Phylogenetic relationships were inferred for six Central American and Mexican pine species by analysing RAPD marker differences among pooled DNA samples. This population level pooling strategy discounts low-frequency allelic variation within taxa, thus obtaining a 'cumulative genotype' to compare among taxa. We used the morphologically based taxonomy of pines as the basis for inference concerning molecular marker divergence. Only RAPD polymorphisms that were repeatable and intensely amplified were used. The resulting data set was found to have strong hierarchical structure. Phylogenetic analyses were carried out using both neighbour-joining and maximum parsimony. The phylogenetic relationships indicated by analysis of the pine RAPD data provide new insights on pine taxonomy. The phylogenetic analyses of the RAPD marker data placed Pinus tecunumanii and P. patula clearly into two different subgroups, strongly supporting the classification of P. tecunumanii as a distinct species, and raise a new set of issues concerning the distinctions between Pinus tecunumanii and P. caribaea . Phylogenetically informative RAPD markers will be useful tools to address ex-situ conservation issues that involve taxonomic identification and species admixture.  相似文献

Creating, restoring, and sustaining forests in urban areas are complicated by habitat fragmentation, invasive species, and degraded soils. Although there is some research on the outcomes of urban reforestation plantings during the first 5 years, there is little research on longer term outcomes. Here, we compare the successional trajectories of restored and unrestored forest sites 20 years after initiating restoration. The sites are located within the Rodman's Neck area of Pelham Bay Park, in the northeast corner of the Bronx in New York City (NYC), U.S.A. Compared with unrestored sites, we saw improvements in species diversity, greater forest structure complexity, and evidence of the regeneration and retention of native tree species in restored sites. In addition, we found differences in restoration outcomes depending on the level of intervention: clearing exotic shrubs and vines and planting native trees and shrubs improved tree diversity and canopy closure to a greater extent than clearing exotics alone, and the mechanical removal of invasive plants after the native plantings further improved some measures of restoration, such as tree species diversity and native tree regeneration. The results of this study suggest that the goal of a sustainable forest ecosystem dominated by native trees and other plant species may not be achievable without continued human intervention on site. In addition, these results indicate that the restoration approach adopted by NYC's reforestation practitioners is moving the site toward a more desirable vegetative community dominated by native species.  相似文献

BackgroundDespite availability of effective treatment, tuberculosis (TB) remains an important cause of morbidity and mortality globally, with low- and middle-income countries most affected. In many such settings, including Malawi, the high burden of disease and severe shortage of skilled healthcare workers has led to task-shifting of outpatient TB care to lay health workers (LHWs). LHWs improve access to healthcare and some outcomes, including TB completion rates, but lack of training and supervision limit their impact. The goals of this study are to improve TB care provided by LHWs in Malawi by refining, implementing, and evaluating a knowledge translation strategy designed to address a recognized gap in LHWs’ TB and job-specific knowledge and, through this, to improve patient outcomes.Methods/designWe are employing a mixed-methods design that includes a pragmatic cluster randomized controlled trial and a process evaluation using qualitative methods. Trial participants will include all health centers providing TB care in four districts in the South East Zone of Malawi. The intervention employs educational outreach, a point-of-care reminder tool, and a peer support network. The primary outcome is proportion of treatment successes, defined as the total of TB patients cured or completing treatment, with outcomes taken from Ministry of Health treatment records. With an alpha of 0.05, power of 0.80, a baseline treatment success of 0.80, intraclass correlation coefficient of 0.1 based on our pilot study, and an estimated 100 clusters (health centers providing TB care), a minimum of 6 patients per cluster is required to detect a clinically significant 0.10 increase in the proportion of treatment successes. Our process evaluation will include interviews with LHWs and patients, and a document analysis of LHW training logs, quarterly peer trainer meetings, and mentorship meeting notes. An estimated 10–15 LHWs and 10–15 patients will be required to reach saturation in each of 2 planned interview periods, for a total of 40–60 interview participants.DiscussionThis study will directly inform the efforts of knowledge users within TB care and, through extension of the approach, other areas of care provided by LHWs in Malawi and other low- and middle-income countries.

