The mouse insulin-like growth factor II (Igf2) and H19 genes are located adjacent to each other on chromosome 7q11-13 and are reciprocally imprinted. It is believed that the allelic expression of these two genes is regulated by the binding of CTCF insulators to four parent-specific DNA methylation sites in an imprinting control center (ICR) located between these two genes. Although monoallelically expressed in peripheral tissues, Igf2 is biallelically transcribed in the CNS. In this study, we examined the allelic DNA methylation and CTCF binding in the Igf2/H19 imprinting center in CNS, hypothesizing that the aberrant CTCF binding as one of the mechanisms leads to biallelic expression of Igf2 in CNS. Using hybrid F1 mice (M. spretus males x C57BL/6 females), we showed that in CNS, CTCF binding sites in the ICR were methylated exclusively on the paternal allele, and CTCF bound only to the unmethylated maternal allele, showing no differences from the imprinted peripheral tissues. Among three other epigenetic modifications examined, histone H3 lysine 9 methylation correlated well with Igf2 allelic expression in CNS. These results suggest that CTCF binding to the ICR alone is not sufficient to insulate the Igf2 maternal promoter and to regulate the allelic expression of the gene in the CNS, thus challenging the aberrant CTCF binding as a common mechanism for lack of Igf2 imprinting in CNS. Further studies should be focused on the identification of factors that are involved in histone methylation and CTCF-associated factors that may be needed to coordinate Igf2 imprinting.  相似文献

利用C分带、基因组原位杂交并结合分子生物学手段,对12份巨穗小麦种质材料中的外源遗传物质进行了检测.结果表明,12份材料染色体数均为42,其中5份材料均具有一对小麦-黑麦(Triticum aestivum-Secalecereal)1BL/1RS易位染色体和一对中间偃麦草(Agropyron intermedium Garten)染色体、3份材料只具有一对中间偃麦草染色体、3份材料只具一对1BL/1RS染色体、1份材料无1BL/1RS和中间偃麦草染色体.进一步细胞学分析表明,此中间偃麦草染色体代换了普通小麦(Triticum aestivum L.)中的2D染色体,因其良好的同源补偿性,表示为2Ai.同时对2Ai在巨穗小麦种质中存在的遗传学意义及小麦遗传改良中的应用进行了讨论.  相似文献

Epigenetic modifications may be one mechanism linking early life factors, including parental socioeconomic status (SES), to adult onset disease risk. However, SES influences on DNA methylation patterns remain largely unknown. In a US birth cohort of women, we examined whether indicators of early life and adult SES were associated with white blood cell methylation of repetitive elements (Sat2, Alu and LINE-1) in adulthood. Low family income at birth was associated with higher Sat2 methylation (β = 19.7, 95% CI: 0.4, 39.0 for lowest vs. highest income quartile) and single parent family was associated with higher Alu methylation (β = 23.5, 95% CI: 2.6, 44.4), after adjusting for other early life factors. Lower adult education was associated with lower Sat2 methylation (β = -16.7, 95% CI: -29.0, -4.5). There were no associations between early life SES and LINE-1 methylation. Overall, our preliminary results suggest possible influences of SES across the life-course on genomic DNA methylation in adult women. However, these preliminary associations need to be replicated in larger prospective studies.  相似文献

We report an analysis of allele-specific expression (ASE) and parent-of-origin expression in adult mouse liver using next generation sequencing (RNA-Seq) of reciprocal crosses of heterozygous F1 mice from the parental strains C57BL/6J and DBA/2J. We found a 60% overlap between genes exhibiting ASE and putative cis-acting expression quantitative trait loci (cis-eQTL) identified in an intercross between the same strains. We discuss the various biological and technical factors that contribute to the differences. We also identify genes exhibiting parental imprinting and complex expression patterns. Our study demonstrates the importance of biological replicates to limit the number of false positives with RNA-Seq data.  相似文献

为了进一步研究Elymus sibiricus L.、E.nutans Griseb.和E.burchan-buddae(Neuski)Tzelev [=Roegneria nutans(Keng)Keng]的外部形态差异及其系统学关系,本文对这三种植物的6个穗部形 态性状进行了观测和比较,并对这三个Elymus种进行了种间杂交及杂种F1的减数分裂染色体配对行 为的分析研究。结果表明:这三个Elymus种的穗长及颖长等性状均变异很大,而内稃的长、宽则变异 不大并具有明显的种间差异。E. nutans×E.barchan-buddae及E.nutans×E.sibiricus的杂种F1均完 全不育,减数分裂不规则。E.nutans×E.burchan-buddae杂种F1的减数分裂构型为:7.70I+13.40 Ⅱ+0.06Ⅲ+O.08 Ⅳ,而E.sibiricus×E.nutans杂种F1的构型为11.98 Ⅰ+9.61Ⅱ+O.64Ⅲ+0.39 Ⅳ+0.01V。由本实验的形态学和细胞学的研究结果得出以下结论:1.利用内稃形态性状结合穗部 其它性状的差异能对这三个物种进行较准确的鉴定;2.E.nutans与E.burchan-buddae的亲缘关系较 近,而E.nutans与E.sibiricus的亲缘关系则较远;3. Restriction fragment length polymorphism (RFLP) of the total DNA ofBipolaris andCurvularia species was analysed using arbitrarily chosen genomic clones of DNA fromCurvularia lunata andBipolaris maydis as probes. Clear differences among species in both genera, resulting in different banding positions, were obtained with some probe-enzyme combinations. Intraspecific polymorphism in banding positions with these probe-enzyme combinations was slight. These analyses allow discrimination between the species. DNA fingerprinting with intrageneric probes is a potentially useful tool for species separation and identification inBipolaris andCurvularia when coupled with another characteristic such as conidial morphology.Curvularia aeria comb. nov. was proposed forCurvularia lunata var.aeria on the basis of differences in RFLP banding patterns and differences in conidial morphology.  相似文献

