The defensive mechanisms of the wild potato, solanum berthaultii Hawkes, to larvae of the Colorado potato beetle, Leptinotarsa decemlineata (Say), were studied by selective removal of glandular trichomes and trichome exudates from leaflets, and by comparing performance on S. berthaultii and on the cultivated potato, S. tuberosum L., which lacks defensively active type A and B glandular trichomes. Removal of type A trichomes increased the proportion of larvae that fed on S. berthaultii. Removal of the exudate from type B trichomes increased the proportion of larvae that fed and led to a decrease in mortality. The predominant active compounds in type B exudate, i.e. fatty acid esters of sucrose, were only effective in the presence of type A trichomes. Sucrose esters did not affect larval feeding on S. tuberosum leaflets or on S. berthaultii leaf discs from which the type A trichomes had been removed. Growth of surviving larvae was not significantly affected by removing type A trichomes or type B exudate. Growth of larvae was significantly increased when S. berthaultii leaflets were presented in artificial diet which eliminated the physical barrier of the type B stalks. Growth was no different on artificial diet containing either S. berthaultii or S. tuberosum leaf material (fresh or lyophilized powder) but was poorer on these diets than on S. tuberosum leaflets. The presence of type A trichomes is a fundamental requirement for expression of S. berthaultii resistance to L1 L. decemlineata. Type B droplets containing sucrose esters increase the expression of resistance in the presence of defensively-active type A trichomes.
Résumé Les mécanismes de défense de la pomme de terre sauvage, S. berthaultii Hawkes, aux larves de Leptinotarsa decemlineata Say, ont été étudiés par ablation sélective des trichomes glandulaires et par l'élimination de leur exsudat des folioles, et par comparaison avec S. tuberosum L. qui a perdu les trichomes glandulaires défensifs A et B. L'ablation des trichomes A a augmenté la proportion de larves ayant consommé S. berthaultii. L'élimination de l'exsudat des trichomes B a augmenté la proportion de consommatrices et réduit la mortalité. Les principaux composés actifs de l'exsudat B, c'est-à-dire des esters d'acides gras de sucrose, n'étaient actifs qu'en présence de trichomes A. Les esters de sucrose n'ont pas modifié la consommation larvaire sur folioles de S. tuberosum, ou sur disques de feuilles de S. berthaultii dont les trichomes A avaient été enlevés. La croissance des larves survivantes n'a pas été modifiée significativement par l'ablation des trichomes A ou l'élimination de l'exsudat de B. La croissance des larves a été significativement augmentée quand les folioles de S. berthaultii ont été incorporés dans l'aliment artificiel après élimination de la barrière physique due aux pédoncules B. La croissance a été de même importance sur aliments artificiels contenant des feuilles (fraiches ou en poudre lyophylisée) de S. berthaultii ou de S. tuberosum, mais plus faible que sur folioles de S. tuberosum. La présence de trichomes A est indispensable à la résistance de S. berthaultii aux L, de L. decemlineata. Background and Aims
Oil-producing flowers related to oil-bee pollination are a major innovation in Neotropical and Mexican Iridaceae. In this study, phylogenetic relationships were investigated among a wide array of New World genera of the tribes Sisyrinchieae, Trimezieae and Tigridieae (Iridaceae: Iridoideae) and the evolution of floral glandular structures, which are predominantly trichomal elaiophores, was examined in relation to the diversification of New World Iridaceae.Methods
Phylogenetic analyses based on seven molecular markers obtained from 97 species were conducted to produce the first extensive phylogeny of the New World tribes of subfamily Iridoideae. The resulting phylogenetic hypothesis was used to trace the evolutionary history of glandular structures present in the flowers of numerous species in each tribe. Hypotheses of differential diversification rates among lineages were also investigated using both topological and Binary-State Speciation and Extinction methods.Key Results and Conclusions
Floral glandular structures and especially trichomal elaiophores evolved multiple times independently in the American tribes of Iridoideae. The distribution pattern of species displaying glandular trichomes across the phylogeny reveals lability in the pollination system and suggests that these structures may have played a significant role in the diversification of the Iridoideae on the American continent. 相似文献175.

