DNA damage bypass pathways promote the replication of damaged DNA when replication forks stall at sites of DNA damage. Template switching is a DNA damage bypass pathway in which fork-reversal helicases convert stalled replication forks into four-way DNA junctions called chicken foot intermediates, which are subsequently extended by replicative DNA polymerases. In yeast, fork-reversal is carried out by the Rad5 helicase using an unknown mechanism. To better understand the mechanism of Rad5 and its specificity for different fork DNA substrates, we used a FRET-based assay to observe fork reversal in real time. We examined the ability of Rad5 to bind and catalyze the reversal of various fork DNA substrates in the presence of short gaps in the leading or lagging strand as well as in the presence or absence of RPA and RNA primers in the lagging strand. We found that Rad5 preferentially reverses fork DNA substrates with short gaps (10 to 30 nt.) in the leading strand. Thus, Rad5 preferentially reverses fork DNA substrates that form chicken foot intermediates with 5′ overhangs that can be extended by replicative DNA polymerases during the subsequent steps of template switching.  相似文献

Truss analysis and length measurements were made on 168 striped red mullet Mullus surmuletus. Multivariate statistical analyses with principal component analysis and partial redundancy analysis (pRDA) were used on these measurements to evaluate the influence of maturity, sex and geographical area distribution on body shape. Truss measurements were important to quantify and discriminate changing body shape, presumably due to changing environmental conditions. Sexual dimorphism was not observed and juveniles could be distinguished from adults based on their body shape. More importantly, M. surmuletus occurring in different geographical areas could be differentiated using this method. Based on pRDA, a significant difference of head morphological dimensions was observed between populations occurring in the eastern English Channel and those occurring in the Bay of Biscay, suggesting that fish from these areas could represent two subpopulations.  相似文献

Lizards of the family Eublepharidae exhibit interspecific diversity in body size, sexual size dimorphism (SSD), head size dimorphism (HSD), occurrence of male combat, and presence of male precloacal pores. Hence, they offer an opportunity for testing hypotheses for the evolution and maintenance of sexual dimorphism. Historical analysis of male agonistic behaviour indicates that territoriality is ancestral in eublepharid geckos. Within Eublepharidae, male combat disappeared twice. In keeping with predictions from sexual selection theory, both events were associated with parallel loss of male-biased HSD and ventral scent glands. Eublepharids therefore provide new evidence that male-biased dimorphic heads are weapons used in aggressive encounters and that the ventral glands probably function in territory marking rather than in intersexual communication. Male-biased SSD is a plesiomorphic characteristic and was affected by at least three inversions. Shifts in SSD and male combat were not historically correlated. Therefore, other factors than male rivalry appear responsible for SSD inversions. Eublepharids demonstrate the full scope of Rensch's rule (small species tend to be female-larger, larger species male-larger). The serum levels of choline decreased by approximately 50% in patients having a surgery under general as well as epidural anesthesia. The decrease is lasts for two days after surgery. Intravenous administration of succinylcholine, either by a single bolus injection or by a slow continuous infusion, increased the serum choline levels several folds during surgery. In these patients, a significant decrease in the serum choline levels was observed one and two days after surgery. In 16 pregnant women at the term, serum choline levels were higher than the value observed in 19 non-pregnant women. The serum choline levels decreased by about 40% or 60% after having a child-birth either by vaginal delivery or caesarean section, respectively. Serum choline levels in blood obtained from 9 patients with traumatic head injury were significantly lower than the observed levels in blood samples obtained from healthy volunteers. These observations show that serum choline levels increase during pregnancy and decrease during stressful situations in humans.  相似文献
