Secondary product accumulation by suspension cultures of Glycyrrhiza glabra has been studied. The cultures did not accumulate either glycyrrhizin of glycyrrhetic acid but did produce the isoflavonoid formononetin. The effect of initial pH, sucrose concentrations, medium composition, auxin and cytokinins on formononetin accumulation was studied. The isoflavone glucosides daidzin, genistin and ononin, the isoflavones daidzein and formononetin, and glyceollins I-III accumulated in soybean leaves inoculated with phytopathogenic bacteria. Treatment of leaves with sodium iodoacetate or yeast extract also led to isoflavonoid accumulation. Various other stress-inducing treatments were not effective. Bacterially-induced accumulation of isoflavone glucosides and the occurrence of ononin and formononetin in soybean are reported for the first time.  相似文献

In current study, a bioinformatic‐based network pharmacology was used to identify the osteosarcoma (OGS)‐pathological targets and formononetin (FN)‐treated targets before the main core predictive biotargets were screened. In addition, all core targets were selected through a number of bioinformatic databases, followed by identification of predominant biological processes and signalling pathways of FN anti‐OGS. Further, top three core targets of FN anti‐OGS were determined as oestrogen receptor 1 (ESR1), tumour protein p53 (TP53), receptor tyrosine‐protein kinase erbB‐2 (ERBB2) respectively. In clinical biochemical data, the plasma samples of OGS showed the increased trends of alkaline phosphatase, triglyceride, blood glucose, lactate dehydrogenase, high‐sensitive C‐reactive protein and some immune cell counts when referenced to medical criteria. In clinicopathological examination, histological OGS sections resulted in increased positive cell counts of neoplastic ESR1, TP53, ERBB2. To further validate these corn proteins in experimental study in vivo, FN‐treated tumour‐bearing nude mice showed intracellular reductions of ESR1, TP53, ERBB2 positive expressions, accompanied with visibly reduced tumour weights. Collectively, our bioinformatic and experimental findings disclosed main core targets, biological processes and signalling pathways of FN anti‐OGS. Interestingly, the top core targets were representatively validated following FN treatment in vivo. Therefore, we reasoned that these predictive targets might be the potential biomarkers for screening and treating osteosarcoma.  相似文献

Diabetic neuropathy is commonly observed complication in more than 50 % of type 2 diabetic patients. Histone deacetylases including SIRT1 have significant role to protect neuron from hyperglycemia induced damage. Formononetin (FMNT) is known for its effect to control hyperglycemia and also activate SIRT1. In present study, we evaluated effect of FMNT as SIRT1 activator in type 2 diabetic neuropathy. Type 2 diabetic neuropathy was induced in rats by modification of diet for 15 days using high fat diet and administration of streptozotocin (35 mg/kg/day, i. p.). FMNT treatment was initiated after confirmation of type 2 diabetes. Treatment was given for 16 weeks at 10, 20 and 40 mg/kg/day dose orally. FMNT treatment‐controlled hypoglycemia and reduced insulin resistance significantly in diabetic animals. FMNT treatment reduced oxidative stress in sciatic nerve tissue. FMNT treatment also reduced thermal hyperalgesia and mechanical allodynia significantly. It improved conduction velocity in nerve and unregulated SIRT1 and NGF expression in sciatic nerve tissue. Results of present study indicate that continuous administration of FMNT protected diabetic animals from hyperglycemia induced neuronal damage by controlling hyperglycemia and increasing SIRT1 and NGF expression in nerve tissue. Thus, FMNT can be an effective candidate for treatment of type 2 diabetic neuropathy.  相似文献

A steadily growing culture of genetically transformed roots of a valuable medicinal Altaic plant Hedysarum theinum Krasnob. was established using a Ri-plasmid (pRi) T-DNA of A4 wild strain of Agrobacterium rhizogenes. The composition of secondary substances accumulated in the in vitro roots and the roots of H. theinum intact seedlings was investigated. Isoflavonoids were found to represent by their main low-molecular metabolites. Preparative HPLC made it possible to isolate four substances from the methanolic extract of H. theinum cultured roots. Using 1H- and 13C-NMR-spectrometry, these substances were identified as formononetin, ononin (formononetin glycoside), malonyl ononin, and texasin glucoside. The qualitative composition of secondary metabolites of the genetically transformed roots and the roots of H. theinum seedlings was essentially the same, except that malonyl ononin was not found in the latter. The technique of producing artificial seeds on the basis of H. theinum roots cultured in vitro was tested, and the possibility of their use as a rhizogenic inoculum was substantiated. The culture of H. theinum roots is considered as a potential source of ecologically pure raw material for medicinal preparations, and the artificial seeds with root inoculum are a promising vehicle for propagation and conservation of this valuable plant.  相似文献