We describe a method for establishing the culture of bovine tracheal submucosal gland (BTG) cells, in which we have also examined the influence of a reconstituted basement membrane matrix derived from the Engelbreth-Holm-Swarm tumor (EHS) on the growth and morphological differentiation of these cells. BTG cells have been isolated by tissue enzymatic digestion using trypsin, deoxyribonuclease I, elastase, hyaluronidase and EGTA for 1 hr at 37 degrees C. Afterwards, cells and tissue were collected by centrifugation and were incubated for 15 min with 15% newborn calf serum to inactivate the proteolytic enzymes. Enzymatic digestion using only trypsin, centrifugation and inactivation steps were repeated three times. Using this protocol, we obtained 15 +/- 4 (X 10(6] cells per g of tracheal submucosa with 72 +/- 2% (n = 5) cell viability. On microscopic observation, isolated cells were mainly composed of serous type glandular cells. Cells were cultured in a 1:1 medium of Dulbecco's Modified Eagle's/Ham's F12 supplemented with 10% fetal calf serum and subcultured in either plastic flasks or flasks coated with EHS matrix. On the plastic, the BTG cells exhibited at confluency an epithelioid appearance. They stained positively with the immunofluorescent anticytokeratin antibody and contained PAS-staining granules. By electron microscopy, lactoferrin, a protein marker specific to the serous cells, was demonstrated immunocytochemically in small secretory vesicles. BTG cells cultured on EHS matrix revealed a significantly increased growth in comparison to those cultured on plastic. In post-confluent culture of BTG cells on EHS matrix, we observed numerous dome-like structures formed by differentiated cells which were joined together around luminal spaces. 相似文献

In order to clarify whether the structures observed at the base of the petiole of the genus Ilex are colleters resulting from stipules, the anatomy, vascularization and secretions of these supposed glandular structures were analysed in nine species. This is the first report of colleters in Ilex. Stipular colleters replace the stipules in all species studied and are characterized by the presence of vascular traces. In addition to the stipular colleters, three other types of colleter were distinguished: standard and lachrymiform colleters found on the leaf teeth or crenations, and sessile colleters found on the margins of the floral bracts. Their basic structure consists of a central core of parenchymatous cells surrounded by one layer of palisade secretory epidermal cells. Histochemical tests were also performed on secretions; proteins were found in the secretions studied, but glucose was not. The glandular origin of the stipular colleters is confirmed on the basis of their position, secretions and anatomy. The histological components of the leaf were studied in dried herbarium material of the three Origanum vulgare subspecies (subsp. hirtum , subsp. viridulum and subsp. vulgare ) grown wild in Greece. These three, geographically distinct, taxa showed remarkable differences in their leaves. The leaves of subsp. hirtum grown in the Mediterranean climatic zone of Greece are characterized by thick cutinized outer walls of the epidermal cells and a thick mesophyll with highly developed chlorophyllous tissues. Peltate glandular trichomes and stomata are numerous on bom leaf surfaces. The thickness of the mesophyll decreases in the other two subspecies grown in the northern part of the country, where a Continental type of climate occurs. The number of glandular trichomes and stomata also decreases. Besides these differences, a noticeable reduction in the size of the essential oil-accumulating subcuticular chamber of the glandular trichomes and in the number of the peribasal cells, has also been recorded in die plants of subsp. vulgare and viridulum. 相似文献

Abstract Two types of glandular hairs have been found in Cistus laurifolius L. (Cistaceae). Both types present a great degree of morphological and histochemical variability, not only at population level but even within a single plant. This fact excludes that the hairs of C. laurifolius could be given a diagnostic value. The two hair types have a mixed hydrophilous and lipophilous secretion in which we have identified, especially in one type of them, cathecolic tannins. The powerful antimycotic activity of such tannins, makes the hairs particularly interesting for the possible ecological implications. 相似文献

Alterations in essential oil gland number, distribution and fine structure, and the oil content in the leaf of Mentha arvensis
L. were examined during its growth and senescence. Accumulation of essential oil occurred predominantly during the rapid leaf
expansion phase followed by a similar decline. The oil gland (trichome) number increased upto leaf maturation and declined
thereafter. Initially, cuticle remains tightly apposed to the secretory head of oil glands but progressively a sub-cuticular
space appears to be created for the oil. Considerable enlargement of vacuole with ageing is witnessed, whereas cytoplasm gradually
decreases to a thin peripheral layer. Some secretory cells from senescing leaf were found almost empty, having only a few
remnant oil droplets.
