Life history theory predicts a trade‐off between immunostimulation and growth. Using a cross‐sectional study design, this study aims to test the hypothesis that C‐reactive protein (CRP) is negatively associated with height‐for‐age z‐scores (HAZ scores) and BMI‐for‐age z‐scores (BAZ scores) among 6‐ to 19‐year olds (N = 426) residing in five Nepalese communities. Dried blood spot (DBS) samples were collected and assayed for CRP using an in‐house enzyme immunoassay (EIA). Sex‐ and age‐group‐specific CRP quartiles were used to examine its association with growth in linear mixed‐effects (LME) models. A significant difference was found in the proportion of elevated CRP (>2 mg/L, equivalent to ~3.2 mg/L serum CRP) between 13‐ and 19‐year‐old boys (12%) and girls (4%). Concentrations of CRP were positively associated with HAZ score among adolescent (13–19 years) boys, which may indicate that individuals with greater energy resources have better growth and a better response to infections, thus eliminating the expected trade‐off between body maintenance (immunostimulation) and growth. Adolescent boys with low BAZ and HAZ scores had low CRP values, suggesting that those who do not have enough energy for growth cannot increase their CRP level even when infected with pathogens. Among adolescent girls a positive association was observed between CRP and BAZ scores suggesting the possible effects of chronic low‐grade inflammation due to body fat rather than infection. The association between CRP and growth was less evident among children (6–12 years) compared with adolescents, indicating that the elevated energy requirement needed for the adolescent growth spurt and puberty may play some role. The main cofactors of Photosystem II (PSII) are borne by the D1 and D2 subunits. In the thermophilic cyanobacterium Thermosynechococcus elongatus, three psbA genes encoding D1 are found in the genome. Among the 344 residues constituting the mature form of D1, there are 21 substitutions between PsbA1 and PsbA3, 31 between PsbA1 and PsbA2, and 27 between PsbA2 and PsbA3. In a previous study (Sugiura et al., J. Biol. Chem. 287 (2012), 13336-13347) we found that the oxidation kinetics and spectroscopic properties of TyrZ were altered in PsbA2-PSII when compared to PsbA(1/3)-PSII. The comparison of the different amino acid sequences identified the residues Cys144 and Pro173 found in PsbA1 and PsbA3, as being substituted in PsbA2 by Pro144 and Met173, and thus possible candidates accounting for the changes in the geometry and/or the environment of the TyrZ/His190 phenol/imidizol motif. Indeed, these amino acids are located upstream of the α-helix bearing TyrZ and between the two α-helices bearing TyrZ and its hydrogen-bonded partner, D1/His190. Here, site-directed mutants of PSII, PsbA3/Pro173Met and PsbA2/Met173Pro, were analyzed using X- and W-band EPR and UV-visible time-resolved absorption spectroscopy. The Pro173Met substitution in PsbA2-PSII versus PsbA3-PSII is shown to be the main structural determinant of the previously described functional differences between PsbA2-PSII and PsbA3-PSII. In PsbA2-PSII and PsbA3/Pro173Met-PSII, we found that the oxidation of TyrZ by P680+● was specifically slowed during the transition between S-states associated with proton release. We thus propose that the increase of the electrostatic charge of the Mn4CaO5 cluster in the S2 and S3 states could weaken the strength of the H-bond interaction between TyrZ and D1/His190 in PsbA2 versus PsbA3 and/or induce structural modification(s) of the water molecules network around TyrZ.  相似文献

The aerodynamic role of the elytra during a beetle's flapping motion is not well-elucidated, although it is well-recognized that the evolution of elytra has been a key in the success of coleopteran insects due to their protective function. An experimental study on wing kinematics reveals that for almost concurrent flapping with the hind wings, the flapping angle of the elytra is 5 times smaller than that of the hind wings. Then, we explore the aerodynamic forces on elytra in free forward flight with and without an effect of elytron-hind wing interaction by three-dimensional numerical simulation. The numerical results show that vertical force generated by the elytra without interaction is not sufficient to support even its own weight. However, the elytron-hind wing interaction improves the vertical force on the elytra up to 80%; thus, the total vertical force could fully support its own weight. The interaction slightly increases the vertical force on the hind wind by 6% as well.  相似文献