Gastric cancer (GC) was responsible for over 1 000 000 new cases in 2018 and an estimated 783 000 deaths, making it still the fifth most frequently diagnosed cancer and the third leading cause of cancer deaths in both sexes worldwide. Divergent trends for GC incidence were observed in the USA. Incidence rates, particularly for non‐cardia GC, were stable or increasing among persons aged <50 years. In an analysis of data from a public hospital database in Hong Kong, treatment of Helicobacter pylori infection was associated with a lower risk of GC, particularly in older subjects who received treatment ≥10 years before. Based on the results of a 16‐year endoscopy‐based follow‐up eradication trial, patients with incomplete‐type intestinal metaplasia (IM) should receive endoscopic surveillance upon H. pylori eradication therapy. Updated guidelines on the endoscopic surveillance of preneoplastic conditions of the stomach (MAPS II) have been published. In the RAINFALL trial, the addition of ramucirumab to a backbone chemotherapy as a first‐line regimen failed to improve overall survival (OS) of patients with metastatic disease. Also, pembrolizumab did not prolong OS when compared to paclitaxel in the second‐line treatment of patients with advanced GC or esophagogastric junction (EGJ) cancer. Trifluridine/tipiracil improved OS by 2.1 months in the third or further treatment line of patients with advanced GC. In a systematic investigation conducted on Chinese patients with GC, CLDN18‐ARHGAP26/6 fusion was associated with signet‐ring cell content and was prognostic for a worse outcome and predictive for no benefit from oxaliplatin/fluoropyrimidine‐based chemotherapy. Organoid cultures represent an appealing model that may be applied for therapy response testing in the near future.  相似文献

Candidatus Liberibacter asiaticus (Ca. L. asiaticus) is a Gram‐negative bacterium and the pathogen of Citrus Greening disease (Huanglongbing, HLB). As a parasitic bacterium, Ca. L. asiaticus harbors ABC transporters that play important roles in exchanging chemical compounds between Ca. L. asiaticus and its host. Here, we analyzed all the ABC transporter‐related proteins in Ca. L. asiaticus. We identified 14 ABC transporter systems and predicted their structures and substrate specificities. In‐depth sequence and structure analysis including multiple sequence alignment, phylogenetic tree reconstruction, and structure comparison further support their function predictions. Our study shows that this bacterium could use these ABC transporters to import metabolites (amino acids and phosphates) and enzyme cofactors (choline, thiamine, iron, manganese, and zinc), resist to organic solvent, heavy metal, and lipid‐like drugs, maintain the composition of the outer membrane (OM), and secrete virulence factors. Although the features of most ABC systems could be deduced from the abundant experimental data on their orthologs, we reported several novel observations within ABC system proteins. Moreover, we identified seven nontransport ABC systems that are likely involved in virulence gene expression regulation, transposon excision regulation, and DNA repair. Our analysis reveals several candidates for further studies to understand and control the disease, including the type I virulence factor secretion system and its substrate that are likely related to Ca. L. asiaticus pathogenicity and the ABC transporter systems responsible for bacterial OM biosynthesis that are good drug targets. Proteins 2012. © 2012 Wiley Periodicals, Inc.  相似文献

Random amplified polymorphic DNA(RAPD) markers were investigated to clarify the taxonomic positions ofVicia linearifolia andV. bifolia, and to assess the genomic diversity among the 9 populations ofV. unijuga, each of which represents a geographical variation or infraspecific taxa in southern Korea. These species are characterized by unijugate leaves in East Asia and have been controversial as to infra-or interspecific classification. The polymorphic markers among the populations examined were observed for fifteen decamer primers. The degree of band sharing was used to calculate genetic similarity between populations, and a phenogram using UPGMA cluster analysis was generated based on the Dice similarity coefficient. The taxa studied were divided into two main groups and the populations ofV. unijuga were all grouped together in the phenogram. The genetic similarities ofV. unijuga were very high among the populations and did not show distinctions between the infraspecific taxa, although the populations of Mt. Odae and adjacent areas in eastern Korea were different from others of the species.V. linearifolia fell within the range of the genomic variation among the populations ofV. unijuga, whileV. bifolia was grouped withV. venosa var.cuspidata having multijugate leaves rather thanV. unijuga. The result from studying RAPD markers suggested thatV. linearifolia should be integrated intoV. unijuga and that species with unijugate leaves ofV. bifolia andV. unijuga are polyphyletic.  相似文献

An overview is presented on the status of studies on multiple codes in genetic sequences. Indirectly, the existence of multiple codes is recognized in the form of several rediscoveries of Second Genetic Code that is different each time. A due credit is given to earlier seminal work related to the codes often neglected in literature. The latest developments in the field of chromatin code are discussed, as well as perspectives of single-base resolution studies of nucleosome positioning, including rotational setting of DNA on the surface of the histone octamers.  相似文献
