This study was undertaken to investigate the effects of both nitrogen (N) and potassium (K) rates on rice resistance to brown spot, caused by the fungus Bipolaris oryzae. Rice plants (cultivar ‘Metica 1’) were grown in soil corrected with 0, 25, 50, 75 and 100 mg of N / kg (as NH4NO3) of soil as well as with 25, 50, 75, 125 and 150 mg of K / kg (as KCl) of soil. Thirty‐three‐day‐old plants were inoculated with a suspension of Bipolaris oryzae conidia and the incubation period (IP), number of lesions (NL) per cm2 of leaf area and disease severity was evaluated. Disease severity was scored at 24, 48, 72, 96, 120 and 144 h after inoculation and data were used to obtain the area under brown spot progress curve (AUBSPC). Soil plant analysis development (SPAD) index, plant dry weight and concentration of N and K in leaf tissues were also determined for both non‐inoculated (NI) and inoculated (IN) plants. Concentration of N in leaf tissue increased as the N rates in the soil increased. Concentration of K in leaf tissue increased sharply as the K rates in the soil increased for both NI and IN plants. Concentration of K in leaf tissue was not affected by N rates. The IP increased as the N rates increased, but was somewhat less impacted by increasing K rates. The NL decreased as the N rates increased. The NL dramatically declined at the highest K rates. The AUBSPC dramatically declined as the N and K rates in the soil increased. SPAD index values increased as the N and K rates in the soil increased for both NI and IN plants. Plant dry weight increased as the N and K rates in the soil increased for both NI and IN plants. Results from this study suggest that combining high N and K rates may contribute to reducing the intensity of brown spot in rice while improving plant development. 相似文献

Summary Cryopreservation (liquid nitrogen, −196°C) represents the only safe and cost-effective option for long-term conservation of
germplasm of non-orthodox seed species, vegetatively propagated species, and of biotechnology products. Classical cryopreservation
techniques, which are based on freeze-induced dehydration, are mainly employed for freezing undifferentiated cultures and
apices of cold-tolerant species. New cryopreservation techniques, which are based on vitrification of internal solutes, are
successfully employed with all explant types, including cells suspensions and calluses, apices, and somatic and zygotic embryos
of temperate and tropical species. The development of cryopreservation protocols is significantly more advanced for vegetatively
propagated species than for recalcitrant seed species. Even though its routine use is still limited, there are a growing number
of examples where cryopreservation is employed on a large scale for different types of materials, including seeds with orthodox
and intermediate storage behaviour, dormant buds, pollen, biotechnology products, and apices sampled from in vitro plantlets of vegetatively propagated species. Cryopreservation can also be employed for uses other than germplasm conservation,
such as cryoselection, i.e., the selection through freezing of samples with special properties, or cryotherapy, i.e., the
elimination of viruses from infected plants through apex cryopreservation. Because of its high potential, it is expected that

Measurement of nitrogen isotopic composition (15N) of plants and soil nitrogen might allow the characteristics of N transformation in an ecosystem to be detected. We tested the measurement of 15N for its ability to provide a picture of N dynamics at the ecosystem level by doing a simple comparison of 15N between soil N pools and plants, and by using an existing model. 15N of plants and soil N was measured together with foliar nitrate reductase activity (NRA) and the foliar NO3– pool at two sites with different nitrification rates in a temperature forest in Japan. 15N of plants was similar to that of soil NO3– in the high-nitrification site. Because of high foliar NRA and the large foliar NO3– pool at this site, we concluded that plant 15N indicated a great reliance of plants on soil NO3– there. However, many 15N of soil N overlapped each other at the other site, and 15N could not provide definitive evidence of the N source. The existing model was verified by measured 15N of soil inorganic N and it explained the variations of plant 15N between the two sites in the context of relative importance of nitrification, but more information about isotopic fractionations during plant N uptake is required for quantitative discussions about the plant N source. The model applied here can provide a basis to compare 15N signatures from different ecosystems and to understand N dynamics. 相似文献