Summary Instantaneous oxygen consumption, muscle potential frequency, thoracic and ambient temperature were simultaneously measured during heating in individual workers and drones of honey bees. Relationships between these parameters and effects of thoracic temperature on power input and temperature elevation were studied. Oxygen consumption increased above basal levels only when flight muscles became active. Increasing muscle potential frequencies correlated with elevated oxygen consumption and raised thoracic temperature. The difference between thoracic and ambient temperature and oxygen consumption were linearly related. Oxygen consumption per muscle potential (l O2 · g –1 thorax · MP–1) was two-fold higher in drones than in workers. However, oxygen consumption for heating the thorax (l O2 · g –1 thorax · (Tth-Ta) · °C–1) was nearly the same in workers and drones. Thoracic temperature affected the amount of oxygen consumed per muscle potential (R10=1.5). Achieved temperature elevation per 100 MP was more temperature sensitive in drones (R10=6–10) than in workers (R10=3.6). Q10 values for oxygen consumption were 3 in workers and 4.5–6 in drones. Muscle potential frequency decreased with a Q10=1.8 in workers and 2.7 in drones. Heating behaviour of workers and drones was different. Drones generated heat less continuously than workers, and showed greater interindividual variability in predilection to heat.   • 1 Foraging routes of worker and queen bumble bees (Bombus kirbyellus Curtis) collecting nectar from flowers of the alpine sky pilot, Polemonium viscosum Nutt., were followed and the corolla tube length, corolla diameter, floral scent, and number of flowers on plants visited or bypassed by bees were monitored. Additionally, the number and proportion of flowers visited per inflorescence and distance flown from each to the next were recorded. Queens and workers differed significantly in choice of flowers. However, intra-inflorescence visitation rates and departure distances were similar between castes. Castes differed in the extent to which visitation reflected patch quality versus individual floral traits.
  • 2 Both queens and workers failed to visit skunky-flowered plants more often than they failed to visit sweet-flowered ones, and preferred large over small inflorescences. However, queens visited large-flowered plants more often than small-flowered ones, while workers preferred flowers with shorter corolla tubes, regardless of their diameter. Although a number of studies have documented caste specialization on alternate species of host plants, ours is one of the first to show that morphological preferences promote comparable foraging differences between castes on monospecific plant resources.
  • 3 Queens, once on a plant, responded to floral traits by probing more flowers on large inflorescences, as well as on those with broader floral form. Workers did not alter intra-inflorescence visitation rate in response to floral traits.
  • 4 For workers, no significant relationship was demonstrated between the likelihood of passing by a plant and the number of flowers probed on the previous inflorescence visited. Thus, workers appeared to accept or reject each plant of P. viscosum independently. However, queens passed by fewer plants when leaving rich inflorescences than poor ones. These results suggest that workers use only individual plant acceptability in choosing which plants to visit, whereas queens base plant choice on patch and individual attributes. Such differences between castes in foraging rules when exploiting the same floral resource have received little attention, and provide insights into the heterogeneity of harvestable reward distributions from the perspective of the forager population.

Question Are the patterns of alien conifer (Pinaceae, Cupressaceae) invasions different between continents, and how is invasion success influenced by commercial forestry practices? Location Temperate and subtropical countries and regions (n = 60) from five continents spanning both hemispheres. Methods We used generalized linear mixed models to test how continent identity, region area and use in commercial forestry affect probabilities of Pinaceae and Cupressaceae species to escape following introduction and cumulative logit regression models to assess how these predictors affect the likelihood that a species becomes naturalized or invasive. Results Sixty Pinaceae of a global total of 232 and 26 Cupressaceae of a total of 142 species have escaped from cultivation across the study regions examined. Average numbers of both alien Pinaceae and Cupressaceae species per region were highest in Oceania, followed by Africa. Moreover, the probability of alien Cupressaceae and Pinaceae becoming naturalized or invasive was particularly high in these two continents. For both families, species used in commercial forestry have a significantly higher probability of escape than those which are only introduced for ornamental or other purposes. In the case of Pinaceae, forestry species also become naturalized or invasive more frequently than non‐forestry species, while no such effect was detectable for Cupressaceae. Conclusions We found that non‐native conifers are more likely to escape from cultivation, naturalize and turn into invasive weeds on the continents of the Southern Hemisphere. In addition to this biogeographic signal, introduction effort strongly determines the behaviour of introduced Pinaceae, and less so, Cupressaceae. A clear conflict exists between the economic benefits of conifer forestry and the risks to the environment from invasions. Future expansion of commercial forestry should address spatial planning to ecosystems vulnerable to invasion and adopt comprehensive risk assessment procedures.  相似文献

