Studies examining intraspecific variation in plant species with widespread distributions and disjunct populations have mainly concentrated on temperate species. Here, we determined the genetic structure of a broadly distributed wetland tropical tree, Pterocarpus officinalis (Jacq.), from eight Neotropical populations using amplified length fragment polymorphisms (AFLP). AFLPs proved highly variable with almost half (48%) of the genetic variation at these loci occurring among individuals within populations. Nonetheless, there was a strong geographical pattern in the distribution of AFLP variation within P. officinalis. Caribbean and continental populations fell into two well-defined genetic clusters supported by the presence of a number of unique AFLP bands. Within these two regions, there were also strong genetic differences among populations, caused mainly by frequency differences in AFLP bands, making it difficult to determine the evolutionary relationships among populations. In addition, our analysis of P. officinalis revealed striking differences in the levels of AFLP variation among the eight populations sampled. In general, Caribbean populations had lower genetic diversity than continental populations. Moreover, there was a clear loss in AFLP diversity with distance from the continent among Caribbean populations. The overall genetic pattern within P. officinalis suggests that past colonization history, coupled with genetic drift within local populations, rather than contemporary gene flow are the major forces shaping variation within this species.  相似文献

Mitochondrial DNA (mtDNA) D-loop sequences (381 bp) from 100 thoroughbreds in 19 of the most common matrilineal female families were used to reconstruct a founder female population for the thoroughbred ( approximately 1650-1750 AD). Seventeen haplotypes were found to have contributed to the 19 female lineages. In order to place the reconstructed founder population in wider historical context, we examined, using both single strand conformation polymorphism and direct sequence analysis, variation in a 343 bp mtDNA fragment in that population and 13 other horse populations of disparate provenance. Interpopulation diversity analyses revealed no significant difference in variation between the thoroughbred founder population and the 13 other diverse horse populations and suggested a non-random partitioning of diversity among geographically diverse horse populations. Within thoroughbreds, almost half of the female families, which are each considered from pedigrees to have matrilines converging to one of >30 historically recognized female ancestors, contained sequences which were inconsistent with common descent. Examination of the anomalies in the context of pedigree records suggests the majority might be best explained as confusion of mares at the foundation stages, although some could have some bearing on more recent (19th century - 1980) lineages. We have used this combination of molecular and historical information to identify some of the founder dams and to make new interpretations about the early history of the thoroughbred.  相似文献

The east coast of Greenland is inhabited in only two places: in Angmagssalik live the direct descendants of the 413 Eskimo discovered in 1884; 1000 km further north, a small isolated settlement, Scoresbysund, was founded in 1925 by 70 Eskimo from Angmagssalik.Several biological features were studied both on the parent population and its descendants and the founders themselves and their progeny. Moreover, detailed and periodically maintained genealogical records from the time of their discovery provide exceptional complete information on these two groups and make them particularly favourable for the study of certain anthropological and genetic problems.Regarding various hereditary anthropological characteristics (blood groups, finger patterns, anthropometric measurements), the isolate shows certain particularities compared to the parent population. There is a much greater ressemblance between the settlement's founders and their present descendants than between these two groups and the parent population and its descendants. A set of converging elements indicate that this isolate represents a good example of the founder effect for various anthropological characteristics.  相似文献

Pond-dwelling copepods have colonized habitats throughout North America after glaciers have receded. Most species are passively transported via resting eggs into new habitats. Colonists originating in a glacial refugium could lose some of the ancestral genetic diversity when they establish new populations and the attenuation may be substantial in populations far removed from the refugium due to multiple founder events. Genetic variation was measured in Heterocope septentrionalis from 27 populations at arctic sites near potential refugia and those more recently deglaciated to determine the effects of postglacial dispersal on patterns of genetic relatedness and diversity. Some populations were more distant, genetically, from others within the same site than those from other more distant sites. Eleven polymorphic enzyme loci were significantly more variable (F [1,294 df] = 5.94, P < 0.025) among individuals from populations near the Alaskan refuge than those at the eastern limit of their distribution. Because populations are typically extremely large and stable this loss of genetic diversity is attributed primarily to repeated founder events during colonization. This result suggests profound genetic changes may occur on a continental scale in passively dispersed copepods due to founder events alone. Their potential for divergence and speciation is greater than currently recognized.  相似文献

