Abundance and Production of Riparian Trees in the Lowland Floodplain of the Queets River, Washington
Riparian zones associated with alluvial rivers are spatially dynamic, forming distinct vegetative mosaics that exhibit sharp
contrasts in structure and processes related to the underlying biophysical template. The productivity of riparian plants,
especially trees, influences streamside community characteristics as well as the forms and fluxes of organic matter to adjacent
streams – thereby strongly impacting patterns of channel morphology, water flow, sedimentation, and habitat in rivers. As
part of a comprehensive investigation of riparian dynamics in coastal rain forest rivers of the Pacific Northwest (USA), we
examined riparian tree abundance (density, basal area, and biomass) and rates of production (basal area growth [BAI] and bole
wood biomass increase [P]) of seven common species – red alder (Alnus rubra), Sitka spruce (Picea sitchensis), bigleaf maple (Acer macrophyllum), western hemlock (Tsuga heterophylla), black cottonwood (Populus trichocarpa), vine maple (Acer circinatum) and willow (Salix spp.) – in the lowland floodplain of the Queets River (Olympic National Park), Washington. Measurements were made annually
for three years (1999 – 2001) in 16 permanent plots on three biophysical templates that formed a toposequence – active floodplain,
young terrace and mature terrace. Stem density was highest in the active floodplain (∼27,000 stems/ ha), decreasing in the
young terrace (∼2,700 stems /ha) and the mature terrace (∼500 stems/ha). Basal area and total stem biomass were lowest in
the active floodplain (∼16 m2/ha and ∼18 Mg dry weight/ha, respectively) and higher on the young terrace (∼32 m2/ha and ∼134
Mg dry weight/ha) and on the mature terrace (∼69 m2/ha and ∼540 Mg dry weight /ha). Total plot-scale BAI was not significantly
different among the physical templates with mean values ranging from approximately 1.4 (low terrace) to approximately 2.8
m2/ha/y (active floodplain). In contrast, P was significantly higher on the mature terrace (10.3 Mg/ha) than the active floodplain
(3.2 Mg/ha) but there was no significant difference between young terrace (6.5 Mg/ha) and mature terrace. For the entire Queets
River floodplain (57 km2 over 77 km of river length), the mature terrace contributed 81% of the total annual production (28,764 Mg) whereas the active
floodplain and young terrace accounted only for 5 and 14%, respectively. Overall, we show that riparian trees grow quickly
in this coastal Pacific Northwest system and that the older riparian forests on mature terraces are the main contributors
to stem production at the plot and floodplain scales for at least 350 years after stand initiation. This suggests that, in
combination with the rapid lateral migrations of many alluvial rivers, the older riparian forests on those terraces are important
and sustained sources of organic matter (especially large woody debris, LWD) that, over decades to centuries, shape the character
of coastal rivers in the Pacific Northwest. 相似文献

In citrus, damage produced by salinity is mostly due to toxic ion accumulation, since this salt-sensitive crop adjusts osmotically with high efficiency. In spite of this observation, the putative role of sugars as osmolites under salinity remains unknown. In this work, we have studied carbohydrate contents (total hexoses, sucrose and starch) in leaves and roots of citrus grown under increasing salinity. The experimental system was characterized through the analyses of several parameters known to be strongly affected by salinity in citrus, such as chloride accumulation, photosynthetic rate, ethylene production and leaf abscission. Three-year-old plants of the Clementina de Nules cultivar grafted on Carrizo citrange rootstock were watered with three different levels of salinity (NaCl was added to the watering solutions to achieve final concentrations of 30, 60 and 90 mM). Data indicate that salt stress caused an accumulation of chloride ions in a way proportional to the external increase in NaCl. The adverse conditions reduced CO2 assimilation, increased ethylene production and triggered abscission of the injured leaves. Data also show that salinity induced progressive depletions of carbohydrates in leaves and roots of citrus plants. This observation clearly indicates that sugar accumulation is not a main component of the osmotic adjustment machinery in citrus. 相似文献

Several methods have been described to enhance antibody-dependent cellular cytotoxicity (ADCC) using different host cells that produce antibody with reduced levels of fucose on their carbohydrates. We compared the suitability of these methods for the serum-free fed-batch production of antibody for clinical trials and commercial uses. Recombinant anti-human CD20 chimeric IgG1-producing clones were established from host-cells that have been shown to produce more than 90% fucose-negative antibody. The cell lines were a FUT8 (alpha-1,6-fucosyltransferase) knockout Chinese hamster ovary (CHO) cell line, Ms704, and two Lens culinaris agglutinin (LCA)-resistant cell lines, one derived from a variant CHO line, Lec13 and the other from a rat hybridoma cell line, YB2/0. The amount of fucose-negative antibody produced by Lec13 and YB2/0 significantly decreased with the culture. The increase in fucosylation was due to remaining synthesis of GDP-fucose via de novo pathway for the CHO line and the elevation of FUT8 expression by the YB2/0 cells. In contrast, Ms704 cells stably produced fucose-negative antibody with a consistent carbohydrate structure until the end of the culture. The productivity of the Ms704 cells reached 1.76 g/L with a specific production rate (SPR) of 29 pg/cell/day for 17 days in serum-free fed-batch culture using a 1 L spinner bioreactor. Our results demonstrate that FUT8 knockout has the essential characteristics of host cells for robust manufacture of fucose-negative therapeutic antibodies with enhanced ADCC. 相似文献

