The wild type (WT) of Scenedesmus obliquus and a mutant lacking chlorophyll b and the light-harvesting complexes (WT-LHC1) were synchronized by a light-dark regime. Both cultures contained the same type of carotenoids. However, concentrations and patterns of carotenoids were different during their synchronous life cycles. The concentration of total carotenoids followed more or less that of chlorophyll. The WT contained more carotenoids per cell mass, but slightly less per chlorophyll. It is discussed that part of the carotenoids of the mutant, lacking the peripheral antenna of PSII, might be located in the chlorophyll b-less apoprotein or in an enlarged core antenna of PSII. During the life cycle of Scenedesmus the carotenes are initially synthesized and most of the α-carotene is immediately oxidized to lutein which is inserted in the antennae systems of PSII and PSI. The further oxidation of lutein to loroxanthin seems to depend on both the change from dark to light, and on stages of the life cycle itself. Although the major part of β-carotene appears to be inserted in the reaction centers, a fraction of the total pool is rapidly converted to violaxanthin, following the onset of illumination. The conversion may serve to protect against photooxidation. Further conversion of violaxanthin to neoxanthin occurs to a greater extent in the mutant, WT-LHC1. The results demonstrate (1) the close connection between the carotenoid pattern and the modulation of the photosynthetic apparatus during the life cycle of Scenedesmus and (2) the flexibility of the organism in compensating for the absence of the light-harvesting complexes of photosystems II by adjusting the carotenoid distribution.  相似文献

The polar organelle, a structure associated with the flaggelar apparatus of bacteria, has been demonstrated in whole unstained cells of the photosynthetic bacterium Rhodopseudomonas palustris. It is subpolarly located close to the surface of the bacterial cell and has a round or ellipsoidal shap. It shows a strong ATPase activity which enables its cytochemical electron microscopical visualization.  相似文献

Trophic morphology plays a major role in niche diversification among the highly diverse fish fauna of coral reefs. It gives insight into feeding types and is useful in taxonomy, as it can be used to differentiate between species and to estimate phylogenetic relationships. The gobiid genus Gobiodon comprises obligate coral dwelling species that are widespread on Red Sea and Indo-Pacific reefs. Yet very little is known about their ecology and behaviour of feeding. In this study, viscerocranial and oral jaw morphology was investigated in seven species of Gobiodon that occur in the Red Sea. Osteological analyses were carried out on enzyme cleared and alizarin red stained specimens. Shape variation of the viscerocranium was analyzed using a landmark-based geometric morphometric method on digital scans of hand drawings and visualized in thin-plate splines. Shape was determined by placing landmarks on skeletal elements in their articulated state. Morphology of the dentary and premaxilla was examined in further detail after disarticulation. A combination of landmarks and semilandmarks was applied to digital photographs of the oral jaws. It was shown that orbit size influences viscerocranial morphology by delimiting the lower jaw articulation angle. The premaxilla differs both in its extent and direction of curvature and in the structure of the articular process. Form changes in the dentary are mainly caused by variation of curvature and depth. A significant relationship between oral jaw shape and the number of anterior rows of teeth was found. Morphological diversity of selected elements is used to generate hypotheses concerning the trophic biology and niche diversification of Gobiodon . Brief dietary and in situ behavioural observations were carried out to further develop and test hypotheses.  相似文献

An innovative Towed Trawl Simulator Sledge (TTSS) which has been tested in the field is described. The new instrument, which incorporates six water sampling bottles and video observation equipment, is designed to simulate the effects of the passage of otter trawl groundropes over seafloor sediments. The TTSS is towed from a surface vessel at speeds similar to those of commercial trawls and the water bottles, triggered simultaneously by electrical or acoustic signals, collect water samples in the plume of disturbed sediment behind the groundrope. The TTSS has been tested in the continental shelf fishing grounds of Heraklion Bay (Eastern Mediterranean) where quantitative measurements of the physical (sediment resuspension) and chemical (nutrient release) effects on surface sediments caused by the passage of otter trawl groundrope typical of that used in the Cretan fishery trawling were performed. The performance of the TTSS is also validated against an instrumented full-scale trawl. It is suggested that the TTSS should be used for the study and modelling of the effects of otter trawling and similar fishing processes on nutrient supply from seafloor sediments and the resulting ecosystem responses.  相似文献

