Two amphipathic protein fractions soluble in organic solvents as well as in water have been isolated from the ganglioside fraction of bovine erythrocyte membranes by successive chromatography in chloroform-methanol mixture on DEAE-Sephadex, silicic acid, and α-hydroxypropylated Sephadex G50 (LH60) columns. These two fractions contained a similar low molecular weight protein but with distinctively different amino acid composition. One of these proteins has been characterized by having a strong Paul-Bunnell antigen activity and had a binding affinity to ganglioside. A similar protein without Paul-Bunnell antigen activity was isolated as the major ganglioside-associated protein. 相似文献

Summary Cornish-Bowden and Marson have recently suggested that the finite sampling component of Q, a measure of nonrandomness in the amino acid composition of proteins, may have been underestimated because it was calculated on the basis of the genetic code table frequencies rather than on the basis of the average natural abundance with which the twenty amino acids actually occur in proteins. This underestimate would lead to an overestimate of Qc a measure of selective effects above and beyond those imposed by the average natural abundance of the amino acids. In this paper the finite sampling component of Q is quantitatively estimated on the basis of these natural abundances and found to reduce Qc from its previous average value of 24.3 to the lower value of 9.7, with the standard deviation of the population of Qc values being 12.5. Individual Qc values are given for 81 protein families of mean composition per 61 codons of Ala5.3 Arg2.4 Asn3.0 Asp3.6 Cys1.5 Gln2.6 Glu3.5 Gly4.7 His1.3 Ile3.4 Leu4.5 Lys4.2 Met1.0 Phe2.3 Pro2.3 Ser4.2 Thr3.6 Trp0.8 Tyr2.6 Val4.2. The mean Qc value of 9.7 is notably small, and indicates that quantitatively minimal adjustments away from the average protein composition are necessary to maintain many different biological functions. This small value, however, is shown to differ significantly from the value of zero expected were the natural abundances of the amino acids the only selective constraint. Leaf wax of a glaucous variety of Poa ampla contains hydrocarbons (5%, C23–C35), esters (9%, C36–C56), free acids (3%, C16–C34), free alcohols (6%, mainly C26); hentriacontane-14,16-dione (14%), 5-oxohentriacontane-14,16-dione (1%); hydroxy β-diketones (56%) and unidentified material (6%). The hydroxy β-diketones, which are more abundant in this wax than in others, were shown by 13C NMR to consist of 4-hydroxy (15%), 5-hydroxy (70%) and 6-hydroxy (15%) hentriacontane-14,16-diones. 相似文献

The thermal stability of excitation transfer from pigment proteins to the Photosystem II reaction center of Nerium oleander adjusts by 10 Celsius degrees when cloned plants grown at 20°C/15°C, day/night growth temperatures are shifted to 45°C/32°C growth temperature or vice versa. Concomitant with this adjustment is a decrease in the fluidity of thylakoid membrane polar lipids as determined by spin labeling. The results are consistent with the hypothesis that there is a limiting maximum fluidity compatible with maintenance of native membrane structure and function. This limiting fluidity was about the same as for a number of other species which exhibit a range of thermal stabilities. Inversely correlated shifts in lipid fluidity and thermal stability occurred during the time course of acclimation of N. oleander to new growth temperatures. Thus, the temperature at which the limiting fluidity was reached changed during acclimation while the limiting fluidity remained constant. Although the relative proportion of the major classes of membrane polar lipids remained constant during adjustments in fluidity, large changes occured in the abundance of specific fatty acids. These changes were different for the phospho- and galacto-lipids suggesting that the fatty acid composition of these two lipid classes is regulated by different mechanisms. Comparisons between membrane lipid fluidity and fatty acid composition indicate that fluidity is not a simple linear function of fatty acid composition. 相似文献

Ultrasound (A-scan mode) and skinfold methods were evaluated in the measurement of subcutaneous fat thickness and prediction of total fat weight (by whole body potassium counting). Based on intraobserver correlations on 39 men at 15 body sites, skinfold caliper measurements were more reproducible than ones obtained by ultrasound. Measurements made with the two techniques at the same site typically produced different mean estimates of fat thickness. However, scores were often highly correlated with each other, indicating similar relative rankings of subjects by each technique. Skinfolds were more highly correlated with total fat weight than were ultrasound measurements, but body weight and anthropometric measures had even higher correlations with total fat weight. Anthropometric measurements were highly correlated with fatness because of their association with body weight, and when this relationship was statistically controlled for, they typically lost their predictive effectiveness. Multiple regression analyses revealed that the best predictors of fat weight were body weight along with skinfold and ultrasound measurements. These results suggest that skinfolds are a more effective means of assessing subcutaneous fat than ultrasound, especially when the large difference in cost of equipment is considered. 相似文献

