Field trial analysis of nitrate reductase co-suppression: a comparative study of 38 combinations of transgene loci 总被引:7,自引:0,他引:7
Co-syppression of host genes and 35S transgenes encoding nitrate reductase was previously reported in transgenic tobacco plants (Nicotiana tabacum cv. Paraguay or Burley) using either a full-length cDNA or fragments devoid of the 3 and/or 5 UTR. Co-suppression was previously shown to affect a limited fraction of the progeny of one transgenic tobacco line homozygous for a single transgene locus, and the phenomenon occurred at each generation. In this work, 38 combinations of transgene loci derived from 13 independent transgenic lines homozygous for a single transgene locus were field-tested under two different conditions in an attempt to determine the corresponding frequencies of co-suppression, i.e. the percentage of plants showing co-suppression.Each of the 13 homozygous lines exhibited a different frequency of co-suppression, ranging from 0% to 57%. High frequencies were found to be associated with transgene loci carrying a high number of copy of the transgene, suggesting a transgene dose effect. Combinations carrying 2 non-allelic transgene loci in a hemizygous state exhibited frequencies of co-suppression between those of each of the 2 transgene loci in a homozygous state, while combinations carrying 2 non-allelic transgene loci in a homozygous state exhibited frequencies of co-suppression higher than the sum of those of the 2 transgene loci alone in a homozygous state, clearly confirming a transgene dose effect.Co-suppression frequencies were increased when the plants were grown initially in vitro, suggesting some environmental effect. Black raspberry necrosis virus (BRNV) reaches only very low concentrations in herbaceous plants and is difficult to maintain in culture. However, in a mixed culture with an unrelated virus, Solanum nodiflorum mottle (SNMV), in the genus Sobemovirus, the concentration of BRNV particles increases about 1000‐fold. In attempts to produce monoclonal antibodies (MAbs) to BRNV for diagnostic use, purified virus particles from the mixed virus culture were used as immunogen and the resultant antibodies screened against cultures of SNMV alone, BRNV+SNMV and healthy plant extracts. None of the virus‐specific MAbs obtained in this way was specific to BRNV but six were specific to SNMV. Although the original objective was not achieved, the SNMV MAbs were characterised and used to study serological properties of SNMV and other Sobemoviruses. Characterisation of the six SNMV MAbs showed that four were IgG3, one IgG1 and the other IgG2b. SNMV was detected by all six MAbs in ELISA, by five in Western blotting, by three in agarose gel double diffusion tests, but only one was suitable for trapping virus particles in immuno‐electron microscopy (IEM). In Western blotting using virus in sap extracts of Nicotiana clevelandii, each of the five MAbs detected a single major band of Mc. 31 000 in sap containing SNMV, and additional bands of lower mass attributed to degradation of coat protein. In various serological tests, no cross‐reactions were detected between SNMV and seven other viruses from the genus Sobemovirus. However, in IEM but not in Western blotting, significant cross‐reactions were observed between SNMV and Velvet tobacco mottle virus, another species from the genus Sobemovirus. The incidence of disease caused by tobacco leaf curl geminivirus (TbLCV) in ten tobacco growing areas of India ranged from 1.2% to 77%. The highest incidence of disease was observed in Andhra Pradesh (77%) followed by Gujarat (59%), Karnataka (17%), Bihar (11.6%) and West Bengal (5.4%). Under field conditions, an average of 32 adult whiteflies (Bemisia tabaci) per plant were recorded in Andhra Pradesh followed by Gujarat (20), Karnataka (12), Bihar (8) and West Bengal (5). In sequential sowings at Bangalore, all the plants were infected within 90 days in plots planted from February to June. Infection in plots planted later was progressively less. There was a positive correlation between whitefly catches and the final incidence of leaf curl disease in plantings. TbLCV was transmitted by Bemisia tabaci to 35 plant species, including Beta vulgaris, Capsicum annuum, Carica papaya, Cymopsis tetragonoloba, Lycopersicon esculentum, Sesamum indicum, Phaseolus vulgaris and Petunia hybrida. Forty five TbLCV isolates from different parts of India induced four distinct types of symptoms on tobacco cultivars Samsun and Anand 119. Group 1 isolates caused severe curling and cup-shaped enations; group II isolates induced pale green leaves, pit-like depressions and thorny enations: group III isolates caused leathery leaves, narrow and tiny protruding enations between the veins, and group IV isolates induced irregular thickening and swelling of veins and green flap-like enations on veins. Nylon net covers protected tobacco seedlings in nursery beds for 45 days. Ricinus communis and Helianthus annuus sown around the tobacco nursery bed as barrier crops attracted adult whiteflies and decreased the number found on tobacco. 相似文献

The expression of nitrite reductase (NiR; EC 1.7.7.1), the second enzyme in the nitrate assimilatory pathway, is regulated
by nitrate as well as by end-products of nitrate assimilation, namely, glutamine (Gln) and asparagine (Asn). Nitrate induces
expression of the NiR gene. Previously, using deletion analysis of the spinach (Spinacia oleracea L.) NiR gene promoter in transgenic tobacco (Nicotiana tabacum L.) and in-vivo dimethyl sulfate footprinting, we had identified the region between −230 bp and −180 bp as being critical
for nitrate inducibility of this gene. In the present study, we show that the region from +1 to +67, which forms part of its
untranslated leader, is important for minimal induction in the presence of nitrate. Electrophoretic mobility shift assays
reveal concentration-dependent and competitive binding of a factor in tobacco nuclear extracts to this region. In the presence
of Gln or Asn, the expression of spinach NiR is repressed. This repression is observed with the full-length NiR promoter (−3100 bp)
as well as with the shortest promoter (−230 bp) that gives nitrate induction, which includes the +67 bp leader sequence. The
repressed expression of the gene is not the result of reduced nitrate accumulation in the presence of the nitrogen metabolites.

利用安徽省凤阳县1955~1996年烤烟上中等烟比重资料,划分出该县烤烟质量优劣年景.采用统计学方法研究烤烟质量与气象条件的关系,结果表明:旱涝灾害、成熟期高温热害和强烈日照是造成凤阳烤烟劣质的主要气象灾害.运用谐波分析法分析了烤烟劣质或严重劣质年景与气象灾害的关系及其随时间变化的规律,并尝试了预测未来若干年内气象灾害年的分布;探讨了防灾减灾对策. 相似文献
