Soaks (areas of mesotrophic/minerotrophic vegetation within acid bog) add to the overall heterogeneity and biodiversity of raised bog landscapes due to the presence of flora and fauna communities not typically associated with acid bog systems. A field experiment was set up to investigate the potential to restore the minerotrophic and aquatic communities that previously occurred within a soak of an oceanic raised bog in Ireland, which has recently undergone acidification with the expansion of acid bog type vegetation. Three different treatments, control (intact sphagnaceous raft), permeable (sphagnaceous raft removed), and enclosed (sphagnaceous raft removed and plots isolated from surrounding surface water influence) were applied to a total of six plots (each measuring 4 × 4 m), each treatment consisting of two replicates. Within 3 years a sphagnaceous raft with similar vegetation to the surroundings had developed in both permeable plots, while aquatic communities similar to those that occurred at the site in the past had established within the enclosed pots. Our results show that with manipulation of local hydrology it is possible to recreate conditions suitable for aquatic plant communities that once characterized the site. The results also give an insight into the likely processes responsible for the initial terrestrialization of the entire soak over the past century. Application of the results in relation to the site and the widespread practice of restoring bog vegetation on degraded peatlands are discussed.  相似文献

Peter Karlson and Martin Lüscher used the term pheromone for the first time in 19591 to describe chemicals used for intra-species communication. Pheromones are volatile or non-volatile short-lived molecules2 secreted and/or contained in biological fluids3,4, such as urine, a liquid known to be a main source of pheromones3. Pheromonal communication is implicated in a variety of key animal modalities such as kin interactions5,6, hierarchical organisations3 and sexual interactions7,8 and are consequently directly correlated with the survival of a given species9,10,11. In mice, the ability to detect pheromones is principally mediated by the vomeronasal organ (VNO)10,12, a paired structure located at the base of the nasal cavity, and enclosed in a cartilaginous capsule. Each VNO has a tubular shape with a lumen13,14 allowing the contact with the external chemical world. The sensory neuroepithelium is principally composed of vomeronasal bipolar sensory neurons (VSNs)15. Each VSN extends a single dendrite to the lumen ending in a large dendritic knob bearing up to 100 microvilli implicated in chemical detection16. Numerous subpopulations of VSNs are present. They are differentiated by the chemoreceptor they express and thus possibly by the ligand(s) they recognize17,18. Two main vomeronasal receptor families, V1Rs and V2Rs19,20,21,22, are composed respectively by 24023 and 12024 members and are expressed in separate layers of the neuroepithelium. Olfactory receptors (ORs)25 and formyl peptide receptors (FPRs)26,27 are also expressed in VSNs.Whether or not these neuronal subpopulations use the same downstream signalling pathway for sensing pheromones is unknown. Despite a major role played by a calcium-permeable channel (TRPC2) present in the microvilli of mature neurons28 TRPC2 independent transduction channels have been suggested6,29. Due to the high number of neuronal subpopulations and the peculiar morphology of the organ, pharmacological and physiological investigations of the signalling elements present in the VNO are complex.Here, we present an acute tissue slice preparation of the mouse VNO for performing calcium imaging investigations. This physiological approach allows observations, in the natural environment of a living tissue, of general or individual subpopulations of VSNs previously loaded with Fura-2AM, a calcium dye. This method is also convenient for studying any GFP-tagged pheromone receptor and is adaptable for the use of other fluorescent calcium probes. As an example, we use here a VG mouse line30, in which the translation of the pheromone V1rb2 receptor is linked to the expression of GFP by a polycistronic strategy.  相似文献