Proteomics is a research field aiming to characterize molecular and cellular dynamics in protein expression and function on a global level. The introduction of proteomics has been greatly broadening our view and accelerating our path in various medical researches. The most significant advantage of proteomics is its ability to examine a whole proteome or sub-proteome in a single experiment so that the protein alterations corresponding to a pathological or biochemical condition at a given time can be considered in an integrated way. Proteomic technology has been extensively used to tackle a wide variety of medical subjects including biomarker discovery and drug development. By complement with other new technique advances in genomics and bioinformatics, proteomics has a great potential to make considerable contribution to biomarker identification and to revolutionize drug development process. High deposition of gaseous/particulate fluorides and other air pollutants has resulted in an acidification and probable formation of soluble AlFx complexes in the soil in the vicinity of the industrial complex of Cubatão, SE Brazil. With the present field study we aimed at determining the contribution of F and Al uptake from fluoride-contaminated soil, supposedly as AlFx complexes, to the increase of foliar F and Al contents in saplings of an Al-accumulator tree species (Tibouchina pulchra) which were concomitantly exposed to fluoride-contaminated air and also the proportional contribution of both air and soil contamination to the mentioned foliar accumulation of these elements. The seasonal variations in F and Al accumulation and possible metabolic changes in the plants due to F and Al accumulation were also investigated. The saplings were exposed during three consecutive periods of 16 weeks to: (a) air and soil from a reference site (PVnoF); (b) air or soil from two polluted sites (CM-high air pollution, low F and MV-high air pollution, high F); and (c) both air and soil from these polluted sites. After exposure, the changes in the foliar concentrations of F and Al, the relations between both element contents and their relationships with oxidative stress indicators were determined. The data were grouped in three matrices: PVnoF–CMlwF and PVnoF–MVhgF, taking in account the possible air/soil exposure combinations in each, and soil/air from all sites. The slight F accumulation in plants of PVnoF–CMlwF matrix was a result of higher uptake from soil than from air (54 and 46%, respectively). At PVnoF–MVhgF matrix, the extremely high F accumulation in leaves of T. pulchra could be attributed to the combination of both air and soil contamination (83 and 17%, respectively). T. pulchra always showed higher foliar Al concentrations than 1000 g g–1 dry mass, mainly after exposure to air and soil of both polluted sites (CMlwF and MVhgF). A highly significant linear regression was estimated between molar Al and F contents, taking in account the data obtained for saplings of T. pulchra cultivated in the different soils and exposed to ambient air of PVnoF, suggesting that both elements were taken as Al–F complexes from soil. The uptake of fluorides from air and/or soil of MVhgF caused significant metabolic changes in T. pulchra, but visible injury supposedly induced by fluorides were observed only when the foliar F contents surpassed 700 g g–1 dry mass. A field experiment was conducted in the South of Portugal with young (0-3 years old) non-bearing ‘Lane Late’ orange trees. Five nitrogen (N) rates were used in a randomised block design with three replicates. The 180 g N tree−1 over three years led to the greatest canopy width (176 cm) and volume (2,697 dm3). The greatest rate applied (720 g N tree−1 in the three years) led to the largest flower yield. Nitrogen concentration in the flowers significantly increased with fertilizer N, and also with the flowering period up to the 23rd day, declining thereafter. Flower yield was strongly correlated (r = 0.99, p < 0.001) with flower N concentration. Nutrient composition of flowers and of mature leaves from the spring flush was compared. Significant correlations were found for N (r = 0.47, p <0.01), P (r = -0.49, p <0.01), K (r = 0.44, p <0.05) and Ca (r = 0.87, p <0.001), suggesting that flowers can be used as a tool to diagnose the nutritional status of trees. Canonical analysis (with N treatment as dummy-variables) showed strong relationships between canopy width and N, which were greater at the larger rates of fertilizer application, and strong and inverse relationships between K and Mg, also with the greatest N rates. 相似文献

In anti-malaria operations the use of DDT for indoor residual spraying has declined substantially over the past 30years, but this insecticide is still considered valuable for malaria control, mainly because of its low cost relative to alternative insecticides. Despite the development of resistance to DDT in some populations of malaria vector Anopheles mosquitoes (Diptera: Culicidae), DDT remains generally effective when used for house-spraying against most species of Anopheles, due to excitorepellency as well as insecticidal effects. A 1990 cost comparison by the World Health Organization (WHO) found DDT to be considerably less expensive than other insecticides, which cost 2 to 23 times more on the basis of cost per house per 6 months of control. To determine whether such a cost advantage still prevails for DDT, this paper compares recent price quotes from manufacturers and WHO suppliers for DDT and appropriate formulations of nine other insecticides (two carbamates, two organophosphates and five pyrethroids) commonly used for residual house-spraying in malaria control programmes. Based on these 'global' price quotes, detailed calculations show that DDT is still the least expensive insecticide on a cost per house basis, although the price appears to be rising as DDT production declines. At the same time, the prices of pyrethroids are declining, making some only slightly more expensive than DDT at low application dosages. Other costs, including operations (labour), transportation and human safety may also increase the price advantages of DDT and some pyrethroids vs. organophosphates and carbamates, although possible environmental impacts from DDT remain a concern. However, a global cost comparison may not realistically reflect local costs or effective application dosages at the country level. Recent data on insecticide prices paid by the health ministries of individual countries showed that prices of particular insecticides can vary substantially in the open market. Therefore, the most cost-effective insecticide in any given country or region must be determined on a case-by-case basis. Regional coordination of procurement of public health insecticides could improve access to affordable products. 相似文献

Bacillus strains producing useful enzymes, have been isolated from various environments. Some of these enzymes, such as proteases
and cellulases from alkaliphilic Bacillus strains, have been commercialized and have brought great advantages to industry and domestic life. To support further development
of the enzyme industry, we initiated analysis of the genome of Bacillus halodurans C-125, which is 4.25 Mb in size, and constructed a physical and genetic map for comparison with the Bacillus subtilis chromosome. Systematic sequencing of the whole genome of Bacillus halodurans C-125 has been automated since the beginning of May 1998, and sequencing of 98% of the whole genome has been done so far.
Through genome analysis, it became apparent that the genome organization of alkaliphilic Bacillus halodurans C-125 is totally different from that of B. subtilis orthologues.
