Extracellular proteins from Botrytis cinerea CBS 144.55 were separated by use of isoelectric focusing in polyacrylamide gel and thin layer isoelectric focusing. The involvement of the endomenbrane system of barley (Hordeum vulgare L.) aleurone cells in the secretion of gibberellin-induced hydrolases has been investigated at the biochemical level. Our results show that at least 40–60% of the -amylase activity in homogenates of aleurone layers occurs in a membrane-bound, latent form. The latent -amylase can be assayed quantitatively following disruption of membranes by treatment with Triton X-100, ethanol, sonication, or osmotic shock and shear. The association of -amylase with the membrane is not an artifact arising from homogenization of the tissue, and acid protease is also enriched in the same subcellular fraction as the -amylase. The membrane fraction with which the -amylase is associated has many properties of the endoplasmic reticulum (ER). When membranebound -amylase is prepared in buffers containing 3 mM MgCl2 two fractions from a sucrose step gradient contain most of the -amylase activity. These fractions are enriched in the ER marker enzyme, NADH-dependent cytochrome-c reductase, and show densities characteristic of smooth and rough ER during subsequent purification on continuous gradients. In step gradients prepared with ethylenediaminete-traacetic-acid-treated membranes, -amylase activity is contained primarily in one fraction having the density of smooth ER. Electron microscopy of the purified fractions is consistent with -amylase being associated with smooth and rough ER. However, it has not been ruled out that the enzyme is also associated with plasma membrane, Golgi membranes, or tonoplast. Summary The influence of repair and replication on the frequency of spontaneous chromosome aberrations and of those induced by gamma-irradiation is reported.Using the technique of labelling DNA with radioactive 3H-thymidine and measuring the radioactivity of DNA isolated from embryos, the time of initiation and the duration of DNA synthesis in barley seeds was studied after the soaking of the seeds had begun. The average duration of each phase of the first DNA synthesis cycle in soaking barley seeds was found to be as follows: pre-DNA synthesis stage, 10–11 hrs; DNA synthesis stage, 8 hrs. After gamma-irradiation, the intensity of DNA synthesis decreased and the beginning of DNA synthesis was delayed.It was found that the inhibition of repair by caffeine led to an increase in the frequency of both spontaneous and induced chromosome aberrations. Caffeine enhanced several times the frequency of chromosome and chromatid aberrations at the time of the maximal activity of repair enzymes. During DNA replication, caffeine had a lower effect on the realization of premutational lesions.An inhibitor of DNA replication — hydroxyurea — had no influence on the frequency of spontaneous chromosome aberrations during the replication period, whereas after gamma-irradiation, hydroxyurea enhanced the frequency of aberrations mainly at the stage of DNA replication.The relatively small mutagenic action of both agents (caffeine and hydroxyurea) was observed during all stages of the cell cycle of germinating barley seeds.  相似文献

Abstract The respiratory activity of cysts of Azotobacter vinelandii has been compared with that of vegetative cells. Whole cysts had a much reduced respiratory activity which was less sensitive to KCN. Substrate oxidation rates by membrane preparations from cysts were reduced approximately 10-fold and sensitivity to KCN was decreased by a similar factor. Difference spectra of cyst membranes revealed changes in cytochrome content. Cytochrome oxidase d was apparently absent, cytochrome a 1 levels were approximately halved whilst those of cytochrome oxidase o were almost doubled. Cytochromes of the b and c -type were present in similar amounts to those in vegetative cells.  相似文献

The complete genetic information contained in the influenza virus RNA segment 7 of the A/Bangkok/ 179 (H3N2) strain has been cloned by in vitro synthesis of the complementary dsDNA and its insertion into plasmid pBR322. The nucleotide sequence of the viral RNA segment has been determined from the cDNA insert. It is 1027 nucleotides long, and contains two open reading frames, as shown for other influenza virus strains. When compared with the previously published sequence for the A/Udorn/72 (H3N2) strain, 15 nucleotide exchanges are observed, most of them silent mutations, and only two causing amino acid changes in each of the M1 and M2 protein sequences.  相似文献

An endogenous soluble protease has been demonstrated to unmask a Ca2+-stimulated ATPase activity in purified dog gastric microsomes. The presence of ATP during protease treatment appears essential for the manifestation of the gastric Ca2+-stimulated ATPase activity. The endogenous protease appears to have trypsin-like activity, since soybean trypsin inhibitor completely blocks the protease effect. Manifestation of the Ca2+-stimulated ATPase occurs without affecting the microsomal (H+ +K+)-ATPase activity and associated H+ uptake ability. The unmasked Ca2+-stimulated ATPase appears insensitive to calmodulin. Possible roles of the enzyme in the regulation of gastric H+ transport have been discussed.  相似文献

