The integral membrane protein complex, menaquinol:fumarate oxidoreductase (mQFR) has been purified, identified and characterized from the thermophilic green filamentous anoxygenic photosynthetic bacterium Chloroflexus aurantiacus. The complex is composed of three subunits: a 74 kDa flavoprotein that contains a covalently bound flavin adenine dinucleotide, a 28 kDa iron-sulfur cluster-containing polypeptide, and a 27 kDa transmembrane polypeptide, which is also the binding site of two b-type hemes and two menaquinones. The purified complex has an apparent molecular mass of 260 kDa by blue-native PAGE, which is indicative of a native homodimeric form. The isolated complex is active in vitro in both fumarate reduction and succinate oxidation. It has been analyzed by visible absorption, redox titration, chemical analysis and EPR spectroscopy. In addition, phylogenetic analysis shows that the QFR of both C. aurantiacus and Chlorobium tepidum are most closely related to those found in the delta-proteobacteria. 摘要 目的:探讨五灵胶囊联合富马酸丙酚替诺福韦片对慢性乙型肝炎(CHB)患者氧化应激、CD4+CD25+调节性T细胞和血清基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶组织抑制因子-1(TIMP-1)的影响。方法:纳入苏州大学附属传染病医院2021年6月~2022年12月期间收治的CHB患者122例,采用随机数字表法分为对照组(n=61,富马酸丙酚替诺福韦片)和研究组(n=61,五灵胶囊联合富马酸丙酚替诺福韦片)。对比两组疗效、氧化应激指标、CD4+CD25+调节性T细胞、肝功能指标[总胆红素(TBIL)、谷丙转氨酶(ALT)、谷氨酰转肽酶(GGT)]、血清MMP-1、MMP-2、TIMP-1水平和不良反应发生情况。结果:研究组的临床总有效率高于对照组(P<0.05)。两组治疗后丙二醛(MDA)下降,且研究组低于对照组同时间点;超氧化物歧化酶(SOD)升高,且研究组高于对照组同时间点(P<0.05)。两组治疗后CD4+CD25+调节性T细胞下降,且研究组低于对照组同时间点(P<0.05)。两组治疗后MMP-1、MMP-2、TIMP-1下降,且研究组低于对照组同时间点(P<0.05)。两组治疗后TBIL、ALT、GGT下降,且研究组低于对照组同时间点(P<0.05)。两组不良反应总发生率组间对比未见差异(P>0.05)。结论:五灵胶囊联合富马酸丙酚替诺福韦片治疗CHB患者,可有效减轻机体氧化应激,调节CD4+CD25+调节性T细胞和血清MMP-1、MMP-2、TIMP-1水平。 相似文献

The characteristic respiratory metabolism of parasites consists of fermentation to carbon-rich, highly reduced volatile fatty acids which are excreted, and electron transport systems emphasising fumarate reductase and b-type cytochromes. The taxonomic groups that contribute major parasites (the heterogeneous protozoa and the helminths) have their evolutionary origins in environments from which oxygen was absent or present in very low concentrations. The Ediacarian period, about 700 million years ago, contains fossils of the appropriate grade of organisation to be contemporaneous with the ancestors of platyhelmiths, nematodes and acanthocephalans. With the oxygen transition, carbon flow in the biosphere resulted in conservative, anoxic environments together with oxygen rich ones. The organisms of the former retained their emphasis on anaerobic energy generation, while cytochrome systems were as much concerned with oxygen detoxification as energy generation. This study investigated the effects of disodium fumarate (DF) on methane emission, ruminal fermentation and microbial abundance in goats under different forage (F) : concentrate (C) ratios and fed according to maintenance requirements. Four ruminally fistulated, castrated male goats were used in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments and the main factors being the F : C ratios (41 : 59 or 58 : 42) and DF supplementation (0 or 10 g/day). DF reduced methane production (P < 0.05) on average by 11.9%, irrespective of the F : C ratio. The concentrations of total volatile fatty acids, acetate and propionate were greater in the rumen of goats supplemented with DF (P < 0.05), whereas the abundance of methanogens was lower (P < 0.05). In high-forage diets, the abundance of Selenomonas ruminantium, a fumarate-reducing bacterium, was greater in the rumen of goats supplemented with DF. The abundance of fungi, protozoa, Ruminococus flavefaciens and Fibrobacter succinogenes were not affected by the addition of DF. Variable F : C ratios affected the abundance of methanogens, fungi and R. flavefaciens (P < 0.05), but did not affect methane emission. The result implied that DF had a beneficial effect on the in vivo rumen fermentation of the goats fed diets with different F : C ratios and that this effect were not a direct action on anaerobic fungi, protozoa and fibrolytic bacteria, the generally recognized fiber-degrading and hydrogen-producing microorganisms, but due to the stimulation of fumarate-reducing bacteria and the depression of methanogens. 相似文献

Jumonji C domain-containing (JMJD) proteins are mostly epigenetic regulators that demethylate histones. However, a hitherto neglected subfamily of JMJD proteins, evolutionarily distant and characterized by their relatively small molecular weight, exerts different functions by hydroxylating proteins and RNA. Recently, unsuspected proteolytic and tyrosine kinase activities were also ascribed to some of these small JMJD proteins, further increasing their enzymatic versatility. Here, we discuss the ten human small JMJD proteins (HIF1AN, HSPBAP1, JMJD4, JMJD5, JMJD6, JMJD7, JMJD8, RIOX1, RIOX2, TYW5) and their diverse physiological functions. In particular, we focus on the roles of these small JMJD proteins in cancer and other maladies and how they are modulated in diseased cells by an altered metabolic milieu, including hypoxia, reactive oxygen species and oncometabolites. Because small JMJD proteins are enzymes, they are amenable to inhibition by small molecules and may represent novel targets in the therapy of cancer and other diseases. 相似文献

Overexpression of γ-tubulin leads to the formation of filaments, but nothing is known about such filaments with regard to possible presence in cells, structure and probable dynamics. Here, we used mammalian cell lines to investigate the ability of γ-tubulin to form filaments. We found that γ-tubulin produces fibers called γ-tubules in a GTP-dependent manner and that γ-tubules are made up of pericentrin and the γ-tubulin complex proteins 2, 3, 5 and 6. Furthermore, we noted that the number of cells with cytosolic γ-tubules is increased in non-dividing cells. Our experiments showed that γ-tubules are polar structures that have a low regrowth rate compared to microtubules. Also, we observed that γ-tubules were disassembled by treatment with cold, colcemid, citral dimethyl acetal, dimethyl fumarate or mutation of γ-tubulin GTPase domain, but were increased in number by treatment with taxol or by stable expression of the γ-tubulin1–333 GTPase domain. Our results demonstrate that γ-tubulin forms filaments, and such assembly is facilitated by the GTPase domain of γ-tubulin. 相似文献
