This study aimed to investigate whether the biological response modifiers (BRM) interferon (IFN) and tumour necrosis factor (TNF) could enhance the cytotoxic action of cisplatin on ovarian tumour cells in vitro. The sensitivity of four cell lines (OAW42, GG, JAM and PE01) to drugs and drug combinations was tested by a radiolabelled-thymidine incorporation assay. Cell lines demonstrated a range of sensitivity to cisplatin and the innate cytotoxic effect of each of the BRM. When IFN was used in combination with cisplatin, a significant enhancement of cisplatin toxicity occurred in three of four cell lines. TNF demonstrated such an effect in two cell lines but diminished the toxicity of cisplatin in one cell line. A purely additive effect of the agents may explain the enhanced toxicity of cisplatin in some of these cases. However, in one cell line at least (PEO1), both TNF and IFN demonstrated a clearly synergistic effect with cisplatin. These BRM used in conjunction with cisplatin may provide better antitumour regimen than cisplatin alone in some patients with ovarian cancer, but the response is likely to be heterogeneous between patients.  相似文献

The current studies demonstrate that MOPC-315 tumor cells secrete large amounts of interleukin-10 (IL-10), which contributes to the inhibitory activity of MOPC-315 culture supernatants for the in vitro generation of antitumor cytotoxicity by MOPC-315-immune spleen cells. Moreover, addition of neutralizing monoclonal anti-IL-10 antibody to the in vitro stimulation cultures of cells from the tumor infiltrated spleens of mice bearing a large MOPC-315 tumor resulted in the generation of enhanced anti-MOPC-315 cytotoxicity. In contrast, addition of monoclonal anti-IL-10 antibody to the in vitro stimulation cultures of splenic cells from mice that are in the final stages of immune-mediated tumor eradication as a consequence of low-dose melphalan (l-phenylalanine mustard; L-PAM) therapy (and whose spleens no longer contain metastatic tumor cells) did not lead to enhancement in the in vitro generation of antitumor cytotoxicity. The cessation of IL-10 secretion as a consequence of low-dose L-PAM therapy of MOPC-315 tumor bearers was found to be accompanied by the acquisition of the ability to secrete interferon (IFN) by the splenic cells. In addition, by day 2 after low-dose L-PAM therapy a drastic decrease in the amount of IL-10 secreted by the s.c. tumor nodules was noted, which preceded the accumulation of tumor-infiltrating lymphocytes capable of secreting IFN. Thus, low-dose L-PAM therapy of mice bearing a large MOPC-315 tumor leads to a shift in cytokine production from a Th2-type cytokine to a Th1-type cytokine, and it is conceivable that this shift in cytokine production plays an important role in the low-dose L-PAM-induced acquisition of antitumor immunity by hitherto immunosuppressed mice bearing a large MOPC-315 tumor.Supported by research grant IM-435 from the American Cancer Society and CA54413 from the National Cancer InstituteIn partial fulfillment of the requirements for the Doctor of Philosophy DegreeRecipient of career development award CA-01350 from the National Cancer Institute  相似文献

A membrane preparation from rat brain catalyzed the hydrolysis of [2-3H]glycerol-labeled lysophosphatidylinositol (lysoPI) to yield monoacylglycerol (MG) and inositolphosphates. This phospholipase C activity had an optimal pH of 8.2. The membrane preparation did not require the addition of Ca2+ for its maximum activity, but the activity was inhibited by addition of 0.1 mM EDTA to the assay mixture and was restored by simultaneous addition of 0.2 mM Ca2+. The activity was found to be localized in synaptic plasma membranes prepared by Ficoll and Percoll density gradients. The phospholipase C was highly specific for lysoPI; diacylglycerol formation from phosphatidylinositol, and MG formation from lysophosphatidylcholine, lysophosphatidylethanolamine, and lysophosphatidylserine were below 5% of that observed with lysoPI under the conditions used. The conversion of 5-hydroxytryptamine to several potential metabolites was examined in the annelid earthworm (Lumbricus terrestris). 5-hydroxytryptamine and some related amines were found to be present in several tissues of the earthworm. Injection of 5-hydroxytryptamine into the body cavity of the earthworm resulted in the production of a -glutamyl conjugate of 5-hydroxytryptamine. Incubations of the anterior nerve cord of the earthworm resulted in the accumlation of considerable amounts of 5-hydroxytryptamine and -glutamyl 5-hydroxytryptamine in the incubation medium. The earthworm did not produce any N-acetyl 5-hydroxytryptamine and only very little 5-hydroxyindoleacetic acid. Experiments involving the injection of radiolabeled 5-hydroxytryptamine or coninjection of radiolabeled glutamic acid with unlabeled 5-hydroxytryptamine into the earthworm resulted in the production of radiolabeled -glutamyl 5-hydroxytryptamine. This work demonstrates that the enzymatic conversion of 5-HT in the earthworm is markedly different from that of vertebrates and insects.  相似文献

