Together with flow injection analysis (FIA), a chemiluminescence (CL) fiber optic biosensor system has been developed for determining glutamine in animal cell cultures. Glutaminase (GAH) and glutamate oxidase (GLO) were onto separate porous aminopropyl glass beads via glutaraldehyde activation and packed to form an enzyme column. These two enzymes acted in sequence on glutamine to produce hydrogen peroxide, which was then reacted with luminol in the presence of ferricyanide to produce a light signal. An anion exchanger was introduced on-line to eliminate interfering endogenous glutamate in view of its negative charge at pH above 3.22 (isoelectric pH). Among several resins tested, the acetate form was most effective, and this type of ion exchanger also effectively adsorbed uric acid, acetaminophen, and aspartic acid.There was an excellent linear relationship between the CL response and standard glutamine concentration in the range 1 to 100 muM. A complete analysis could be performed in 2 min, including sampling and washing with a good reproducibility (+/- 4.4%). Both the bi-enzymic and ion exchange columns were useful for at least 500 analyses when the biosensor system was applied for the glutamine determination in murine hybridoma cell cultures and insect cell cultures. The third phase of Wright's shifting-balance theory involves the export of adaptive gene combinations from one subpopulation to another. Previous results have demonstrated that this can occur at very low migration rates, but it has been argued that this simply reflects the ability of migration to overcome selection and fix any (even deleterious) alleles. Here, previous analyses are extended by concentrating on the critical balance between forward and reverse migration rates that still allows phase III to proceed. It is shown that selective advantage, dominance, recombination rate, and the number of loci all affect the ability of a genotype to invade and become fixed in a new subpopulation, but it is unlikely that phase III will occur in the absence of differential migration unless the invading genotype consists of a few dominant loci with a large selection advantage, spreading into a few populations of lower fitness. Therefore, as was envisioned by Wright, differential migration from more to less fit populations will be necessary for phase III to occur under most circumstances. 相似文献

Pyrethroid resistance inHelicoverpa armigera provides a model system in which to study evolution in natural populations. Resistance is seen to evolve as a consequence of selection pressure that varies within and between life-stages and gene flow. Although three different mechanisms are involved, present day fluctuations in phenotype frequency can be explained by variation in only one of these, metabolic resistance, that is inherited as a single, incompletely dominant gene,mfo. Selective mortality of phenotypes occurs in both larvae and adults in the presence of the pyrethroid insecticides. Although most individuals of all three genotypes are killed in young larvae, selection in this age-class contributes significantly to evolution of resistance. While there is some evidence of reduced fitness of resistant pupae during winter diapause, most of the decline in resistance frequencies each spring occurs as a result of immigration of susceptible individuals into insecticide-treated populations. 相似文献

ty) from the two ears in response to various changes in body and head positions. Upon assuming a lateral recumbent position, the T
ty on the lower side increased while that on the upper side decreased. Pressure application over a wide area of the lateral chest only caused inconsistent and obscure asymmetric changes in T
ty. A lateral flexion of the head with the subject sitting upright and a rotation of the head to the side in a supine position induced an increase in the T
ty on the lower side compared to that on the upper side. The temperature and blood flow of the forehead often decreased on the lower side and increased on the upper side, although such responses were not always concomitant with the asymmetric changes in T
ty. A dorsal flexion of the head with the subject in a reclining position caused a slight increase in the T
ty, whereas raising the head upright induced a slight decrease in them. Two additional experiments were carried out with single photon emission computed tomography using 99mTc-hexamethylpropyleneamine oxime as tracer, and a slight, relative decrease in counts was noted in the right hemisphere during rotation of the head to the right. These results would strongly suggest that unilateral increases and decreases in T

Climate change is altering hydrological cycles globally, and in Mediterranean (med-) climate regions it is causing the drying of river flow regimes, including the loss of perennial flows. Water regime exerts a strong influence over stream assemblages, which have developed over geological timeframes with the extant flow regime. Consequently, sudden drying in formerly perennial streams is expected to have large, negative impacts on stream fauna. We compared contemporary (2016/17) macroinvertebrate assemblages of formerly perennial streams that became intermittently flowing (since the early 2000s) to assemblages recorded in the same streams by a study conducted pre-drying (1981/82) in the med-climate region of southwestern Australia (the Wungong Brook catchment, SWA), using a multiple before-after, control-impact design. Assemblage composition in the stream reaches that remained perennial changed very little between the studies. In contrast, recent intermittency had a profound effect on species composition in streams impacted by drying, including the extirpation of nearly all Gondwanan relictual insect species. New species arriving at intermittent streams tended to be widespread, resilient species including desert-adapted taxa. Intermittent streams also had distinct species assemblages, due in part to differences in their hydroperiods, allowing the establishment of distinct winter and summer assemblages in streams with longer-lived pools. The remaining perennial stream is the only refuge for ancient Gondwanan relict species and the only place in the Wungong Brook catchment where many of these species still persist. The fauna of SWA upland streams is becoming homogenised with that of the wider Western Australian landscape, as drought-tolerant, widespread species replace local endemics. Flow regime drying caused large, in situ alterations to stream assemblage composition and demonstrates the threat posed to relictual stream faunas in regions where climates are drying. 相似文献

