Investigation of protein‐ligand interactions obtained from experiments has a crucial part in the design of newly discovered and effective drugs. Analyzing the data extracted from known interactions could help scientists to predict the binding affinities of promising ligands before conducting experiments. The objective of this study is to advance the CIFAP (compressed images for affinity prediction) method, which is relevant to a protein‐ligand model, identifying 2D electrostatic potential images by separating the binding site of protein‐ligand complexes and using the images for predicting the computational affinity information represented by pIC50 values. The CIFAP method has 2 phases, namely, data modeling and prediction. In data modeling phase, the separated 3D structure of the binding pocket with the ligand inside is fitted into an electrostatic potential grid box, which is then compressed through 3 orthogonal directions into three 2D images for each protein‐ligand complex. Sequential floating forward selection technique is performed for acquiring prediction patterns from the images. In the prediction phase, support vector regression (SVR) and partial least squares regression are used for testing the quality of the CIFAP method for predicting the binding affinity of 45 CHK1 inhibitors derived from 2‐aminothiazole‐4‐carboxamide. The results show that the CIFAP method using both support vector regression and partial least squares regression is very effective for predicting the binding affinities of CHK1‐ligand complexes with low‐error values and high correlation. As a future work, the results could be improved by working on the pose of the ligands inside the grid.  相似文献

Internodal elongation in floating rice ( Oryza sativa L. cv. Habiganj Aman II) is known to be enhanced by treatment with ethylene or gibberellic acid (GA3) at high relative humidity (RH). However, ethylene-induced internodal elongation is inhibited at low RH. while GA3-induced internodal elongation is hardly affected by humidity. We examined the possible involvement of osmoregulation in the stimulation by GA3 of the elongation of internodes at low RH. Submergence and treatment with ethylene or GA33 at 100% RH increased the osmotic potential in internodes of excised stem segments, while GA3 at 20% RH maintained the osmotic potential at a low level. In internodes of stem segments that had been treated with GA3 at 20% RH, the activity of invertase and the level of soluble sugars were almost 2- and 1.5-fold higher, respectively, than those in internodes that had been treated with GA3 at 100% RH. These results indicate that one of the possible mechanisms by which GA3 promotes elongation of internodes at low RH involves the osmoregulation that is achieved by promotion of the synthesis of invertase.  相似文献

Polysaccharides, which have been explored to possess gelling properties and a wide margin of safety, were used to formulate single-unit floating matrix tablets by a direct compression technique. This work has the aim to allow continuous slow release of famotidine above its site of absorption. The floating approach was achieved by the use of the low density polypropylene foam powder. Polysaccharides (κ-carrageenan, gellan gum, xyloglucan, and pectin) and blends of polysaccharides (κ-carrageenan and gellan gum) and cellulose ethers (hydroxypropylmethyl cellulose, hydroxypropylcellulose, sodium carboxymethyl cellulose) were tried to modulate the release characteristics. The prepared floating tablets were evaluated for their floating behavior, matrix integrity, swelling studies, in vitro drug release studies, and kinetic analysis of the release data. The differential scanning calorimetry and Fourier transform infrared spectroscopy studies revealed that changing the polymer matrix system by formulation of polymers blends resulted in formation of molecular interactions which may have implications on drug release characteristics. This was obvious from the retardation in drug release and change in its mechanistics.  相似文献

城市绿色开敞空间是城市公共空间的重要组成部分,合理配置城市绿色开敞空间对城市宜居环境建设至关重要。空间可达性常用于评价公共服务设施空间布局的合理性,两步移动搜索法即是一种直观、运算简便、应用广泛的可达性计算方法,但该方法未考虑需求主体之间的差异性。本研究以南京市中心城区为例,利用手机信令数据提取需求者属性特征以及空间分布,通过分类细化需求主体改进搜索策略,改善两步移动搜索法传统研究未考虑不同群体差异性、统计数据滞后、空间尺度粗等弊端。结果表明,南京市中心城区绿色开敞空间可达性呈现出明显的空间分异特征,江南主城可达性总体比江北新区高,低值区集中在人口密度高、绿色开敞空间资源规模小的中心城区周边;不同人群的绿色开敞空间可达性总体格局相似,但在数值上老年人可达性均值是其他年龄段的14.60%,工作人群可达性均值是居住人群的86.02%,老年人作为弱势群体在享受绿色开敞空间服务中被边缘化。研究结果可为优化南京市绿色开敞空间布局提供科学依据,也可为其他公共服务设施可达性评价提供借鉴。  相似文献

Phenol is a commonly found organic pollutant in industrial wastewaters. Its ecotoxicological significance is well known and, therefore, the compound is often required to be removed prior to discharge. In this study, plant-bacterial synergism was established in floating treatment wetlands (FTWs) in an attempt to maximize the removal of phenol from contaminated water. A common wetland plant, Typha domingensis, was vegetated on a floating mat and augmented with three phenol-degrading bacterial strains, Acinetobacter lwofii ACRH76, Bacillus cereus LORH97, and Pseudomonas sp. LCRH90, to develop FTWs for the remediation of water contaminated with phenol. All of the strains are known to have phenol-reducing properties, and grow well in FTWs. Results showed that T. domingensis was able to remove a small amount of phenol from the contaminated water; however, bacterial augmentation enhanced the removal potential significantly, i.e., 0.146 g/m2/day vs. 0.166 g/m2/day, respectively. Plant biomass also increased in the presence of bacterial consortia; and inoculated bacteria displayed successful colonization/survival in the rhizosphere, root interior and shoot interior of the plant. Similarly, highest reduction in chemical oxygen demand (COD), biochemical oxygen demand (BOD5), and total organic carbon (TOC) was achieved by the combined application of plants and bacteria. The study demonstrates that the plant-bacterial synergism in a FTW may be a more effective approach for the remediation of phenol-contaminated water.  相似文献
