Two sub-strains of the anoxygenic photosynthetic green sulfur bacterium Chlorobium vibrioforme NCIB 8327 were derived from the same clone and could be discriminated only by their possession of either bacteriochlorophyll (BChl) c or d as the major pigment in the peripheral light-harvesting antenna system, chlorosome (Saga Y et al. (2003) Anal Sci 19: 1575–1579). In the presence of a proper amount of oxygen in the initial culture medium, the BChl d strain showed longer retardation on its growth initiation than the BChl c strain, indicating that the latter was advantageous for survival under aerobic light conditions which produced reactive oxygen species in vivo. The result would be ascribable to the difference of the midpoint potentials between two kinds of chlorosomes formed by self-aggregates of BChl c and d as measured by their fluorescence quenching.  相似文献

The teeth of the oral jaws of two sympatric species of Argyrosomus, Argyrosomus japonicus and Argyrosomus inodorus, found along the South African coast developed first on the premaxilla and then on the dentary of the lower jaw. Teeth were observed on the premaxilla of A. inodorus [head length (LH) = 1·0 mm; notochord length (LN) = 2·7 mm] at a smaller size than in A. japonicus (LH = 1·2 mm; LN = 4·7 mm). The ventral elements of the gill arches (hypo‐ and basibranchials) were not ossified by the end of preflexion. The fifth ceratobranchial began ossifying and possessed pharyngeal teeth by 1·2 mm LH (LN = 4·7 mm) in A. japonicus and 1·1 mm LH (LN = 3·2 mm) in A. inodorus. To complement the osteological data, stomach contents were also analysed as a proxy for feeding apparatus functionality. Prey were first present in the stomach of A. japonicus at 1·2 mm LH (LN = 4·7 mm) and only 22% of the stomachs contained no prey suggesting that A. japonicus is already actively foraging by preflexion. In comparison, 83% of the stomachs of A. inodorus contained no prey and a single prey item was present in the largest examined specimen (LH = 1·6 mm; LN = 5·4 mm). Elements of the feeding apparatus begin to ossify early during ontogeny. While the overall pattern of ossification is similar between the two species, A. japonicus may be able to begin feeding at smaller head lengths relative to A. inodorus in their nursery habitats.  相似文献

ABSTRACT Bacteria grow on avian eggshells and thus can potentially cause diseases in developing embryos. Little is known about culturable bacteria colonizing avian eggshells in free‐living birds, with most studies restricted to poultry. Our objective was to examine the culturable bacterial array growing on eggshells during incubation that could negatively affect hatching success of Pied Flycatchers (Ficedula hypoleuca) in a temperate montane habitat in central Spain. Cloacal culturable bacteria of females were also analyzed because bacteria can be vertically transmitted from females to eggs. We used fecal samples as surrogates of cloacal samples due to the small size of sampled birds. We found that eggshells and female cloacae of Pied Flycatchers harbored 24 and 40 bacterial families and species, respectively, but only a few in each clutch and each cloaca. Rod‐shaped gram‐negative bacteria and bacteria in the family Pseudomonadaceae were the most common bacteria on eggshells during early and late incubation and in female cloacae. Although based on small sample sizes, we found that females with rod‐shaped gram‐negative bacteria in their cloacae laid eggs that also had these bacteria, providing possible evidence for vertical transmission. We found no evidence for vertical transmission of Pseudomonadaceae, suggesting a possible environmental source for these bacteria. The prevalence of bacterial morphological types and major taxonomical categories on eggshells did not vary from early to late stages of incubation, providing support for the hypothesis that incubation may have bacteriostatic effects on bacterial proliferation on eggshells. Despite being primary egg invaders in poultry, we detected no effects of culturable Pseudomonadaceae or Pseudomonas luteola on hatching success. Our study represents the first to examine the culturable bacteria growing on the eggshells of a wild bird in a temperate habitat and additional studies based on culture‐independent techniques are required to confirm our results.  相似文献

Elucidating functions of commensal microbial genes in the mammalian gut is challenging because many commensals are recalcitrant to laboratory cultivation and genetic manipulation. We present Temporal FUnctional Metagenomics sequencing (TFUMseq), a platform to functionally mine bacterial genomes for genes that contribute to fitness of commensal bacteria in vivo. Our approach uses metagenomic DNA to construct large‐scale heterologous expression libraries that are tracked over time in vivo by deep sequencing and computational methods. To demonstrate our approach, we built a TFUMseq plasmid library using the gut commensal Bacteroides thetaiotaomicron (Bt) and introduced Escherichia coli carrying this library into germfree mice. Population dynamics of library clones revealed Bt genes conferring significant fitness advantages in E. coli over time, including carbohydrate utilization genes, with a Bt galactokinase central to early colonization, and subsequent dominance by a Bt glycoside hydrolase enabling sucrose metabolism coupled with co‐evolution of the plasmid library and E. coli genome driving increased galactose utilization. Our findings highlight the utility of functional metagenomics for engineering commensal bacteria with improved properties, including expanded colonization capabilities in vivo.  相似文献

The protonmotive force in several sulfate-reducing bacteria has been determined by means of radiolabelled membrane-permeant probes (tetraphenyl-phosphonium cation, TPP+, for , and benzoate for pH). In six of ten freshwater strains tested only the pH gradient could be determine, while the membrane potential was not accessible due to nonspecific binding of TPP+. The protonmotive force of the other four strains was between –110 and –155 mV, composed of a membrane potential of –80 to –140 mV and a pH gradient between 0.25 and 0.8 (inside alkaline) at pHout=7. In Desulfobulbus propionicus the pH gradient decreased with rising external pH values. This decrease, however, was compensated by an increasing membrane potential. Sulfate, which can be highly accumulated by the cells, did not affect the protonmotive force, if added in concentrations of up to 4 mM. The highest sulfate accumulation observed (2500-fold), which occurred at external sulfate concentrations below 5 M, could be explained by a symport of three protons per sulfate, if equilibrium with the protonmotive force was assumed. At higher sulfate concentrations the accumulation decreased and suggested an electroneutral symport of two protons per sulfate. 本文通过从污水处理厂活性污泥中分离得到一株高效降解轻油(煤油)细菌5092-2,通过形态学、16S rRNA鉴定该菌为Mangroveibacter sp.。进一步对其生长条件进行了研究,发现该菌在23℃到38℃、pH为5.0到9.0范围内生长无明显差异。在温度为35℃,pH 7.2~7.4,160 r/min培养7 d,菌株降解煤油的能力达到52.3%,具有进一步研究的价值。  相似文献

Bacterial communities and metabolites in kimchi fermented under conventional conditions (CC) compared to CO2-rich environments (CO2) were analyzed. After a 20-day fermentation, lactic and acetic acid productions were 54 and 69 mM under CC, and 19 and 12 mM under CO2, respectively. The final pH of kimchi fermented under CC (CC-fermenting) and CO2 (CO2-fermenting) were 4.1 and 4.7, respectively. For bacterial communities, OTU and Chao1 indices were both 35 in fresh kimchi, 10 and 15 in CC-fermenting kimchi, and 8 and 24 in CO2-fermenting kimchi, respectively. Shannon and Simpson indices were 3.47 and 0.93 in fresh kimchi, 1.87–0.06 and 0.46–0.01 in CC-fermenting kimchi, and 1.65–0.44 and 0.63–0.12 in CO2-fermenting kimchi, respectively. Non-lactic acid bacteria were eliminated in fermenting kimchi after 12 days under CC and 6 days under CO2. I conclude that carbon dioxide can alter bacterial communities, reduce metabolite production, and improve fermented kimchi quality.  相似文献