Hydrocotyle ranunculoides L.f. is a stoloniferous perennial plant with floating and emergent leaves that is native to Europe. It is commonly used as an aquarium plant with little published information on its biology and natural range. In 1983 H. ranunculoides was first observed in the urban drainage network in the Canning River Regional Park, Western Australia. By 1991 the plant had extended throughout the drainage network into the river and adjacent wetlands. H. ranunculoides formed extensive mats, disrupting the ecology and recreational uses of the waterways, and posed a threat to other waterways. It is not known to be invasive in other Australian waterways. A group of state and local government and community members assessed environmental, technical and social interactions and developed an integrated managementstrategy for the weed, using a combination of physical, chemical and ecological techniques. The environmental significance of the affected waterways required the programme to be accompanied by appropriate ecological surveillance. The initial short-term control phase was completed successfully. An assessment of water quality and aquatic invertebrates during the initial phase showed only short-term disruption of river ecology following physical and chemical control. Dispersal on floating seaweeds depends on availability, viability, and trajectories of the rafts. In the southern hemisphere, the bull kelp Durvillaea antarctica is one of the most common floating seaweeds, but phylogeographic studies had shown low connectivity between populations from continental Chile, which could be due to limitations in local supply and dispersal of floating kelps. To test this hypothesis, the spatiotemporal dynamics of kelp strandings were examined in four biogeographic districts along the Chilean coast (28°–42°S). We determined the biomass and demography of stranded individuals on 33 beaches for three subsequent years (2013, 2014, 2015) to examine whether rafting is restricted to certain districts and seasons (winter or summer). Stranded kelps were found on all beaches. Most kelps had only one stipe (one individual), although we also frequently found coalesced holdfasts with mature males and females, which would facilitate successful rafting dispersal, gamete release, and reproduction upon arrival. High biomasses of stranded kelps occurred in the northern‐central (30°S–33°S) and southernmost districts (37°S–42°S), and lower biomasses in the northernmost (28°S–30°S) and southern‐central districts (33°S–37°S). The highest percentages and sizes of epibionts (Lepas spp.), indicative of prolonged floating periods, were found on stranded kelps in the northernmost and southernmost districts. Based on these results, we conclude that rafting dispersal can vary regionally, being more common in the northernmost and southernmost districts, depending on intrinsic (seaweed biology) and extrinsic factors (shore morphology and oceanography) that affect local supply of kelps and regional hydrodynamics.  相似文献

The basic objective of this study was to explore the application of Gelucire 43/01 for the design of multi-unit floating systems of a highly water-soluble drug diltiazem HCl. Diltiazem HCl-Gelucire 43/01 granules were prepared by melt granulation technique. The granules were evaluated for in vitro and in vivo floating ability, surface topography, and in vitro drug release. Aging effect on storage was vvaluated using scanning electron microscopy, hot stage polarizing microscopy (HSPM), differential scanning calorimetry (DSC), and in vitro drug release. Granules were retained in stomach at least for 6 hours. Approximately 65% to 80% drug was released over 6 hours with initial fast release from the surface. Surface topography, HSPM, DSC study of the aged samples showed phase transformation of Gelucire. The phase transformation also caused significant increase in drug release. In conclusion, hydrophobic lipid, Gelucire 43/01, can be considered as an effective carrier for design of a multi-unit floating drug delivery system of highly water-soluble drugs such as diltiazem HCl.  相似文献

Biological systematics provides taxonomic information and expertise for biogeography and biological invasion research and management. However, the systematic classification of most taxa, including some alien and invasive species, relies only on morphology. This applies for example to Sertularella tongensis Stechow, 1919 Stechow, E. (1919). Zur Kenntnis der Hydroidenfauna des Mittelmeeres, Amerikas und anderer Gebiete, nebst Angaben über einige Kirchenpauer'sche Typen von Plumulariden. Zoologische Jahrbuecher (Systematik), 42, 1172. [Google Scholar] (Cnidaria: Sertulariidae), an alien hydrozoan recently reported in the Mediterranean. Its genus affiliation is still unclear and controversial. Historically, it has been assigned to several existing genera. However, molecular data (COI, 16S, 18S, 28S) provided here do not support these previous assignments. Instead, integrative analyses combining morphological and molecular data support reassigning this species to a new genus, Bicaularia gen. nov. A biogeographic review revealed that this species is widely distributed in the mid- and low-latitude waters of the Indo-Pacific region, and is spreading to some tropical and temperate regions including the Mediterranean and Caribbean Seas, as well as Korea and Japan. This expansion may be attributable to human activity such as shipping through the Suez and the Panama Canals. However, the new record attached to a floating Sargassum algal fragment in the surface water of the open Pacific, might suggest a neglected cryptic spread pathway with the assistance of floating biological organisms. Our findings shed light on the complexity of the phylogeny and spread pathway of the investigated hydrozoan species. We regard the fine systematics as the first step towards future global sampling for molecular studies aiming to elucidate the origins and pathways for alien and invasive species.