Autotomy is the ability to spontaneously self‐amputate a limb or other appendage, often as a reflexive action. This limb amputation typically occurs as a specialized defensive response to an attack from a predator and thereby enables the prey to escape from predation. Despite the benefits of escape, autotomized organisms lose the body part and its associated function. Here, we investigated the jumping behavior and performance of one‐leg‐autotomized and intact rice grasshoppers, Oxya yezoensis, to examine changes in jumping behavior after autotomy. The take‐off elevation of autotomized grasshoppers was 7.8° lower than in intact grasshoppers, resulting in nearly a 45° angle of take‐off, which maximized the jumping distance. Kinematic analyses of the jumping manner revealed that the angle of the femur during jumping differed between intact and autotomized grasshoppers, suggesting that the grasshoppers behaviorally change the take‐off elevation after autotomy. According to analyses of jumping performance, the degree of decline in performance differed between horizontal distance and vertical height. Even though they jumped on only one hind leg, one‐leg‐autotomized grasshoppers realized 69% performance along a horizontal distance relative to intact grasshoppers. In contrast, autotomized grasshoppers realized only a 44% performance in vertical height compared to intact grasshoppers. The difference in take‐off elevation between autotomized and intact grasshoppers is likely related to the observed difference in the magnitude of the decline in performance between horizontal distance and vertical height. These results suggest that rice grasshoppers may alter their take‐off elevation after limb autotomy to minimize the reduction in jumping distance.  相似文献

The reduction of insect herbivory is one of the services provided by tree diversity in forest ecosystems. While it is increasingly acknowledged that the compositional characteristics of tree species assemblages play a major role in triggering associational resistance to herbivores, underlying mechanisms are less well known. We addressed this question in the ORPHEE experiment by assessing pine processionary moth infestations (Thaumetopoea pityocampa) across a tree diversity gradient from pine monocultures to five species mixtures. We showed that tree species richness per se had no effect on the probability of attack by this pest. By contrast, the infestation rate was strongly dependent on plot composition. Mixtures of pines (Pinus pinaster) and birches (Betula pendula) were less prone to T. pityocampa infestations, whereas mixtures of pines and oaks (Quercus spp.) were more often attacked than pine monocultures. By taking into account the relative height of pines and associated broadleaved species, this effect could be explained by pine apparency. Pines were on average 343 ± 5 cm height. Birches, as fast growing trees, were slightly taller than pines (363 ± 6 cm), while oak trees were significantly smaller (74 ± 1 cm). Host trees of T. pityocampa were then partly hidden in mixtures of pines and birches but more apparent in mixtures with oaks. We suggest that reduced pine apparency disrupted visual cues used by female moths to select host trees prior to oviposition. This study highlights the need to take into account tree traits such as growth rate when selecting the tree species that have to be associated in order to improve forest resistance to pest insects.  相似文献

Biotin synthase (BioB) catalyses the final step in the biosynthesis of biotin. Aerobically purified biotin synthase contains one [2Fe-2S]2+ cluster per monomer. However, active BioB contains in addition a [4Fe-4S]2+ cluster which can be formed either by reconstitution with iron and sulfide, or on reduction with sodium dithionite. Here, we use EPR spectroscopy to show that mutations in the conserved YNHNLD sequence of Escherichia coli BioB affect the formation and stability of the [4Fe-4S]1+ cluster on reduction with dithionite and report the observation of a new [2Fe-2S]1+ cluster. These results serve to illustrate the dynamic nature of iron-sulfur clusters in biotin synthase and the role played by the protein in cluster interconversion.  相似文献

An N-terminal top-down sequencing approach was developed for IgG characterization, using high-resolution HPLC separation and collisionally activated dissociation (CAD) on a single-stage LCT Premier time of flight (TOF) mass spectrometer. Fragmentation of the IgG chains on the LCT Premier was optimized by varying the ion guide voltage values. Ion guide 1 voltage had the most significant effect on the fragmentation of the IgG chains. An ion guide 1 voltage value of 100 V was found to be optimum for the N-terminal fragmentation of IgG heavy and light chains, which are approximately 50 and 25 kDa, respectively. The most prominent ion series in this CAD experiment was the terminal b-ion series which allows N-terminal sequencing. Using this technique, we were able to confirm the sequence of up to seven N-terminal residues. Applications of this method for the identification of N-terminal pyroglutamic acid formation will be discussed. The method described could be used as a high-throughput method for the rapid N-terminal sequencing of IgG chains and for the detection of chemical modifications in the terminal residues.  相似文献

The kinetics of the formation and relaxation of transmembrane electric potential (Δψ) during the complete single turnover of CcO was studied in the bovine heart mitochondrial and the aa3-type Paracoccus denitrificans enzymes incorporated into proteoliposome membrane. The real-time Δψ kinetics was followed by the direct electrometry technique. The prompt oxidation of CcO and formation of the activated, oxidized (OH) state of the enzyme leaves the enzyme trapped in the open state that provides an internal leak for protons and thus facilitates dissipation of Δψ (τapp ≤ 0.5-0.8 s). By contrast, when the enzyme in the OH state is rapidly re-reduced by sequential electron delivery, Δψ dissipates much slower (τapp > 3 s). In P. denitrificans CcO proteoliposomes the accelerated Δψ dissipation is slowed down by a mutational block of the proton conductance through the D-, but not K-channel. We concluded that in contrast to the other intermediates the OH state of CcO is vulnerable to the elevated internal proton leak that proceeds via the D-channel.  相似文献
