Basic knowledge of the previous forest types or ecosystem present in an area ought to be an essential part of all landscape restoration. Here, we present a detailed study of forest and land use history over the past 2,000 years, from a large estate in southernmost Sweden, which is currently undergoing a restoration program. In particular, the aim was to identify areas with long continuity of important tree species and open woodland conditions. We employed a multidisciplinary approach using paleoecological analyses (regional and local pollen, plant macrofossil, tree ring) and historical sources (taxation documents, land surveys, forest inventories). The estate has been dominated by temperate broad‐leaved trees over most of the studied period. When a forest type of Tilia, Corylus, and Quercus started to decline circa 1,000 years ago, it was largely replaced by Fagus. Even though extensive planting of Picea started in mid‐nineteenth century, Fagus and Quercus have remained rather common on the estate up to present time. Both species show continuity on different parts of the estate from eighteenth century up to present time, but in some stands, for the entire 2,000 years. Our suggestions for restoration do not aim for previous “natural” conditions but to maintain the spatial vegetational pattern created by the historical land use. This study gives an example of the spatial and temporal variation of the vegetation that has historically occurred within one area and emphasizes that information from one methodological technique provides only limited information about an area’s vegetation history.  相似文献

A sensitivity factor that quantifies the responsiveness of stomata to xylem sap abscisic acid concentration ([ABA]xyl) is described, using the relation between [ABA]xyl and maximum leaf conductance (gmax). Plotting gmax against this factor results in a common linear relationship for woody and herbaceous species from boreal to (semi-) arid climates. The global distribution of the sensitivity factor reveals an unexpected pattern which is inverse to rainfall, i.e., plants in humid climates respond more sensitively to ABA than plants in arid areas. The implications for the response of natural vegetation and consequences for agriculture are discussed.  相似文献

Currently, the identification of groups of amino acid residues that are important in the function, structure, or interaction of a protein can be both costly and prohibitively complex, involving vast numbers of mutagenesis experiments. Here, we present the application of a novel computational method, which identifies the presence of coevolution in a data set, thereby enabling the a priori identification of amino acid residues that play an important role in protein function. We have applied this method to the heat shock protein (Hsp) protein-folding system, studying the network between Hsp70, Hsp90, and Hop (heat shock-organizing protein). Our analysis has identified functional residues within the tetratricopeptide repeat (TPR) 1 and 2A domains in Hop, previously shown to be interacting with Hsp70 and Hsp90, respectively. Further, we have identified significant residues elsewhere in Hop within domains that have been recently proposed as being important for Hop interaction with Hsp70 and/or Hsp90. In addition, several amino acid sites present in groups of coevolution were identified as 3-dimensionally or linearly proximal to functionally important sites or domains. Based on our results, we also investigate a further functional domain within Hop, between TPR1 and TPR2A, which we suggest as being functionally important in the interaction of Hop with both Hsp70 and Hsp90 whether directly or otherwise. Our method has identified all the previously characterized functionally important regions in this system, thereby indicating the power of this method in the a priori identification of important regions for site-directed mutagenesis studies.  相似文献

It currently comprises libraries of temperature-sensitive mutants in essential genes, and insertional mutants in non-essential genes, available for screening by visiting workers. Background

Tautomerization of dopachrome to 5,6-dihydroxyindole-2-carboxylic acid (DHICA) is a biologically crucial reaction relevant to melanin synthesis, cellular antioxidation, and cross-talk among epidermal cells. Since dopachrome spontaneously converts into 5,6-dihydroxyindole (DHI) via decarboxylation without any enzymes at physiologically usual pH, the mechanism of how tautomerization to DHICA occurs in physiological system is a subject of intense debate. A previous work has found that Cu(II) is an important factor to catalyze the tautomerization of dopachrome to DHICA. However, the effect of Cu(II) on the tautomerization has not been clarified at the atomic level.

Methods

We propose the reaction mechanism of the tautomerization to DHICA by Cu(II) from density functional theory-based calculation.

Results

We clarified that the activation barriers of α-deprotonation, β-deprotonation, and decarboxylation from dopachrome are significantly reduced by coordination of Cu(II) to quinonoid oxygens (5,6-oxygens) of dopachrome, with the lowest activation barrier of β-deprotonation among them. In contrast to our previous work, in which β-deprotonation and quinonoid protonation (O5/O6-protonation) were shown to be important to form DHI, our results show that the Cu(II) coordination to quinonoid oxygens inhibits the quinonoid protonation, leading to the preference of proton rearrangement from β-carbon to carboxylate group but not to the quinonoid oxygens.

Conclusion

Integrating these results, we conclude that dopachrome tautomerization first proceeds via proton rearrangement from β-carbon to carboxylate group and subsequently undergoes α-deprotonation to form DHICA.

General significance

This study would provide the biochemical basis of DHICA metabolism and the generalized view of dopachrome conversion which is important to understand melanogenesis.  相似文献