The GTPase ADP-ribosylation factor related protein 1 (ARFRP1) controls the recruitment of proteins such as golgin-245 to the trans-Golgi. ARFRP1 is highly expressed in adipose tissues in which the insulin-sensitive glucose transporter GLUT4 is processed through the Golgi to a specialized endosomal compartment, the insulin-responsive storage compartment from which it is translocated to the plasma membrane in response to a stimulation of cells by insulin. In order to examine the role of ARFRP1 for GLUT4 targeting, subcellular distribution of GLUT4 was investigated in adipose tissue specific Arfrp1 knockout (Arfrp1ad−/−) mice. Immunohistochemical and ultrastructural studies of brown adipocytes demonstrated an abnormal trans-Golgi in Arfrp1ad−/− adipocytes. In addition, in Arfrp1ad−/− adipocytes GLUT4 protein accumulated at the plasma membrane rather than being sequestered in an intracellular compartment. A similar missorting of GLUT4 was produced by siRNA-mediated knockdown of Arfrp1 in 3T3-L1 adipocytes which was associated with significantly elevated uptake of deoxyglucose under basal conditions. Thus, Arfrp1 appears to be involved in sorting of GLUT4.  相似文献

The three-dimensional organization of the tear film, which is produced and drained by the different structures of the ocular adnexa, is essential for maintainance and protection of the ocular surface. This is facilitated by a class of large, highly glycosylated, hydrophilic glycoproteins, the mucins, which are usually expressed in association with a class of peptides having a well-defined, structurally conserved trefoil domain, the mammalian trefoil factor family (TFF) peptides. In this review, the latest information regarding mucin and TFF peptide function and regulation in the human lacrimal system, the tear film and the ocular surface is summarized with regard to mucous epithelia integrity, rheological and antimicrobial properties of the tear film and tear outflow, age-related changes and certain disease states such as dry eye, dacryostenosis and dacryolith formation.  相似文献

A motor protein complex of the bacterial flagellum, PomA/B from Vibrio alginolyticus, was reconstituted into liposomes and visualized by electron cryomicroscopy. PomA/B is a sodium channel, composed of two membrane proteins, PomA and PomB, and converts ion flux to the rotation of the flagellar motor. Escherichia coli and Salmonella have a homolog called MotA/B, which utilizes proton instead of sodium ion. PomB and MotB have a peptidoglycan-binding motif in their C-terminal region, and therefore PomA/B and MotA/B are regarded as the stator. Energy filtering electron cryomicroscopy enhanced the image contrast of the proteins reconstituted into liposomes and showed that two extramembrane domains with clearly different sizes stick out of the lipid bilayers on opposite sides. Image analysis combined with gold labeling and deletion of the peptidoglycan-binding motif revealed that the longer one, approximately 70 A long, is likely to correspond to the periplasmic domain, and the other, about half size, to the cytoplasmic domain.  相似文献

The Golgi apparatus breaks down at mitosis, resulting in the dispersal of Golgi-resident proteins. In NRK cells, however, subsets of both TGN38 and golgin-97, but not ManII and GM130, remained associated with the centrosome throughout the cell cycle. This centrosome association of TGN38 and golgin-97 was not disrupted by treatment with brefeldin A, additional inducers of retrograde trafficking and inhibitors of either kinases or protein phosphatases. Anchoring of the Golgi apparatus within the juxtanuclear region depends on microtubules; the association of TGN38 and golgin-97 subsets with the centrosome, however, was insensitive to nocodazole treatment. Drugs such as PDMP, which block Golgi dispersal both by nocodazole, despite microtubule depolymerization, and by inducers of retrograde trafficking, strengthened the microtubule-nucleating activity of the centrosome. These observations cumulatively suggest the centrosome is implicated in nucleation of the Golgi apparatus through interactions with Golgi-resident proteins, such as TGN38 and golgin-97.  相似文献
