用幼胚培养方法重新获得的埃塞俄比亚芥(Brassica carinata A.Braun,2n=34)与诸葛菜(Orychophragmus violaceus (L.)O.E.Schulz,2n=24)的属间杂种仍为混倍体(2n=14~34),2n=34细胞的频率最高;绝大多数花粉母细胞(PMCs)表现正常的17个二价体配对和17:17的分离。基因组原位杂交分析结果表明,在所有体细胞和PMCs中不含有整条的诸葛菜染色体,2n=34的体细胞和PMCs中包含了来自黑芥(B.nigra (L.)Koch,2n=16)的16条染色体。这些具有完全或部分埃塞俄比亚芥染色体组成的细胞,可能来源于以前提出的杂种细胞在有丝分裂中完全或部分亲本染色体组分开和染色体复制,并伴随诸葛菜染色体的消除。 相似文献

Organic bulk heterojunction (BHJ) solar cells require energetic offsets between the donor and acceptor to obtain high short‐circuit currents (JSC) and fill factors (FF). However, it is necessary to reduce the energetic offsets to achieve high open‐circuit voltages (VOC). Recently, reports have highlighted BHJ blends that are pushing at the accepted limits of energetic offsets necessary for high efficiency. Unfortunately, most of these BHJs have modest FF values. How the energetic offset impacts the solar cell characteristics thus remains poorly understood. Here, a comprehensive characterization of the losses in a polymer:fullerene BHJ blend, PIPCP:phenyl‐C61‐butyric acid methyl ester (PC61BM), that achieves a high VOC (0.9 V) with very low energy losses (Eloss = 0.52 eV) from the energy of absorbed photons, a respectable JSC (13 mA cm?2), but a limited FF (54%) is reported. Despite the low energetic offset, the system does not suffer from field‐dependent generation and instead it is characterized by very fast nongeminate recombination and the presence of shallow traps. The charge‐carrier losses are attributed to suboptimal morphology due to high miscibility between PIPCP and PC61BM. These results hold promise that given the appropriate morphology, the JSC, VOC, and FF can all be improved, even with very low energetic offsets. 相似文献

Understanding the origin of new species is a central goal in evolutionary biology. Diverging lineages often evolve highly heterogeneous patterns of genetic differentiation; however, the underlying mechanisms are not well understood. We investigated evolutionary processes governing genetic differentiation between the hybridizing campions Silene dioica (L.) Clairv. and S. latifolia Poiret. Demographic modelling indicated that the two species diverged with gene flow. The best‐supported scenario with heterogeneity in both migration rate and effective population size suggested that a small proportion of the loci evolved without gene flow. Differentiation (F ST) and sequence divergence (d XY) were correlated and both tended to peak in the middle of most linkage groups, consistent with reduced gene flow at highly differentiated loci. Highly differentiated loci further exhibited signatures of selection. In between‐species population pairs, isolation by distance was stronger for genomic regions with low between‐species differentiation than for highly differentiated regions that may contain barrier loci. Moreover, differentiation landscapes within and between species were only weakly correlated, suggesting that linked selection due to shared recombination and gene density landscapes is not the dominant determinant of genetic differentiation in these lineages. Instead, our results suggest that divergent selection shaped the genomic landscape of differentiation between the two Silene species, consistent with predictions for speciation in the face of gene flow. 相似文献

Barley (Hordeum vulgare L.) is one of the most Aluminum (Al) sensitive cereal species. In this study, the physiological, biochemical, and molecular response of barley seedlings to Al treatment was examined to gain insight into Al response and tolerance mechanisms. The results showed that superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activity were inhibited to different degrees following Al exposure. The MDA content also significantly increased with increasing Al concentrations. SRAP results indicated significant differences between Al treatments and controls in terms of SRAP profile, and the genomic template stability (GTS) decreased with increasing Al concentration and duration. These integrative results help to elucidate the underlying mechanisms that the barley response to Al toxicity. 相似文献

