫、蚯蚓、小杂鱼等动物生长繁衍的人工模拟生态环境,建立一个稳定的多功能蛇场生态系统,以尖吻蝮蛇100条,在试验以前随机抽取10条测量其体重和体长,并以活体动物饲料喂养法和科学饲养管理法进行试验养殖,并在进入第一、第二次冬眠前均随机抽取10条蛇仔测其体重和体长。结果本次试验与本蛇场室内人工饲养尖吻蝮仔蛇相比,不仅在体重、体长和肥满度方面均相差不大,而且仔蛇的存活率还有所提高。结论表明该人工模拟生态养殖蛇场能够满足尖吻蝮仔蛇生长发育的需要,达到了预期的目的。  相似文献

External phosphorus load to a wetland with two shallow lakes in the Botshol Nature Reserve, the Netherlands, was reduced since 1989, resulting in a rapid reduction of phosphorus levels, phytoplankton biomass and turbidity, and after 4 years, explosive growth of Characeae. The clear water state was unstable, however, and the ecosystem subsequently alternated between clear, high-vegetation and turbid, low-vegetation states. The switch from clear to turbid states occurred at a higher phosphorus level than the switch from turbid to clear states and area covered by macrophytes was significantly correlated to Secchi disc depth (r = 0.86, p < 0.001). Beginning in 1997, phosphorus input from droppings of greylag geese and black-headed gulls increased. The hypothesis that grazing prevented the return of Characeae populations was tested during 1996–1997 using large-scale exclosure experiments. The biomass and cover of Characeae were monitored in 17 exclosures for 2 years. The experiments showed no significant increase in Characeae in the absence of grazers compared with the controls. During the course of the experiments, the entire lake switched from turbid water to clear water and high Chara biomass. These ecosystem developments suggest that light limitation was the main factor controlling the collapse and return of Characeae in Botshol.  相似文献

Engineering recombinant viruses is a pre‐eminent tool for deciphering the biology of emerging viral pathogens such as the severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). However, the large size of coronavirus genomes renders the current reverse genetics methods challenging. Here, we describe a simple method based on “infectious subgenomic amplicons” (ISA) technology to generate recombinant infectious coronaviruses with no need for reconstruction of the complete genomic cDNA and apply this method to SARS‐CoV‐2 and also to the feline enteric coronavirus. In both cases we rescue wild‐type viruses with biological characteristics similar to original strains. Specific mutations and fluorescent red reporter genes can be readily incorporated into the SARS‐CoV‐2 genome enabling the generation of a genomic variants and fluorescent reporter strains for in vivo experiments, serological diagnosis, and antiviral assays. The swiftness and simplicity of the ISA method has the potential to facilitate the advance of coronavirus reverse genetics studies, to explore the molecular biological properties of the SARS‐CoV‐2 variants, and to accelerate the development of effective therapeutic reagents.  相似文献

应用单因素实验设计对干式酶法松籽油提取工艺进行优化。结果表明,干式酶法提取松籽油最佳制备工艺为:0.2%淀粉酶、料液比为6:1、酶解温度为55℃、酶解时间为8 h,所得最大出油率为90.2%,残油率为6.2%。经检测最佳制备条件提取的松籽油不饱和脂肪酸的含量为90.09%,其中油酸含量为26.84%,介酸含量为2.41%、亚油酸含量为46.25%、皮诺敛酸为14.59%;饱和脂肪酸中棕榈酸含量为6.5%、硬脂酸为3.41%。松籽油酸价为2.61 mg·g-1、过氧化值为1.56 mmol·kg-1、丙二醛含量为0.41 mg·kg-1。所有检测结果均高于《中华人民共和国粮食行业标准LS/T 3242-2014》松籽油中质量标准的要求。  相似文献

为了提高实验室人类免疫缺陷病毒(human immunodeficiency virus,HIV)抗体检测能力及对免疫蛋白印迹(Western blot,WB)实验结果的判断能力,对送检至北京市东城区艾滋病确证实验室的268份HIV抗体待确定样本进行确证实验及结果分析。按照试剂说明书和实验室标准作业程序(Standard Operation Procedure,SOP)操作对送检的全部样本进行WB确证实验;了解HIV筛查实验结果与确证实验结果的相关性,并分析不同送检机构、送检人群样本的检测结果差异以及不同试剂、不同检测方法的结果差异。结果显示在筛查出抗体待确定的268份样本中,确证阳性170份,阳性率63.43%;确证阴性51份,阴性率19.03%;不确定结果47份,占筛查有反应的17.54%。确证阳性病例来自监管场所、自愿咨询检测门诊(Voluntary Counseling and Test,VCT)和医疗机构,不同送检单位及不同人群的阳性样本率有显著统计学意义(P<0.01)。WB确证阳性样本带型以全条带和次全条带为主,且所有确证阳性标本均来自双试剂阳性样本。不同检测方法阳性样本率的差异有显著统计学意义(P<0.01),其中化学发光法的样本阳性率占46.27%,酶联免疫吸附实验(ELISA)占88.29%,胶体硒法占43.48%。本研究结果提示,对潜在HIV感染者,应扩大检测面,加强医疗机构检测,并提供一种以上方法的多次检测,以减少漏检的风险。  相似文献

Soil legacy effects are commonly highlighted as drivers of plant community dynamics and species co‐existence. However, experimental evidence for soil legacy effects of conditioning plant communities on responding plant communities under natural conditions is lacking. We conditioned 192 grassland plots using six different plant communities with different ratios of grasses and forbs and for different durations. Soil microbial legacies were evident for soil fungi, but not for soil bacteria, while soil abiotic parameters did not significantly change in response to conditioning. The soil legacies affected the composition of the succeeding vegetation. Plant communities with different ratios of grasses and forbs left soil legacies that negatively affected succeeding plants of the same functional type. We conclude that fungal‐mediated soil legacy effects play a significant role in vegetation assembly of natural plant communities.  相似文献