Dynamic material flow analysis (dMFA) is widely used to model stock-flow dynamics. To appropriately represent material lifetimes, recycling potentials, and service provision, dMFA requires data about the allocation of economy-wide material consumption to different end-use products or sectors, that is, the different product stocks, in which material consumption accumulates. Previous estimates of this allocation only cover few years, countries, and product groups. Recently, several new methods for estimating end-use product allocation in dMFA were proposed, which so far lack systematic comparison. We review and systematize five methods for tracing material consumption into end-use products in inflow-driven dMFA and discuss their strengths and limitations. Widely used data on industry shipments in physical units have low spatio-temporal coverage, which limits their applicability across countries and years. Monetary input–output tables (MIOTs) are widely available and their economy-wide coverage makes them a valuable source to approximate material end-uses. We find four distinct MIOT-based methods: consumption-based, waste input–output MFA (WIO-MFA), Ghosh absorbing Markov chain, and partial Ghosh. We show that when applied to a given MIOT, the methods’ underlying input–output models yield the same results, with the exception of the partial Ghosh method, which involves simplifications. For practical applications, the MIOT system boundary must be aligned to those of dMFA, which involves the removal of service flows, sector (dis)aggregation, and re-defining specific intermediate outputs as final demand. Theoretically, WIO-MFA, applied to a modified MIOT, produces the most accurate results as it excludes massless and waste transactions. The aim of this study was to delineate the flow patterns in a non-unidirectional flow field inside a ventricle-shaped cell
culture chamber, and examine the resulting morphology and integrity of the endothelium in select regions of the monolayer.
The chamber was perfused by pulsatile flow, and the coherent motion of the fluid was studied using flow visualization aided
by image analysis. Four distinct flow patterns were discerned and examined: central jet, flow impingement, flow separation,
and recirculating eddies. The influence of these patterns on endothelial cell morphology was assessed after 20 h of exposure
to flow. There were no signs of damage to the endothelium in the jet region nor was there evidence of cell alignment with
the flow. Yet, there were changes in cell morphology and cytoskeletal architecture as compared to control. By contrast, within
the eddies where the flow was highly disturbed, there was apparent damage to the endothelium. Thus, exposure of cells to random
velocity fluctuations in regions of quasi-static flow compromises the integrity of the monolayer. Identification of such sites
and acquisition of the knowledge necessary to protect the cells from denudation will be valuable for the endothelialization

Migration is ubiquitous and can strongly shape food webs and ecosystems. Less familiar, however, is that the majority of life cycle, seasonal and diel migrations in nature are partial migrations: only a fraction of the population migrates while the other individuals remain in their resident ecosystem. Here, we demonstrate different impacts of partial migration rendering it fundamental to our understanding of the significance of migration for food web and ecosystem dynamics. First, partial migration affects the spatiotemporal distribution of individuals and the food web and ecosystem-level processes they drive differently than expected under full migration. Second, whether an individual migrates or not is regularly correlated with morphological, physiological, and/or behavioural traits that shape its food-web and ecosystem-level impacts. Third, food web and ecosystem dynamics can drive the fraction of the population migrating, enabling the potential for feedbacks between the causes and consequences of migration within and across ecosystems. These impacts, individually and in combination, can yield unintuitive effects of migration and drive the dynamics, diversity and functions of ecosystems. By presenting the first full integration of partial migration and trophic (meta-)community and (meta-)ecosystem ecology, we provide a roadmap for studying how migration affects and is affected by ecosystem dynamics in a changing world. 相似文献

The objective of this study was to examine membrane filtration of a single stranded DNA (ssDNA) with 60 thymine nucleotides, and to elucidate the variables controlling its transmission across track-etched porous membranes. Dead end filtration measurements were performed using different pore size membranes (10, 15, and 30 nm) at different transmembrane pressures in solutions with ionic strength ranging from 0 to 1000 mM NaCl. The diffusivity of the ssDNA was determined using fluorescence recovery after photobleaching, yielding hydrodynamic radii ranging from 1.6 to 2.8 nm, with values decreasing with increasing solution ionic strength. Despite the small ssDNA/membrane pore size, nearly 100% rejection was observed for measurements performed with the 10 and 15 nm pore size membranes under low-ionic strength conditions. These high rejections can be attributed to strong repulsive electrostatic ssDNA-membrane interactions. With increasing ionic strength, electrostatic interactions as well as the effective size of the ssDNA decreases and the flexibility of the ssDNA increases, leading to a reduction in ssDNA rejection. A design of experiments approach was used to plan filtration experiments that adequately covered the variable space with a manageable number of experiments. The results yielded an empirical expression relating ssDNA rejection to pore size, solution ionic strength and transmembrane pressure. There was evidence of flow induced elongation at high-transmembrane pressures in the 30 nm pore size membranes, but not in the smaller pore size membranes. These results are consistent with critical flux estimates developed using a free draining model for the ssDNA. 相似文献
