Neuromuscular electrical stimulation (NMES) is used for preventing muscle atrophy and improving muscle strength in patients and healthy people. However, the current intensity of NMES is usually set at a level that causes the stimulated muscles to contract. This typically causes pain. Quantifying the instantaneous changes in muscle microcirculation and metabolism during NMES before muscle contraction occurs is crucial, because it enables the current intensity to be optimally tuned, thereby reducing the NMES‐induced muscle pain and fatigue. We applied near‐infrared spectroscopy (NIRS) to measure instantaneous tissue oxygenation and deoxygenation changes in 43 healthy young adults during NMES at 10, 15, 20, 25, 30, and 35 mA. Having been stabilized at the NIRS signal baseline, the tissue oxygenation and total hemoglobin concentration increased immediately after stimulation in a dose‐dependent manner (P < 0.05) until stimulation was stopped at the level causing muscle contraction without pain. Tissue deoxygenation appeared relatively unchanged during NMES. We conclude that NIRS can be used to determine the optimal NMES current intensity by monitoring oxygenation changes.   相似文献

Menisci are very important fibrocartilaginous tissue, which maintain biomechanical functions and physiological stabilization of knee joint. Meniscectomy is known as a surgery to recover partial functions from acute meniscus tears. However, the late consequences of total or partial meniscectomy include signs of osteoarthritis and even ligament instability. Infrared attenuated total reflection (IR‐ATR) spectroscopy is a very useful technique, which can reveal molecular characteristics via the analysis of vibrational bands. The present study has employed IR‐ATR spectroscopy to investigate sheep menisci samples after meniscectomy in a label‐free fashion. Several differences of peak absorbance change and peak shift were observed between the native healthy samples and the meniscectomy samples in distinct IR wavenumber regions, such as amide I band, amide II band, C‐H bending band as well as the sugar band region. Combining the results from the collagen protein IR spectra, it can be speculated that six months after meniscectomy collagen fibrils on the incision lose its ordered arrangement and a decrease in the triple helical structure of collagen fibril is observed. In addition, the collagen fibrils and proteoglycan content might also be slight varied after meniscectomy.   相似文献

Synapses are the basic structural and functional units for information processing and storage in the brain. Their diverse properties and functions ultimately underlie the complexity of human behavior. Proper development and maintenance of synapses are essential for normal functioning of the nervous system. Disruption in synaptogenesis and the consequent alteration in synaptic function have been strongly implicated to cause neurodevelopmental disorders such as autism spectrum disorders (ASDs) and schizophrenia (SCZ). The introduction of human‐induced pluripotent stem cells (hiPSCs) provides a new path to elucidate disease mechanisms and potential therapies. In this review, we will discuss the advantages and limitations of using hiPSC‐derived neurons to study synaptic disorders. Many mutations in genes encoding for proteins that regulate synaptogenesis have been identified in patients with ASDs and SCZ. We use Methyl‐CpG binding protein 2 (MECP2), SH3 and multiple ankyrin repeat domains 3 (SHANK3) and Disrupted in schizophrenia 1 (DISC1) as examples to illustrate the promise of using hiPSCs as cellular models to elucidate the mechanisms underlying disease‐related synaptopathy.  相似文献

Objective: Globally, cardiovascular diseases (CVDs) are one of the most leading causes of death. In medical screening and diagnostic procedures of CVDs, electrocardiogram (ECG) signals are widely used. Early detection of CVDs requires acquisition of longer ECG signals. It has triggered the development of personal healthcare systems which can be used by cardio-patients to manage the disease. These healthcare systems continuously record, store, and transmit the ECG data via wired/wireless communication channels. There are many issues with these systems such as data storage limitation, bandwidth limitation and limited battery life. Involvement of ECG data compression techniques can resolve all these issues.Method: In the past, numerous ECG data compression techniques have been proposed. This paper presents a methodological review of different ECG data compression techniques based on their experimental performance on ECG records of the Massachusetts Institute of Technology-Beth Israel Hospital (MIT-BIH) arrhythmia database.Results: It is observed that experimental performance of different compression techniques depends on several parameters. The existing compression techniques are validated using different distortion measures.Conclusion: This study elaborates advantages and disadvantages of different ECG data compression techniques. It also includes different validation methods of ECG compression techniques. Although compression techniques have been developed very widely but the validation of compression methods is still a prospective research area to accomplish an efficient and reliable performance.  相似文献

BackgroundFrequency band optimization improves the performance of common spatial pattern (CSP) in motor imagery (MI) tasks classification because MI-related electroencephalograms (EEGs) are highly frequency specific. Many variants of CSP algorithm divided the EEG into various sub bands and then applied CSP. However, the feature dimension of MI-EEG data increases with addition of frequency sub bands and requires efficient feature selection algorithms. The performance of CSP also depends on filtering techniques.MethodIn this study, we designed a dual tree complex wavelet transform based filter bank to filter the EEG into sub bands, instead of traditional filtering methods, which improved the spatial feature extraction efficiency. Further, after filtering EEG into different sub bands, we extracted spatial features from each sub band using CSP and optimized them by a proposed supervised learning framework based on neighbourhood component analysis (NCA). Subsequently, a support vector machine (SVM) is trained to perform classification.ResultsAn experimental study, conducted on two datasets (BCI Competition IV (Dataset 2b), and BCI competition III (Dataset IIIa)), validated the MI classification effectiveness of the proposed method in comparison with standard algorithms such as CSP, Filter bank CSP (CSP), and Discriminative FBCSP (DFBCSP). The average classification accuracy obtained by the proposed method for BCI Competition IV (Dataset 2b), and BCI Competition III (Dataset IIIa) are 84.02 ± 12.2 and 89.1 ± 7.50, respectively and found significant than that achieved by standard methods.ConclusionAchieved superior results suggest that the proposed algorithm can improve the performance of MI-based Brain-computer interface devices.  相似文献