The distribution of four variants for flower colour ofCrocus scepusiensis in the northern part of the Western Carpathians is described. The frequency of white stigmata morphs declines from east to west. In the center of the area stigmata colour morphs show strikingly patchy distribution. White perianth morphs usually occur at low frequencies and their distribution with minor exceptions is restricted to the central part of the area, where patchy distribution of the morphs is a rule. These distribution patterns suggest that the founder effect has played a major role in determining the genetic composition of individual populations. The cline for stigmata colour may also be explained by the dynamics of population expansion. No influence of selection can be demonstrated, but the association between perianth and stigmata colour, and the excessively low frequency of white perianth morphs may imply that the polymorphisms are not selectively neutral.  相似文献

Within Scandinavia, Carex capitata s.l, is usually divided into two taxa: the lowland to lowalpine C. capitata s.s., and the alpine C. arctogena. The systematic status of these taxa is uncertain, reflected by their taxonomic treatment in different floras. In this study, allozymes were used to assess the degree of genetic differentiation between and within taxa. Ten populations of Carex capitata s.s. and eight of C. arctogena, from Norway and Sweden, were included in the study. Sixteen loci in eleven different enzyme systems were analysed: five were variable within or between taxa, three could be interpreted in all plants. In all, 20 different alleles were found. Four of these alleles were confined to C. capitata s.s., and two to C. arctogena. In C. capitata s.s., three loci (18%) were polymorphic, while all loci were monomorphic in C. arctogena. The number of alleles per polymorphic locus was 2.2 in C. capitata s.l. The taxa had completely different alleles at two loci {Pgi-2, Pgm-1). Nei's genetic identity was 0.86 between taxa, and ranged between 0.95 and 1 in pairwise comparisons of populations of Carex capitata s.s., while it was 1 in all comparisons between populations of C. arctogena. The distinction of C. capitata s.s. and C. arctogena as separate species agrees well with the amount of differentiation found between other congeneric species. It is suggested that the low genetic variation observed in both taxa is due to ancient founder events. The difference in genetic variability between taxa may be due to differences in immigration history: whereas C. capitata s.s. may have colonized Scandinavia from several close refugia after the last glaciation, C. arctogena may have reached Scandinavia after long-distance dispersal of a limited number of propagules.  相似文献

The central coastal region of Tunisia harbours two chromosomal races of the house mouse (2 n  = 22, 22Rb; 2 n  = 40, 40Std), which are genetically differentiated and show a high level of reproductive isolation. This study presents an allozyme analysis of house mice from the inland city of Kairouan 75 km from the coastal region. Results showed that the 22Rb and 40Std mice from Kairouan shared the same high degree of variability, and were not genetically differentiated. This contrasts with the genetic data registered for the two chromosomal races from the coastal towns, in which a particularly low genic diversity was observed in the 22Rb populations. As the two races in Kairouan show the same low hybridization rates as the populations in the coastal region, these results argue in favour of a local differentiation of the Rb race in Tunisia, most likely originating in Kairouan. The data further suggest that the decrease in variability observed in the coastal 22Rb populations is related to a founder effect or a bottleneck following the introduction of mice from Kairouan into one of these localities, prior to their dispersion throughout the coastal region.  © 2005 The Linnean Society of London, Biological Journal of the Linnean Society , 2005, 84 , 515–521.  相似文献