Synopsis The pelagic (589 spp.) and demersal (505 spp.) oceanic ichthyofaunas of the North Atlantic Basin have very different compositions at ordinal and family level. Yet the pattern of relationships between species' maximum size and maximum fecundity from data available (10% of the pelagic, 19% of the demersal species) was similar. A positive relationship between fecundity and weight was confirmed among most teleosts, but was not followed by the elasmobranchs represented. Species' reproductive styles are reviewed in ordinal groupings within a framework of the overall body size/fecundity distribution. Species size (maximum weight) spectra were synthesized for both pelagic and demersal assemblages to assess the allocation of potential reproductive effort throughout the North Atlantic oceanic ichthyofauna. The only available examples of species size spectra and biomass spectra from the pelagic and demersal ichthyofauna in this ocean basin imply geographic and bathymetric variation in overall reproductive effort among fishes whose fecundity is size dependent. Further implications concerning reproductive effort are discussed in the light of food availability.  相似文献

Synopsis Similarities between the freshwater fish faunas of 52 west African rivers have been investigated and three main zoogeographic regions recognized. The Sudanian region includes all rivers from Senegal to the Omo, as well as coastal basins from Ivory Coast to the Cross and the Wouri. The Upper Guinean region comprises the coastal basins from Guinea to Liberia and the Lower Guinean one, the coastal rivers of Cameroon and Gaboon. The Sudanian region can be sub-divided into a Sudanian region sensu stricto, including the Sahelo-Sudanese rivers, and the Eburneo-Ghanean region corresponding to coastal basins from the Cess (or Nipoué, Ivory Coast) to the Pra (Ghana). These delimitations give an highly significant within region faunal homogeneity, even if the effect of geographical proximity between rivers is removed. 21 to 71% of the fish species in each region are endemics. The present patterns of distribution are the result of past climatic and geological events affecting west Africa and, given this framework, the role of alternating wet and dry periods during the early Quarternary is emphasized as well as the importance of mountains as dispersal barriers. Role of recent river connections and links via lagoon is emphasized in explaining river faunal similarities within biogeographical regions.  相似文献

在昆明地区,纵坑切梢小蠹(Tomicus piniperda)成虫蛀梢多集中在蛀干木附近。 种群密度以蛀干木为中心向周围呈指数递减,散布半径约30m。在蛀梢过程中,该种群逐渐向新区扩张。在树冠内,纵坑切梢小蠹主要分布在4-10轮枝上。第7轮枝虫口百分率最高。6-7轮枝受害率最大。 树冠上层受害较其下层严重。从树冠水平层次考察,树冠外层虫量相对集中,约为树冠中、内层虫量之和。 树冠内层虫量最少。纵坑切梢小蠹在树冠内的种群分布系由梢径、种群密度、蛀梢行为、降落方式、光照等因素综合影响的结果。  相似文献

稻瘿蚊为害的空间分布型属聚集分布,田间估计稻瘿蚊为害的最佳取样方法为棋盘法和平行线法,当每丛受害超过1苗时,抽样数量可稳定在151丛以下。标葱率和有效穗相关性测定表明:当标葱率在20%以下时,为害程度与有效穗的多少无显著相关;当标葱率大于20%时,标葱率与有效穗呈显著负相关,每受害加重5%,有效穗将减少0.37个/丛。  相似文献