It has been shown that the propensity of a protein to form amyloid-like fibrils can be predicted with high accuracy from the knowledge of its amino acid sequence. It has also been suggested, however, that some regions of the sequences are more important than others in determining the aggregation process. Here, we have addressed this issue by constructing a set of “sequence scrambled” variants of the first 29 residues of horse heart apomyoglobin (apoMb1-29), in which the sequence was modified while maintaining the same amino acid composition. The clustering of the most amyloidogenic residues in one region of the sequence was found to cause a marked increase of the elongation rate (kagg) and a remarkable shortening of the lag phase (tlag) of the fibril growth, as determined by far-UV circular dichroism and thioflavin T fluorescence. We also show that taking explicitly into consideration the presence of aggregation-promoting regions in the predictive methods results in a quantitative agreement between the theoretical and observed kagg and tlag values of the apoMb1-29 variants. These results, together with a comparison between homologous segments from the family of globins, indicate the existence of a negative selection against the clustering of highly amyloidogenic residues in one or few regions of polypeptide sequences.  相似文献

Lipase from Burkholderia cepacia (lipase BC) and lipase B from Candida antarctica (CALB) show an increase of the transesterification activity in toluene (up to 2.4- and 1.7-fold, respectively), when lyophilized with 18-crown-6. Nevertheless, the increase was observed only for low (less than 100) 18-crown-6/lipase molar ratio, while at higher ratios, the activity decreased for both enzymes to values lower than those obtained in the absence of the additive. In 1,4-dioxane, the activation is lower for lipase BC (1.7-fold) and for CALB (1.5-fold). Concerning enantioselectivity, tested in the kinetic resolution of 6-methyl-5-hepten-2-ol, only in the case of CALB, an effect of the additive (the E value varied from about 120 to 280) was observed. In water, 4% (w/w) of 18-crown-6 caused a loss of activity in the hydrolysis of p-nitrophenyl laurate of about 88 and 99.75%, compared to that observed in the absence of the crown ether for CALB and lipase BC, respectively. These data and the conformational analysis of both lipases, carried out by FT/IR spectroscopy indicate that the enzyme inactivation in water and in organic solvents at 18-crown-6/lipase molar ratios, higher than 100 might be due to conformational changes caused by the additive. Instead, at molar ratios lower than 100, 18-crown-6 might increase the activity - particularly, in toluene - thanks to the fact that in its presence, the enzyme has an hydrogen bonds pattern, more similar to that in water. This suggests that the additive would be able to provide the enzyme with more water.  相似文献

Numerous fungal genomes encode homologs of bacteriorhodopsin (BR), but only two fungal rhodopsins were overexpressed and characterized spectroscopically. Neurospora rhodopsin (NR) is a slow-cycling sensory rhodopsin-like protein, while Leptosphaeria rhodopsin (LR) is a BR-like proton pump. Recently, we found that a conservative replacement of the cytoplasmic proton donor Asp150 by Glu converts LR into an NR-like protein. In this work, we search for structural reasons for the dramatic differences in their photochemistry by mutating the hydrogen-bonding partner of Asp150 (Thr87) and three additional residues (Thr233, Asp248, and Gly271) selected by comparison of the primary structures of NR and LR. We conclude that while these residues may contribute to the differences between LR and NR, they are not crucial for the optimization of the Schiff base reprotonation by Asp150, and that the dramatic effect of the D150E mutation is not a simple result of the introduction of a bulkier glutamate sidechain.  相似文献

Raman spectra provide wealthy but complex information about the chemical constituents of biological samples. Digital processing techniques are usually needed to extract the spectra of chemical constituents and their associated concentration profiles. However, spectral signatures may admit transformations from those recorded on pure constituents and these techniques require a priori knowledge of spectra to be estimated. We propose in this study to analyse paraffin-embedded skin biopsies of malignant and benign tumors dedicated to oncology researches by Raman spectroscopy and advanced signal processing methods. We show that the commonly used principal component analysis (PCA) does not give physically interpretable estimators of spectra and associated concentration profiles. Based on a linear model and taking into account the statistical properties of spectra, independent component analysis (ICA) is used to better estimate the spectra of chemical constituents. The estimators of associated concentration profiles are no longer orthogonal and have only positive values, contrary to PCA. ICA allows to model the paraffin by three Raman spectra and provides good estimators of underlying spectra of the human skin, which is of great interest in oncology since the retrieval of spectral features of different types of skin tumors is sufficient for their discrimination.  相似文献

The plant life cycle form and its spectrum features of different plants in the artificial community of Pinus svlvestris var. mongolica at the botanical garden of Northeast Forestry University were classified by PCA method. For canopy layer plants, the vegetative growth (V) accounted for 46%, the sexual growth (S) accounted for 35%, and the clonal growth (C) accounted for the rest 19%. For succession layer plants, V accounted for more than 50%, and C accounted for a little higher than S did. For herbaceous layer plants, V accounted for nearly 47%, which was only 4% higher than that of S, and C only accounted for 11%. These findings indicate that the plant life cycle forms of canopy layer plants and herbaceous layer plants were VS transition form with V form as the main form, while the succession layer plants were V form. At the viewpoint of artificial community, the life cycle form can be described as V0.49S0.33C0.18, a typical VS transition form, indicating that most of the species studied in this community are in a healthy status with the V form. Moreover, the relatively high (33%) percentage in the S form indicates that the community also tends to sexual growth for offspring, although it will be steady within a period of time for the vegetative growth of bodies.  相似文献