Aim  Island populations of small mammals are often characterized by a larger body size compared with neighbouring mainland or continental populations of the same species. A number of reasons have been put forward to explain this phenomenon. The aim of this study was to test which of these hypotheses can best explain the increase of body size in common shrews ( Sorex araneus ) on islands.
Location  The fieldwork for this study was carried out on the islands of the Inner Hebrides, Clyde Islands and the west coast of Scotland.
Methods  This study compared body sizes of common shrews from mainland and island sites on the west coast of Scotland, based on measurements of hind foot lengths. On 10 of the 13 islands sampled, common shrews were significantly larger than on the mainland. Body size did not vary significantly among mainland populations. We used the directional contrasts method to test the relative contributions of possible factors explaining the large body size observed in the island populations.
Results  We found that body size of common shrews on islands was positively related to distance from mainland, negatively related to average annual temperature, negatively related to island size, and may also be influenced by the presence or absence of pygmy shrews ( Sorex minutus ) on the island.
Main conclusions  Our results suggest a role for founder events, Bergmann's rule and K -selection in determining body size of common shrews on islands.  相似文献

Aim The origins of the Cuban biota are of long‐standing interest in biogeography, and the source of a small live oak (Quercus series Virentes) population on Cuba remains unresolved. Based on morphological evidence, previous authors have hypothesized a Florida origin from either Q. geminata or Q. virginiana or both; a Mexican origin from Q. oleoides; or a hybrid origin from both sources. We use molecular data and taxonomically informative leaf morphology to identify the source species and timing of colonization. Location Cuba, Central America, Mexico and the south‐eastern United States. Methods  We collected representative samples of Cuban oaks and each putative source species and genotyped each sample at 12 nuclear microsatellites and two chloroplast DNA sequences. We estimated population structure using a Bayesian clustering analysis and F‐statistics, pairwise migration rates among taxa, and divergence time using an isolation‐with‐migration model. We measured seven leaf traits and conducted an analysis of similarity (ANOSIM) to determine which putative source species was most similar to Cuban oaks. Results Cuban oak contains one chloroplast DNA haplotype, which is common in southern Florida. Bayesian clustering analysis of microsatellites revealed that the Cuban oak forms a distinct and pure population cluster, and F‐statistics showed that Cuban oaks are differentiated least from Q. virginiana and most from Q. geminata. Migration rates were highest out of Cuba to Q. oleoides. Molecular diversity, the ratio of allelic richness to allele size range, and effective population size of the Cuban oak were relatively low, suggesting a founder effect. Divergence time estimates fell entirely within the Pleistocene (628–6 ka), considering a range of mutation rates and generation times. Cuban oaks were morphologically most similar to Q. virginiana and least similar to Q. geminata. Main conclusions Molecular and morphological data support a Pleistocene dispersal of Q. virginiana from Florida to Cuba, followed by isolation and divergence, then limited dispersal and introgression from Cuba to Q. oleoides in Central America. Birds could have dispersed acorns to Cuba during a glacial period when sea levels were low. These results highlight the varied origin of the Cuban biota and the possible role of Pleistocene glaciations in the establishment of temperate taxa in the tropics.  相似文献

The worldwide destructive epidemic of the fungus Mycosphaerella fijiensis on banana started recently, spreading from South-East Asia. The founder effects detected in the global population structure of M. fijiensis reflected rare migration events among continents through movements of infected plant material. The main objective of this work was to infer gene flow and dispersal processes of M. fijiensis at the continental scale from population structure analysis in recently invaded regions. Samples of isolates were collected from banana plantations in 13 countries in Latin America and the Caribbean and in Africa. The isolates were analysed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and microsatellite molecular markers. The results indicate that a high level of genetic diversity was maintained at the plantation and the plant scales. The loci were at gametic equilibrium in most of the samples analysed, supporting the hypothesis of the existence of random-mating populations of M. fijiensis, even at the plant scale. A low level of gene diversity was observed in some populations from the Africa and Latin America-Caribbean regions. Nearly half the populations analysed showed a significant deviation from mutation-drift equilibrium with gene diversity excess. Finally, a high level of genetic differentiation was detected between populations from Africa (FST = 0.19) and from the Latin America-Caribbean region (FST = 0.30). These results show that founder effects accompanied the recent invasion of M. fijiensis in both regions, suggesting stochastic spread of the disease at the continental scale. This spread might be caused by either the limited dispersal of ascospores or by movements of infected plant material.  相似文献
