A new split β‐lactamase assay promises experimental testing of the interplay of protein stability and function. Proteins are sufficiently stable to act effectively within cells. However, mutations generally destabilize structure, with effects on free energy that are comparable to the free energy of folding. Assays of protein functionality and stability in vivo enable a quick study of factors that influence these properties in response to targeted mutations. These assays can help molecular engineering but can also be used to target important questions, including why most proteins are marginally stable, how mutations alter structural makeup, and how thermodynamics, function, and environment shape molecular change. Processes of self‐organization and natural selection are determinants of stability and function. Non‐equilibrium thermodynamics provides crucial concepts, e.g., cells as emergent energy‐dissipating entities that do work and build their own parts, and a framework to study the sculpting role of evolution at different scales.  相似文献

The sexual ontogeny of butterfish Odax pullus was examined in the Hauraki Gulf of New Zealand through histological analysis of gonad material, size and age information and seasonal patterns of sexual maturation. The patterns of gonad development and schedules of male recruitment were established and sexual ontogeny of O. pullus was diagnosed as monandric protogyny, with all males developing from mature females after female-to-male sex reversal. All individuals underwent an immature female phase before maturing as functional females at 228·7-264·8 mm fork length (L(F) ) and at 1·1-1·5 years of age, and there was no evidence of a juvenile bisexual phase. Degenerating mature oogenic elements were found in the gonad lumen of individuals with developing spermatogenic tissue, providing histological evidence for functional protogyny. Sex change was estimated to occur at 359-379 mm L(F) and 2-3 years of age. The diagnosis of monandric protogyny for O. pullus coincided with the pattern of sexual ontogeny seen in the majority of labrids, particularly those of the same clade (tribe Hypsigenyini) and contrasted with that seen in a number of other temperate labrids. This study suggests that the protogynous mode of sexual development in O. pullus is likely to be lineage-specific, i.e. associated with the phylogeny of labrid sexual development, and is not constrained by environmental effects on the evolution of sex change in temperate regions.  相似文献

Apertures are key characters of pollen grains with systematic importance in angiosperms. They function as sites for pollen tube exit, water uptake, transfer of recognition substances and accommodation of volume changes. Not all pollen has apertures; inaperturate pollen (lacking obvious apertures) characterizes many angiosperm groups, especially in early divergent angiosperms and monocots, but also eudicots. In order to expand our knowledge of the systematic distribution, possible functional significance and development of inaperturate pollen in angiosperms, this review focuses on inaperturate and cryptoaperturate (with hidden apertures) pollen in the large eudicot clade, which comprises about 75% of present‐day angiosperm species. It includes new TEM observations of inaperturate pollen from four exemplar taxa selected from different parts of the eudicot phylogeny. Two categories of inaperturate (including cryptoaperturate) pollen occur in eudicots. (1) Sterile attractant or feeding pollen associated with functional dioecy has evolved iteratively at least six times in conjunction with complex breeding systems in the core eudicots. (2) Fertile pollen has evolved numerous times independently throughout eudicots, though generally in a relatively small number of individual taxa. Notable exceptions are the petaliferous crotonoid Euphorbiaceae s.s., in which fertile inaperturate pollen occurs in c. 1500 species, and two subfamilies of Apocynaceae s.l. (Secamonoideae and Asclepiadoideae) with c. 2500 species with fertile inaperturate pollen in pollinia. Fertile inaperturate pollen is sometimes (but not always) associated with an aquatic habit, parasitism, insectivory, heterostyly, anemophily or pollinia. Most fertile inaperturate pollen has a thin exine, or the exine is largely restricted to isolated components (muri, protuberances, subunits) separated by thinner areas which probably function as apertures. In cryptoaperturate pollen, the aperture is covered by continuous exine which probably has a protective function, similar to an operculum. Developmentally, inaperturate pollen is not associated with any particular tetrad type or meiotic spindle orientation (unlike some apertures) due to the absence of a colpal shield of endoplasmic reticulum or other organelles and hence is independent of microsporogenesis type. Nearest neighbor analysis of immunocytolocalization experiments indicates that the enzymes glyceraldehyde-3-P dehydrogenase, triose-P isomerase and aldolase are located close to one another in the pea leaf chloroplast stroma, and that aldolase is located close to sedoheptulose bisphosphatase. Direct transfer of the triose phosphates between glyceraldehyde-3-P dehydrogenase and triose-P isomerase, and from glyceraldehyde-3-P dehydrogenase and triose-P isomerase to aldolase, is then a possibility, as is direct transfer of sedoheptulose bisphosphate from aldolase to sedoheptulose bisphosphatase. Spatial organization of these enzymes may be important for efficient CO2 fixation in photosynthetic organisms. In contrast, there is no indication that fructose bisphosphatase is co-localized with aldolase, and direct transfer of fructose bisphosphate from aldolase to fructose bisphosphatase seems unlikely.  相似文献

Analysis of genomes has revealed that the total number of human genes is comparable to those of simpler organisms, and thus, the number of genes does not correlate with the complexity and functional diversity of different organisms. Multiple mechanisms, including alternative splicing, are believed to contribute to the molecular complexity in higher eukaryotes. Given the fact that more than half of human genes undergo alternative splicing, however, little is known about the biological relevance of most alternative splicing events and their regulatory mechanisms. Recent work has highlighted the power of reverse genetic approaches in addressing regulated splicing in animal models. Here, we focus on the conditional knockout approach adapted for splicing research with the intention to provide a general guide to the generation of mouse models to study regulated splicing in development and disease.  相似文献

Northeast China Transect (NECT), one of the fifteen International Biosphere-Geosphere Programme (IGBP) terrestrial transects, has been established for 10 years by Prof. Zhang Xin-Shi, through a core project of the IGBP - the Global Change and Terrestrial Ecosystems (GCTE). This transect is located in the mid-latitude semi-arid region, ranging 42-46°N latitude and 110-132癊 longitude. The primary driving force for global change is precipitation and the secondary one is land use intensity. Research progresses have been performed during the past decade in the following aspects: ecological database development, climate and its variability, ecophysiological response of plants to environments, vegetation and landscape changes, biodiversity patterns and their changes, plant functional types and traits with relation to climatic gradient, productivity and carbon dynamics, pollen-vegetation relationship, trace gas emissions, land use and land cover changes, as well as biogeographical and biogeochemical modelling. In order to achieve the higher level of integrated research, the NECT needs the consistent basic data sets within the same framework, further field experiments and observations, integrated simulations of vegetation structure, process and function from patch, landscape to biome scales, intercomparisons of results and simulations within the transect and to other IGBP transects, multidisciplinary research, national and international co-ordinates, and full scientific plan and implementation strategy.  相似文献

The present study contrasts available biological data and results of morphofunctional analyses of the bill and hyoid apparatus in motmots. It shows that these omnivorous birds, which take relatively large food items, possess osteomuscular peculiarities that enable them to process these items as a whole in order to soften or cut them, and make them suited for easy ingestion. For that, they use the crenate edges of their rhamphotheca. Their jaws work as a highly mobile saw-like system. Their mutual movements, enhanced by the fact that particular dispositions of the hyoid apparatus rise the tongue and the supported items high up into buccal cavity, facilitate an effective clamping of items that can be moved along the jaws and be quite appropriately processed.  相似文献
