Approximately 500 species of ascomycetous yeasts, including members of Candida and other anamorphic genera, were analyzed for extent of divergence in the variable D1/D2 domain of large subunit (26S) ribosomal DNA. Divergence in this domain is generally sufficient to resolve individual species, resulting in the prediction that 55 currently recognized taxa are synonyms of earlier described species. Phylogenetic relationships among the ascomycetous yeasts were analyzed from D1/D2 sequence divergence. For comparison, the phylogeny of selected members of the Saccharomyces clade was determined from 18S rDNA sequences. Species relationships were highly concordant between the D1/D2 and 18S trees when branches were statistically well supported.  相似文献

The microbial population dynamics on apples cv. Golden Delicious were analysed every 15 days between bud and harvest in a fully replicated experiment in northern Spain in 1994 and 1995. The total microbial populations varied with developmental stage, and with prevailing climatic conditions. The predominant mycroflora were the filamentous fungi Cladosporium and Alternaria spp. and white and pink yeasts. Other genera isolated included mainly species of Epicoccum, Fusarium and Acremonium. However, the most important post-harvest pathogens Penicillium expansum and Botrytis cinerea were seldom isolated from ripening apples. Maximum total filamentous fungal populations occurred after fruit set and during early ripening [2 × 104cfu (colony-forming units) g-1 approximately] while those of bacteria were maximum at bud stage (3.5 × 105and 3.0 × 104 cfu g-1 in 1994 and 1995 respectively). White yeasts were more numerous than pink yeasts. Endophytic infection of apple buds by Alternaria spp., responsible for core rot, was found in almost all bud tissue. By contrast, Cladosporium spp. were initially isolated later from 12.5–50% of tissue samples during blooming and fruit set. The impact of a four-spray fungicide regime during apple development significantly decreased the total filamentous fungal populations in both years, and that of Cladosporium spp. in 1994. However, bacterial populations were often higher on apples from fungicide-treated plots. Fungicide sprays decreased populations of Cladosporium, Alternaria and white yeasts for a maximum of up to 15–30 days after application. Fungicide application had little effect on endophytic infection of apples by Alternaria spp. between bud and harvest.  相似文献

AIMS: The aim of this study was to analyse the effect of osmotic stress on the biosynthesis of invertase enzyme in nonconventional yeasts. METHODS AND RESULTS: Invertase activities of the nonconventional yeast species belonging to Kluyveromyces, Schwanniomyces and Pichia genus were measured either in the presence or in the absence of various amounts of NaCl. The effect of hyperosmotic stress on the glucose consumption of Saccharomyces cerevisiae and Pichia anomala were also compared. Like S. cerevisiae, derepression of invertase synthesis in Kluyveromyces lactis, Schwanniomyces occidentalis and Pichia jadinii is inhibited by hyperosmotic stress. However, derepression of invertase synthesis in P. anomala is not affected by hyperosmotic stress. In addition, low levels of osmotic stress activated invertase synthesis three- to fourfold in P. anomala and K. lactis. CONCLUSIONS: This study shows that low levels of osmotic stress induces the invertase synthesis at very high levels in P. anomala and K. lactis. Glucose consumption was not influenced at significant levels by the hyperosmotic stress in P. anomala. SIGNIFICANCE AND IMPACT OF THE STUDY: This study shows the activation of invertase synthesis by low levels of osmotic stress in P. anomala and K. lactis.  相似文献

The reproductive stages of Dermocarpella gardneri and D. stellata, which have been reported only once, are described. Formation of baeocytes occurs by cellular divisions that are parallel to the substratum, followed by a series of anticlinal radial divisions. In some cases in D. gardneri, the superior cell, resulting from the first division parallel to the substratum, is liberated prior to radial divisions, and these probably represent the ‘macrogonidia’ originally described by Lemmermann for D. hemisphaerica. The baeocytes are released through a circular apical pore, which develops after the formation of a papilla that eventually dissolves to form a pore.  相似文献

To maintain homeostasis, every cell must constantly monitor its energy level and appropriately adjust energy, in the form of ATP, production rates based on metabolic demand. Continuous fulfillment of this energy demand depends on the ability of cells to sense, metabolize, and convert nutrients into chemical energy. Mitochondria are the main energy conversion sites for many cell types. Cellular metabolic states dictate the mitochondrial size, shape, function, and positioning. Mitochondrial shape varies from singular discrete organelles to interconnected reticular networks within cells. The morphological adaptations of mitochondria to metabolic cues are facilitated by the dynamic events categorized as transport, fusion, fission, and quality control. By changing their dynamics and strategic positioning within the cytoplasm, mitochondria carry out critical functions and also participate actively in inter-organelle cross-talk, assisting metabolite transfer, degradation, and biogenesis. Mitochondrial dynamics has become an active area of research because of its particular importance in cancer, metabolic diseases, and neurological disorders. In this review, we will highlight the molecular pathways involved in the regulation of mitochondrial dynamics and their roles in maintaining energy homeostasis.  相似文献

The currently accepted type species of the genusTrichosporon Behrend isT. beigelii. This species has formerly been regarded as identical toT. cutaneum. However, these fungi are now known to represent separate species with different ecology. The first species described inTrichosporon wasT. ovoides, an agent of human white piedra. A neotype strain is designated for this species, while a lectotype strain is indicated forT. cutaneum. The nameT. beigelii is considered as doubtful and consequently cannot be maintained.  相似文献

Histidine-containing phosphotransfer (HPt) proteins play an essential role in multistep histidine-aspartate phosphorelay signal transduction systems in prokaryotes and eukaryotes. The putative HPt protein in Schizosaccharomyces pombe, Mpr1p (also known as Spy1p), is a 295 amino acid protein that appears to be composed of more than one functional domain. The amino acid sequence of the N-terminal region of Mpr1p lacks homology to other known proteins, whereas the C-terminal domain is predicted to have structural similarity to the Ypd1p HPt protein from Saccharomyces cerevisiae. This study provides both in vitro and in vivo evidence that the C-terminal domain of Mpr1p indeed functions as an HPt protein in shuttling phosphoryl groups from one response regulator domain to another. Furthermore, we find that various deletions of the N-terminal region diminish both the phosphotransfer activity of Mpr1p and its affinity for response regulator domains, suggesting a possible role for the N-terminal domain in HPt-response regulator domain interactions.  相似文献

In the present study we describe the occurrence of fungi in 100 tap water and 16 groundwater samples from Slovenia. We used culture-dependent and culture-independent techniques. 28 fungal species belonging to 16 genera were isolated with selected culturing conditions, targeting human opportunistic yeasts and yeast-like fungi. Of special concern was the detection of Aureobasidium melanogenum, Exophiala dermatitidis, Rhinocladiella similis, Candida parapsilosis and Rhodotorula mucilaginosa. The DGGE analysis of ITS1 rDNA revealed from 6 to 16 bands hypothetically corresponding to different taxa, while pyrosequencing showed the presence of Aspergillus and Exophiala. According to the statistic machine learning methodology, the profile of fungi in water is determined by the concentration of calcium and magnesium ions and the presence of nitrate. Exophiala spp., C. parapsilosis and R. mucilaginosa are known as dominant contaminants of household appliances. It appears that they are transferred with water to dishwashers and washing machines, where they subsequently proliferate.  相似文献