Phosphorus effect on phosphatase activity in endomycorrhizal maize   总被引:3,自引:0,他引:3  
Success of a mycorrhizal symbiosis is influenced by the availability of phosphorus (P) in the soil. Maize ( Zea mays L. cv. Great Lakes 586) plants were grown under five different levels of soil P, either in the presence or absence of formononetin or the vesicular‐arbuscular mycorrhizal (VAM) fungus Glomus intraradices Schenck and Smith. We detected physiological differences in mycorrhizal roots very early in the development of symbiosis, before the onset of nutrient‐dependent responses. Under low P levels, VAM roots accumulated a greater shoot dry weight (13%), root P concentration (15%) and protein concentration (30%) than non-VAM roots, although root growth was not statistically significantly different. At higher P levels, mycorrhizal roots weighed less than non-VAM roots (10%) without a concomitant host alteration of growth or root P concentration. Mycorrhizal colonization decreased as soil P increased. Formononetin-treatment enhanced colonization of the root by G. intraradices and partially overcame inhibition of VAM colonization by high soil P concentrations. This is the first report that formononetin improves root colonization under high levels of soil P. Acid phosphatase (ACP) and alkaline phosphatase (ALP) activities were closely related to the level of fungal colonization in corn roots. ACP activity in corn roots responded more to soil P availability than did ALP activity (38% more). These results suggest that ACP was involved in the increased uptake of P from the soil, while ALP may be linked to active phosphate assimilation or transport in mycorrhizal roots. Thus, soil P directly affected a number of enzymes essential in host-endophyte interplay, while formononetin enhanced fungal colonization.  相似文献

Isoflavones form a group of plant compounds that occur mainly in legumes, soy being the most important source in human diet. The high levels of isoflavones in the diet have been associated with a lowered risk for hormone-dependent diseases, including breast and prostate cancers, osteoporosis and cardiovascular disease. The metabolism of isoflavones in humans has been studied to a certain extent, but detailed studies are lacking. This paper reviews the current knowledge on metabolism of isoflavones and presents some preliminary results of a comprehensive soy feeding study, in which the phase I metabolites of soy isoflavones, daidzein, genistein and glycitein, were identified by GC-MS. 怀槐(Maackia amurensis Rupr.et Maxim.)广泛分布于中国东北三省及内蒙古等地,尤以黑龙江省为多.怀槐作为民间药,其茎、枝可治疗风湿性关节炎; 叶、枝皮可治疗肿瘤; 内皮可治疗深度伤口及久不愈合的溃疡.另外,怀槐树皮在民间还作为祛风湿、消炎、镇痛、健胃、止血等药广泛应用,疗效甚佳.从怀槐的根、枝、叶、果实和心材中分别分得异黄酮、生物碱、NC442类化合物,据报道,其中的异黄酮类化合物表现出对心血管系统的复合作用和明显的抗肿瘤活性[1].作者在对怀槐进行系统的化学成分研究过程中,分离鉴定了9个异黄酮类化合物[2,3],并进行了抑制肿瘤细胞增殖作用研究[4],其中,染料木素(genistein)对胃腺癌细胞(BGC)增殖有一定程度的抑制作用,芒柄花黄素(formononetin)则对白血病细胞(HL-60)增殖有一定的抑制作用.染料木素和芒柄花黄素作为马鞍树属(Maackia Rupr.)植物中普遍存在的2个异黄酮类化合物,其活性研究日益受到重视.采用HPLC法测定染料木素等异黄酮类化合物含量在大豆等其他植物中曾有文献报道[5].为了尽早开发怀槐资源,同时为其资源的综合利用提供科学依据,本文采用HPLC法对怀槐不同部位的染料木素和芒柄花黄素2个有效成分进行了含量测定.  相似文献