The gamma-aminobutyric acidA/benzodiazepine receptor complexes from bovine cerebral cortex were purified by immunoaffinity chromatography, and the main component peptide subunits were characterized. The peptide band originally thought to be a single beta subunit [57,000 Mr band in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE)] is composed of at least four different peptides of 54,000-57,000 Mr. Two peptides of 55,000 and 57,000 Mr were recognized by the beta subunit-specific monoclonal antibody 62-3G1. Peptides in the range of 54,000-57,000 Mr were photoaffinity-labeled with [3H]muscimol. A different 57,000 Mr peptide was photoaffinity-labeled by [3H]flunitrazepam, but neither was recognized by the monoclonal antibody 62-3G1 nor photoaffinity-labeled with [3H]muscimol. Some peptides could be identified by their differential mobility shift in SDS-PAGE after treatment with endoglycosidase H. Two additional subunit peptides of 51,000 and 53,000 Mr were also photoaffinity-labeled by [3H]flunitrazepam and reacted with antiserum A. Mangrove stands in Puttalam lagoon and Dutch bay, two interconnected lagoons situated on the northwestern coast of Sri Lanka have been classified broadly into two groups, i.e., estuarine and island/mainland-fringing stands. Structural diversity of six mangrove stands, representing the two types was studied in terms of floristic composition, density, basal area, mean stand diameter, tree height, standing above-ground biomass and leaf-area index.Rhizophora mucronata andAvicennia marina were the dominant species. Higher mean stand diameters for the mangrove stands in Puttalam lagoon indicated greater maturity than the estuarine mangrove stands in Dutch bay. Nevertheless, estuarine stands in Dutch bay were structurally more complex (complexity indices 8.11–22.7) than the island/mainland-fringing mangrove stands (complexity indices 1.38–6.78). Higher number of species present in the estuarine mangrove stands is the major element that contributes to the higher values for the complexity indices for those stands. This appears to mask the contribution of stand-age to the complexity of a mangrove stand. Therefore complexity indices alone may not be used to explain adequately the structural diversity among mangrove stands. 相似文献

The harpacticoid copepod Tisbe holothuriae Humes was reared for several generations under different photoperiods (white light) and spectral quality (continuous illumination).
The main life cycle parameters were measured and demographic variables were determined. Development time was retarded under
green wavelengths and at photoperiods different from LD 12:12, especially under continuous white light. Total body length
of females and males was shortest under LD 12:12 and longest under constant dark. Photoperiods different from LD 12:12 prolonged
the interval between broods of egg sacs. Short photoperiods caused higher abortion rate, while long photoperiods retarded
the maturation time of the egg sacs. Red wavelengths stimulated the abortion rate. Photoperiod LD 12:12 and blue wavelengths
increased the production of offspring and the life span of females. Highest intrinsic rates of natural increase were estimated
under LD 12:12 (rm =0.304) and blue wavelengths (rm = 0.254). These light conditions proved the most favourable for rearing Tisbe holothuriae. 相似文献

Summary Chaos game representation (CGR) is a novel holistic approach that provides a visual image of a DNA sequence quite different from the traditional linear arrangement of nucleotides. Although it is known that CGR patterns depict base composition and sequentiality, the biological significance of the specific features of each pattern is not understood. To systematically examine these features, we have examined the coding sequences of 7 human globin genes and 29 relatively conserved alcohol dehydrogenase (Adh) genes from phylogenetically divergent species. The CGRs of human globin cDNAs were similar to one another and to the entire human globin gene complex. Interestingly, human globin CGRs were also strikingly similar to human Adh CGRs. Adh CGRs were similar for genes of the same or closely related species but were different for relatively conserved Adh genes from distantly related species. Dinucleotide frequencies may account for the self-similar pattern that is characteristic of vertebrate CGRs and the genome-specific features of CGR patterns. Mutational frequencies of dinucleotides may vary among genome types. The special features of CG dinucleotides of vertebrates represent such an example. The CGR patterns examined thus far suggest that the evolution of a gene and its coding sequence should not be examined in isolation. Consideration should be given to genome-specific differential mutation rates for different dinucleotides or specific oligonucleotides.
