It is well established that somatotropin (GH) antagonizes insulin action in vivo and that supraphysiologic concentrations of GH frequently result in insulin resistance and glucose intolerance. However, the demonstration of an anti-insulin activity by GH in vitro has been difficult. This study, therefore, set out to determine whether cultures of 3T3-L1 adipocytes could be used to examine the anti-insulin activity of GH. The ability of insulin to stimulate glucose utilization by 3T3-L1 adipocytes increases approximately five-fold during the first 4 days following treatment of the cells with a differentiation medium. It was found that glucose utilization in 3T3-L1 adipocytes is regulated in a reciprocal fashion by insulin and GH. Bovine or human GH directly inhibit up to 50% of insulin-stimulated [14C]-glucose incorporation into lipids in a concentration-dependent manner. The 3T3-L1 sensitivity to GH appears to be at the maximum (50% inhibition of an insulin response) immediately following removal of the cells from the differentiation medium and remains essentially constant during the subsequent 4 days. The GH inhibition of insulin action does not appear to be due GH enhancement of cellular degradation of insulin, competitive binding of GH to the insulin receptor, or GH-induced decrease in cell number. The 3T3-L1 adipocyte system appears to be a sensitive and reliable in vitro model with which to study the molecular mechanisms involved in both GH antagonism of insulin action and development of hormone responsiveness during cellular differentiation into adipocytes. 相似文献

研究了重穗型杂交水稻培矮 6 4s/E3 2的灌浆过程和强、弱势颖花中内源IAA、ABA和GA1 GA3水平的动态状况。籽粒发育过程中不同内源激素水平高低依次为 :IAA >GA1 GA3>ABA。IAA和ABA水平在强势颖花中较高而GA1 GA3水平在弱势颖花中较高。 3种激素水平的变化与谷粒增重速率之间均存在正相关 ,两个最高的相关系数值分别存在于单位鲜重样本的IAA含量(ng/gFW ) 与籽粒鲜重的增重速率之间 (r =0 .82 1 8 )和单个籽粒IAA含量 (ng/grain)与籽粒干重的增重速率之间 (r =0 .8485 )。推测启动和维持籽粒灌浆过程可能需要较高的IAA水平 ;ABA可能具有促进籽粒中同化物的累积和种子成熟的作用 ;GA1 GA3可能具有保持弱势颖花活性的特殊作用 相似文献

Summary Participation of calmodulin, clathrin, and actin in receptor mediated endocytosis of gonadotropin-releasing hormone (GnRH) was studied in an in vitro system of dispersed pituitary cells with a triple staining procedure. Cells were incubated in D-Lys6-Pro9-Des10-GnRH-biotin and stained with avidin-peroxidase-diaminobenzidine. Calmodulin, clathrin, and actin as well as luteinizing hormone were identified by indirect immunofluorescence with FITC- and rhodamine-labeled second antibody. 采用全基因分段合成和补丁连接相结合的方法构建了人甲状旁腺素 (hPTH) (1~ 34)肽基因 ,并利用GST融合表达系统 ,在大肠杆菌中克隆和可溶性表达了hPTH(1~ 34)肽基因。融合蛋白表达量占菌体总蛋白质的 2 5 %以上 ,经高密度发酵、亲和纯化后 ,在每升发酵液中得到了 10 g融合蛋白。融合蛋白经肠激酶一步加工并亲合纯化和反相脱盐后 ,最终可以得到约 0 .6g /L人甲状旁腺素 (1~ 34)肽纯品 ,样品的理论回收值达到了4 8.75 %。产物的纯度和性质经HPLC、毛细管电泳、质谱分析、N端测序等得到证明 ,并与化学合成产物的结果相吻合。兔肾皮质细胞测活表明 ,重组产物与化学合成人甲状旁腺素 (1~ 34)肽具有相近的腺苷酸环化酶激活活性。 相似文献

目的:探讨定坤丹对多囊卵巢综合征患者性激素水平、胰岛素抵抗及妊娠情况的的影响。方法:选择2016年5月至2018年5月我院收治的210例多囊卵巢综合征患者,根据随机化原则将其分为两组,每组105例。对照组患者接受达英-35与二甲双胍治疗,研究组在对照组的基础上口服定坤丹治疗。连续治疗3个月后,比较两组治疗前后的性激素水平及胰岛各相关指标的变化。治疗结束后随访1年,比较两组的排卵及妊娠情况。结果:与治疗前相比,两组治疗后的血清睾酮(Testosterone,T)、黄体生成素(Luteinizing hormone,LH)及LH/FSH均明显降低,且研究组以上指标均显著低于对照组(P<0.05)。两组治疗前后的卵泡刺激素(Follicle-stimulating hormone,FSH)水平比较无统计学差异(P>0.05)。与治疗前相比,两组治疗后的空腹胰岛素(Fasting insulin,FINS)、餐后2 h胰岛素(Postprandial 2 hours insulin,2hPINS)及β细胞胰岛素分泌功能(Homeostasis model assessment ofβcell,HOMA-β)均明显升高,胰岛素敏感指数(Homeostasis model assessment-Insulin sensitivity index,HOMA-IS)明显降低,且研究组以上指标较对照组改善更明显(P<0.05)。研究组排卵及成功妊娠率均显著高于对照组(P<0.05)。结论:定坤丹有助于改善多囊卵巢综合征患者性激素水平及胰岛素抵抗,提高排卵及妊娠率。 相似文献

GH plays an essential role in the growing child by binding to the growth hormone receptor (GHR) on target cells and regulating multiple growth promoting and metabolic effects. Mutations in the GHR gene coding regions result in GH insensitivity (dwarfism) due to a dysfunctional receptor protein. However, children with idiopathic short stature (ISS) show growth impairment without GH or GHR defects. We hypothesized that decreased expression of the GHR gene may be involved. To test this, we investigated whether common genetic variants (microsatellites, SNPs) in regulatory regions of the GHR gene region were associated with the ISS phenotype. Genotyping of a GT‐repeat microsatellite in the GHR 5′UTR in a Montreal ISS cohort (n = 37 ISS, n = 105 controls) revealed that the incidence of the long/short (L/S) genotype was 3.3× higher in ISS children than controls (P = 0.04, OR = 3.85). In an Italian replication cohort (n = 143 ISS, n = 282 controls), the medium/short (M/S) genotype was 1.9× more frequent in the male ISS than controls (P = 0.017, OR = 2.26). In both ISS cohorts, logistic regression analysis of 27 SNPs showed an association of ISS with rs4292454, while haplotype analysis revealed specific risk haplotypes in the 3′ haploblocks. In contrast, there were no differences in GT genotype frequencies in a cohort of short stature (SS) adults versus controls (CARTaGENE: n = 168 SS, n = 207 controls) and the risk haplotype in the SS cohort was located in the most 5′ haploblock. These data suggest that the variants identified are potentially genetic markers specifically associated with the ISS phenotype. 相似文献
