Summary The deffects of a purified homologue of tunicamycin (B2-tunicamycin) on the biosynthesis of lipid-linked intermediates participating in protein glycosylation in normal embryonic fibroblasts, 3T3 and virally transformed (simian virus 40 and polyoma virus) mouse fibroblasts grown in culture were investigated. Long incubations (20 h) with the antibiotic caused a higher degree of inhibition of sugar incorporation into glycoproteins in transformed cells. However, the formation of lipid-linked intermediates was inhibited to a similar level in both cell types. When time dependent inhibition experiments were carried out using transformed cells, an earlier and stronger inhibition of the formation of lipid-oligosaccharides occurred (70% inhibition at 30 min). In 3T3 cells, prolonged incubation (6–8 h) was necessary in order to reach a similar degree of inhibition. Formation of lipid-sugar was also inhibited to a greater extent by B2-tunicamycin in transformed cells. This inhibition was not clearly time dependent. Analysis of the newly synthesized glycolipids in 3T3 and in transformed cells after B2-tunicamycin treatment have shown reduction in dolichyl-P-P-sugars as well as in other glycolipids. Dimethylsulfoxide (10%) and linoleic acid (0.5 mg/ml) markedly increased the level of tunicamycin activity in 3T3 cells while phosphatidylcholine (2 mg/ml) partially reversed it. The stronger and faster inhibition of the formation of lipid intermediates of the dolichyl-phosphate cycle caused by B2-tunicamycin in transformed cells, described here for the first time, may therefore be due to differences in penetration of the antibiotic into these cells.Abbreviations DMEM

The wake and sleep-onset times of a patient with a sleep-wake cycle longer than 24 hr were recorded by the patient for 4 years. During this time, the patient found himself unable to maintain a 24-hr sleep-wake schedule. When treated with 1-2 mg clonazepam, taken nightly, he was able to become entrained to a 24-hr day. Glutathione (GSH) dissolved in Eagle's MEM and added to cultures o of V79-E cells in concentrations between 2.5 × 10–4 and 10–3 moles/l for 1 h induces a dose-dependent cell cycle delay, sister chromatid exchanges and clastogenic damage. 7–8% of the metaphases showed endoreduplication at a recovery phase of 25 and 30 h after treatment with 10–3 molesll GSH. Higher concentrations were lethal. The highest tolerated dose corresponds to the intracellular GSH level in V79-E cells. In the same range of concentrations, glutathione disulfide was inactive. Endoreduplication induction by GSH is G2-phase specific and endoreduplication metaphases show a reduced occurrence of single SCEs when extrapolated to the diploid complement. The adverse effects of GSH are independent of the presence of serum in the culture fluid but completely abolished when the treatment is performed in Hank's solution instead of MEM. The mechanism of genotoxicity of exogenous GSH is discussed but, at present, no pertinent explanation can be given.Abbreviations BUdR

The penetration of metacyclic forms of Trypanosoma cruzi into HeLa cells after different treatments was studied. When cell development was synchronized by two different processes, maximum rates of parasitization occurred during the S phase of cell cycle (29.48 and 24.3%). However, when cells were treated with trypsin (0.1%), parasitization rates appeared to be lower than controls, reaching values similar to controls 14 h after the beginning of the treatment. Infection values remained unaltered after treatment with colcemid (0.6 μg ml?1). Cell treatment either with valinomycin (1 μg ml?1) or with actinomycin D (250 μg ml?1) caused a marked decrease in the percentage of parasitization. When cells were treated and infected in the presence of tunicamycin (100 ng ml?1), parasitization rates were increased (14.7%) compared to control cells (6%). On the other hand, no differences in parasitization rates were observed when cells were treated with cycloheximide (100 μg ml?1). Investigations were undertaken to determine the applicability of recently reported specific radioimmunoassays for urinary estrone conjugates and progesterone metabolites for monitoring ovarian function in the cynomolgus macaque (Macaca fasciularis) and other macaque species. Mean estrone conjugate measurements appear to accurately reflect the preovulatory estrogen peak in both conceptive (n = 5) and nonconceptive (n = 6) cycles, as well as to indicate early pregnancy through increases which are significantly elevated by Day + 15 (p less than 0.049) post estrone conjugates peak. The mean luteal phase levels of these progesterone metabolites are significantly elevated by Day + 14 (p less than 0.012) in conceptive cycles when compared to the mean values for nonconceptive cycles. 相似文献

The Q cycle and theb cycle are the main current models of action of the cytochromebc-type complexes of mitochondria, bacteria, and chloroplasts. Both are based on the concept, proposed in 1972, of two sequential one-electron oxidations of (ubi)quinol along two discrete pathways which operate at different redox potentials, and with bound semiubiquinone as an intermediate. The models differ in two respects, viz. in the pathway of electron transfer and the principle of linkage of electron transfer to proton translocation. The firing pattern of neural pulses often show the following features: the shapes of individual pulses are nearly identical and frequency independent; the firing frequency can vary over a broad range; the time period between pulses shows a stochastic scatter. This behaviour cannot be understood on the basis of a deterministic non-linear dynamic process, e.g. the Bonhoeffer-van der Pol model. We demonstrate in this paper that a noise term added to the Bonhoeffer-van der Pol model can reproduce the firing patterns of neurons very well. For this purpose we have considered the Fokker-Planck equation corresponding to the stochastic Bonhoeffer-van der Pol model. This equation has been solved by a new Monte Carlo algorithm. We demonstrate that the ensuing distribution functions represent only the global characteristics of the underlying force field: lines of zero slope which attract nearby trajectories prove to be the regions of phase space where the distributions concentrate their amplitude. Since there are two such lines the distributions are bimodal representing repeated fluctuations between two lines of zero slope. Even in cases where the deterministic Bonhoeffer-van der Pol model does not show limit cycle behaviour the stochastic system produces a limit cycle. This cycle can be identified with the firing of neural pulses. 相似文献