The purpose of this research was to prepare a floating drug delivery system of diltiazem hydrochloride (DTZ). Floating matrix tablets of DTZ were developed to prolong gastric residence time and increase its bioavailability. Rapid gastrointestinal transit could result in incomplete drug release from the drug delivery system above the absorption zone leading to diminished efficacy of the administered dose. The tablets were prepared by direct compression technique, using polymers such as hydroxypropylmethylcellulose (HPMC, Methocel K100M CR), Compritol 888 ATO, alone or in combination and other standard excipients. Sodium bicarbonate was incorporated as a gas-generating agent. The effects of sodium bicarbonate and succinic acid on drug release profile and floating properties were investigated. A 32 factorial design was applied to systematically optimize the drug release profile. The amounts of Methocel K100M CR (X1) and Compritol 888 ATO (X2) were selected as independent variables. The time required for 50% (t50) and 85% (t85) drug dissolution were selected as dependent variables. The results of factorial design indicated that a high level of both Methocel K100M CR (X1) and Compritol 888 ATO (X2) favors the preparation of floating controlled release of DTZ tablets. Comparable release profiles between the commercial product and the designed system were obtained. The linear regression analysis and model fitting showed that all these formulations followed Korsmeyer and Peppas model, which had a higher value of correlation coefficient (r). While tablet hardness had little or no effect on the release kinetics and was found to be a determining factor with regards to the buoyancy of the tablets. Published: September 7, 2007  相似文献

For the high frequency selection of salt-tolerant doubled haploids (DHs) using rice anther culture, the efficiency of anther culture was investigated with different genotype, media condition and NaCl concentrations. The six F1 hybrids obtained by backcross or three-way cross between indica and japonica differed in salt tolerance. The efficiencies of callus induction and plant regeneration was decreased by NaCl concentration and salt tolerance of donor variety, and those in japonicas were higher than those in indicas. The percentages of callus induction in Gyehwa 5 (japonica, tolerant) and IR61633-B-2-2-1 (japonica, sensitive) were 21.1 and 13.5% on agar medium containing 0.3% NaCl, respectively. The plant regeneration of callus derived from anther floating culture in liquid media was less than that on solid medium. In four F1 hybrids, the frequencies of high salt-tolerant DHs were 21.4 and 8.9% in 0.3% NaCl medium and the control, respectively. The high frequency of salt-tolerant DHs could be selected in the callus induction medium (0.3% NaCl) and in the combinations crossed with salt-tolerant japonica as the third parent.  相似文献

We investigated whether rates of net primary production (NPP) and biomass turnover of floating grasses in a central Amazon floodplain lake (Lake Calado) are consistent with published evidence that CO2 emissions from Amazon rivers and floodplains are largely supplied by carbon from C4 plants. Ground‐based measurements of species composition, plant growth rates, plant densities, and areal biomass were combined with low altitude videography to estimate community NPP and compare expected versus observed biomass at monthly intervals during the aquatic growth phase (January–August). Principal species at the site were Oryza perennis (a C3 grass), Echinochloa polystachya, and Paspalum repens (both C4 grasses). Monthly mean daily NPP of the mixed species community varied from 50 to 96 g dry mass m?2 day?1, with a seasonal average (±1SD) of 64±12 g dry mass m?2 day?1. Mean daily NPP (±1SE) for P. repens and E. polystachya was 77±3 and 34±2 g dry mass m?2 day?1, respectively. Monthly loss rates of combined above‐ and below‐water biomass ranged from 31% to 75%, and averaged 49%. Organic carbon losses from aquatic grasses ranged from 30 to 34 g C m?2 day?1 from February to August. A regional extrapolation indicated that respiration of this carbon potentially accounts for about half (46%) of annual CO2 emissions from surface waters in the central Amazon, or about 44% of gaseous carbon emissions, if methane flux is included.  相似文献