燕麦是一种主要生长在温带冷凉地区的粮饲兼用作物,近年来燕麦的营养价值和降低胆固醇特性的发现使人们认识到燕麦及其制品是一种健康食品,促进了燕麦产业发展,对燕麦品种培育提出了更高要求。在此背景下,现代生物技术与常规育种技术结合是满足这一需求的重要途经。本文综述了国内外燕麦分子育种方面的研究进展:(1)我国燕麦种质资源的收集与遗传多样性研究。我国的燕麦种质资源收集工作起始于20世纪50年代,迄今收集并保存了5282份燕麦种质资源。对这些资源的遗传多样性分析研究表明,国内的燕麦种质资源中内蒙古和山西的资源多样性最高;(2)利用各种分子标记构建燕麦遗传连锁图谱研究;(3)一些重要农艺性状的QTL定位研究以及全基因组关联分析研究。包括产量、含油量、β-葡聚糖含量、蛋白质含量、抽穗期、抗病基因、抗冻性等重要农艺性状;(4)标记辅助基因组选择技术在燕麦中的应用;(5)燕麦遗传工程育种研究进展。同时,本文将国内的主要研究进展与国外相关的最新研究成果进行了比较,并对当前分子育种中存在的问题及今后的研究方向进行了探讨,希望能为今后通过生物技术手段培育燕麦新品种提供一定的参考。 相似文献

Biomolecular condensates (BMCs) play an important role in the replication of a growing number of viruses, but many important mechanistic details remain to be elucidated. Previously, we demonstrated that the pan-retroviral nucleocapsid (NC) and HIV-1 pr55Gag (Gag) proteins phase separate into condensates, and that HIV-1 protease (PR)-mediated maturation of Gag and Gag-Pol precursor proteins yields self-assembling BMCs that have HIV-1 core architecture. Using biochemical and imaging techniques, we aimed to further characterize the phase separation of HIV-1 Gag by determining which of its intrinsically disordered regions (IDRs) influence the formation of BMCs, and how the HIV-1 viral genomic RNA (gRNA) could influence BMC abundance and size. We found that mutations in the Gag matrix (MA) domain or the NC zinc finger motifs altered condensate number and size in a salt-dependent manner. Gag BMCs were also bimodally influenced by the gRNA, with a condensate-promoting regime at lower protein concentrations and a gel dissolution at higher protein concentrations. Interestingly, incubation of Gag with CD4+ T cell nuclear lysates led to the formation of larger BMCs compared to much smaller ones observed in the presence of cytoplasmic lysates. These findings suggest that the composition and properties of Gag-containing BMCs may be altered by differential association of host factors in nuclear and cytosolic compartments during virus assembly. This study significantly advances our understanding of HIV-1 Gag BMC formation and provides a foundation for future therapeutic targeting of virion assembly. 相似文献

Interspecific hybridization between closely related mammalian species, including various species of the genus Mus, is commonly associated with abnormal growth of the placenta and hybrid foetuses, a phenomenon known as hybrid placental dysplasia (HPD). The role of HPD in speciation is anticipated but still poorly understood. Here, we studied placental and foetal growth in F1 crosses between four inbred mouse strains derived from two house mouse subspecies, Mus musculus musculus and Mus musculus domesticus. These subspecies are in the early stage of speciation and still hybridize in nature. In accordance with the maternal–foetal genomic conflict hypothesis, we found different parental influences on placental and foetal development, with placental weight most affected by the father's body weight and foetal weight by the mother's body weight. After removing the effects of parents’ body weight, we did not find any significant differences in foetal or placental weights between intra‐subspecific and inter‐subspecific F1 crosses. Nevertheless, we found that the variability in placental weight in inter‐subspecific crosses is linked to the X chromosome, similarly as for HPD in interspecific mouse crosses. Our results suggest that maternal–foetal genomic conflict occurs in the house mouse system, but has not yet diverged sufficiently to cause abnormalities in placental and foetal growth in inter‐subspecific crosses. HPD is thus unlikely to contribute to speciation in the house mouse system. However, we cannot rule out that it might have contributed to other speciation events in the genus Mus, where differences in the levels of polyandry exist between the species. 相似文献