To assess the validity of previously developed risk assessment schemes in the conditions of Central Europe, we tested (1) Australian weed risk assessment scheme (WRA; Pheloung et al . 1999); (2) WRA with additional analysis by Daehler et al . (2004); and (3) decision tree scheme of Reichard and Hamilton (1997) developed in North America, on a data set of 180 alien woody species commonly planted in the Czech Republic. This list included 17 invasive species, 9 naturalized but non-invasive, 31 casual aliens, and 123 species not reported to escape from cultivation. The WRA model with additional analysis provided best results, rejecting 100% of invasive species, accepting 83.8% of non-invasive, and recommending further 13.0% for additional analysis. Overall accuracy of the WRA model with additional analysis was 85.5%, higher than that of the basic WRA scheme (67.9%) and the Reichard–Hamilton model (61.6%). Only the Reichard–Hamilton scheme accepted some invaders. The probability that an accepted species will become an invader was zero for both WRA models and 3.2% for the Reichard–Hamilton model. The probability that a rejected species would have been an invader was 77.3% for both WRA models and 24.0% for the Reichard–Hamilton model. It is concluded that the WRA model, especially with additional analysis, appears to be a promising template for building a widely applicable system for screening out invasive plant introductions.  相似文献

Occupational exposure to cadmium has for long been associated with renal tubular cell dysfunction, osteomalacia with osteoporosis, hypercalciuria and renal stone formation. High environmental exposure in Japan resulting from a stable diet of cadmium contaminated rice caused itai-itai disease, fractures occurring mainly in elderly multiparous women, with a form of osteomalacia, osteoporosis and renal dysfunction. More recently a population based study in Europe, in the vicinity of zinc smelters has shown that low to moderate exposure to cadmium, with a mean urinary excretion of cadmium of the order of 1 microg/g creatinine has been associated with a decrease in bone density, an increased risk of bone fractures in women and of height loss in men. In a population-based study of residents near a cadmium smelter in China, forearm bone density was shown to decrease linearly with age and urinary cadmium in both sexes, suggesting a dose effect relationship between cadmium dose and bone mineral density. A marked increase in the prevalence of fractures was shown in the cadmium-polluted area in both sexes. Concentrations of cadmium in blood and urine were taken as exposure biomarkers, and beta2-microglobulin, retinol binding protein and albumin as biomarkers of effect. A marked dose response relationship between these indicators of exposure and effect was shown. Hypercalciuria, which may progress to osteoporosis, has been taken as a sensitive renal-tubular biomarker of a low level of cadmium exposure. Cadmium may also act directly on bone. Animal studies have shown cadmium to stimulate the formation and activity of osteoclasts, breaking down the collagen matrix in bone. Osteoporosis is the main cause of fracures in post-menopausal women, a common occurrence worldwide, giving rise to disability and a high cost to health services. The identification of cadmium, an environmental pollutant, as one causal factor is highly significant in helping to control the incidence of this complex condition.  相似文献

This study aimed to determine the bacterial species colonizing the nasal and oropharyngeal mucosa of fuel workers in Central Riyadh, Saudi Arabia on a microbiological and molecular level. Throat and nasal swab samples were obtained from 29 fuel station attendants in the period of time extending from March to May 2014 in Riyadh, Saudi Arabia. Microbiological identification techniques were utilized to identify the bacterial species isolated. Antibiotic sensitivity was assessed for each of the bacterial isolates. Molecular identification techniques based on PCR analysis of specific genomic sequences was conducted and was the basis on which phylogeny representation was done for 10 randomly selected samples of the isolates. Blood was drawn and a complete blood count was conducted to note the hematological indices for each of the study participants. Nineteen bacterial species were isolated from both the nasal cavity and the oropharynx including Streptococcus thoraltensis, alpha-hemolytic streptococci, Staphylococcus hominis, coagulase-negative staphylococci, Leuconostoc mesenteroides, Erysipelothrix rhusiopathiae and several others. We found 100% sensitivity of the isolates to ciprofloxacin, cefuroxime and gentamicin. Whereas cefotaxime and azithromycin posted sensitivities of 85.7% and 91.4%, respectively. Low sensitivities (<60% sensitivity) to the antibiotics ampicillin, erythromycin, clarithromycin and norfloxacin were observed. Ninety-seven percent similarity to the microbial bank species was noted when the isolates were compared to it. Most hematological indices recorded were within the normal range. In conclusion, exposure to toxic fumes and compounds within fuel products may be a contributing factor to bacterial colonization of the respiratory tract in fuel workers.  相似文献