将水稻(Oryza sativa L.)幼苗悬浮培养于含有羧基化多壁碳纳米管MWCNTs-COOH(0、2.5、5.0、10.0 mg/L)、50 mmol/L混合盐(1NaCl:9Na2SO4:9NaHCO3:1Na2CO3),以及MWCNTs-COOH+混合盐的复合溶液中,10 d后检测叶片生理生化指标变化,研究MWCNTs-COOH复合盐碱胁迫对水稻幼苗的毒性及生态风险。结果显示,与对照组相比,MWCNTs-COOH单一组诱导下水稻叶片O2·-和H2O2的产生不明显,而混合盐组和混合盐+MWCNTs-COOH复合组均诱导了O2·-和H2O2产物的大量累积。MWCNTs-COOH与混合盐复合后,加剧了O2·-和H2O2的累积,并有明显的浓度效应。活性氧(ROS)作为信号分子在一定程度上诱导了各处理组部分抗氧化酶(SOD、CAT、POD、APX)活性的升高;与混合盐组相比,低浓度混合盐+MWCNTs-COOH复合组中叶绿素a和胡萝卜素含量呈一定程度的升高;MWCNTs-COOH与混合盐复合后,抑制了叶片中可溶性糖(SS)和脯氨酸(Pro)的合成,致使相对电导率(REC)和丙二醛(MDA)含量显著升高。上述抗氧化酶活性及叶绿素a和胡萝卜素含量的升高对缓解水稻叶片氧化损伤、维持正常的光合电子传递及对过剩光能的热耗散是有益的,是水稻幼苗重要的防御机制。本研究表明MWCNTs-COOH单一处理在一定程度上诱导了水稻叶片的氧化胁迫和应激响应,与混合盐复合后加剧了叶片的氧化胁迫和应激损伤。  相似文献

The Cl/HCO 3 exchange mechanism usually postulated to occur in gastric mucosa cannot account for the Na+-dependent electrogenic serosal to mucosal Cl transport often observed. It was recently suggested that an additional Cl transport mechanism driven by the Na+ electrochemical potential gradient may be present on the serosal side of the tissue. To verify this, we have studied Cl transport in guinea pig gastric mucosa. Inhibiting the (Na+, K+) ATPase either by serosal addition of ouabain or by establishing K+-free mucosal and serosal conditions abolished net Cl transport. Depolarizing the cell membrane potential with triphenylmethylphosphonium (a lipid-soluble cation), and hence reducing both the Na+ and Cl electrochemical potential gradients, resulted in inhibition of net Cl flux. Reduction of short-circuit current on replacing Na+ by choline in the serosal bathing solution was shown to be due to inhibition of Cl transport. Serosal addition of diisothiocyanodisulfonic acid stilbene (an inhibitor of anion transport systems) abolished net Cl flux but not net Na+ flux. These results are compatible with the proposed model of a Cl/Na+ cotransport mechanism governing serosal Cl entry into the secreting cells. We suggest that the same mechanism may well facilitate both coupled Cl/Na+ entry and coupled HCO 3 /Na+ exit on the serosal side of the tissue.  相似文献

The stability of the serine proteases from Bacillus amyloliquefaciens (subtillisin BPN') and Bacillus licheniformis (subtilisin Carlsberg) was investigated in various anhydrous solvents at 45 degrees C. The half-life of subtilisin BPN' in dimethyl-formamide dramatically depends on the pH of the aqueous solutions from which the enzyme was lyophilized, increasing from 48 min to 20 h when the pH is raised from 6.0 to 7.9. Both subtilisins exhibited substantial inactivation during multihour incubations in tert-amyl alcohol and acetonitrile when enzymatic activities were also measured in these solvents; however, when the enzymes were assayed in water instead, hardly any loss of activity was detected. This surprising difference appears to stem from the partitioning of the bound water essential for catalytic activity from the enzymes into the solvents. When assayed in organic solvents, this time-dependent stripping of water results in decay of enzymatic activity; however, when assayed in water, where the dehydrated subtilisins can undergo rehydration thereby recovering catalytic activity, little inactivation is observed. In agreement with this hypothesis, the addition of small quantities of water tert-amyl alcohol stabilized the subtilisins in it even when enzymatic activity was measured in the nonaqueous solvent. Ester substrates (vinyl butyrate and trichloroethyl butyrate) greatly enhanced the stability of both subtilisins in organic solvents possibly because of the formation of the acyl-enzymes.  相似文献