Asparaginase isozyme II from Escherichia coli is a popular enzyme that has been used as a therapeutic agent against acute lymphoblastic leukemia. Here, fusion tag systems consisting of the pelB signal sequence and various lengths of repeated aspartate tags were devised to highly express and to release active asparaginase isozyme II extracellularly in E. coli. Among several constructs, recombinant asparaginase isozyme II fused with the pelB signal sequence and five aspartate tag was secreted efficiently into culture medium at 34.6 U/mg cell of specific activity. By batch fermentation, recombinant E. coli produced 40.8 U/ml asparaginase isozyme II in the medium. In addition, deletion of the gspDE gene reduced extracellular production of asparaginase isozyme II, indicating that secretion of recombinant asparaginase isozyme II was partially ascribed to the recognition by the general secretion machinery. Periphyton (epilithon) gross primary production (GPP) was estimated using the DCMU-fluorescence method in the Yenisei River.
In the unshaded littoral zone, chlorophyll a concentration (Chl a) and GPP value varied from 0.83 to 973.74 mg m−2and 2–304,425 O2 m−2 day−1 (0.64–95 133 mg C m−2 day−1), respectively. Positive significant correlation (r = 0.8) between daily GPP and periphyton Chl a was found. Average ratio GPP:Chl a for periphyton was 36.36 mg C mg Chl a m−2 day−1. The obtained GPP values for the Yenisei River have a high significant correlation with values predicted by a conventional
empirical model for stream periphyton. We concluded that the DCMU-fluorescence method can be successfully used for measuring
of gross primary production of stream phytoperiphyton at least as another useful tool for such studies. 相似文献

Aim Propagule size and output are critical for the ability of a plant species to colonize new environments. If invasive species have a greater reproductive output than native species (via more and/or larger seeds), then they will have a greater dispersal and establishment ability. Previous comparisons within plant genera, families or environments have conflicted over the differences in reproductive traits between native and invasive species. We went beyond a genus‐, family‐ or habitat‐specific approach and analysed data for plant reproductive traits from the global literature, to investigate whether: (1) seed mass and production differ between the original and introduced ranges of invasive species; (2) seed mass and production differ between invasives and natives; and (3) invasives produce more seeds per unit seed mass than natives. Location Global. Methods We combined an existing data set of native plant reproductive data with a new data compilation for invasive species. We used t‐tests to compare original and introduced range populations, two‐way ANOVAs to compare natives and invasives, and an ANCOVA to examine the relationship between seed mass and production for natives and invasives. The ANCOVA was performed again incorporating phylogenetically independent contrasts to overcome any phylogenetic bias in the data sets. Results Neither seed mass nor seed production of invasive species differed between their introduced and original ranges. We found no significant difference in seed mass between invasives and natives after growth form had been accounted for. Seed production was greater for invasive species overall and within herb and woody growth forms. For a given seed mass, invasive species produced 6.7‐fold (all species), 6.9‐fold (herbs only) and 26.1‐fold (woody species only) more seeds per individual per year than native species. The phylogenetic ANCOVA verified that this trend did not appear to be influenced by phylogenetic bias within either data set. Main conclusions This study provides the first global examination of both seed mass and production traits in native and invasive species. Invasive species express a strategy of greater seed production both overall and per unit seed mass compared with natives. The consequent increased likelihood of establishment from long‐distance seed dispersal may significantly contribute to the invasiveness of many exotic species. 相似文献

The microsporida Nosema gastroideae and N. equestris, which are highly pathogenic for Leptinotarsa, have been successfully produced in some other chrysomelid species, Gastrophysa polygoni and G. viridula. As the principal target host, Leptinotarsa is very susceptible to these pathogens, and death occurs before massive sporulation by the microsporidia. By contrast, the infected larvae of G. polygoni or G. viridula are able to develop until the adult stage when most of the tissues become filled with spores. In addition, the larvae and adults of these species can be reared in the laboratory on Polygonum aviculare and Rumex obtusifolius. These plants have longer vegetative periods and are better sources of food than potato leaves. In both species of Gastrophysa the yields of spores related to unit weight were about five times higher than in Leptinotarsa. In the adults of G. viridula there was up to 4.8 × 106 spores mg?1 body weight of N. gastroideae, or 9.1 × 106 spores mg?1 of N. equestris. The higher content of microsporidian spores facilitates their purification and isolation. 相似文献

大丽轮枝菌可侵染棉花、马铃薯、番茄等660余种寄主植物,引致黄萎病,造成严重的经济损失。为了深入了解大丽轮枝菌的致病机制,本研究在前期棉花提取物诱导大丽轮枝菌转录组分析的基础上,选择上调差异表达的线粒体甘油-3-磷酸脱氢酶基因VdGut2(VD592_6958_Chr4)和非差异表达的胞质甘油-3-磷酸脱氢酶基因VdGpd(VD592_10256_Chr2),进行了功能分析。结果表明,两个VdGut2敲除突变体菌株的产孢量较野生型菌株分别下降了32%和41%,病情指数分别下降了70%和51%,菌落生长速率也显著下降;VdGpd过表达菌株产孢量和病情指数较野生型菌株显著下降,但在甘油为唯一碳源的培养基上其菌落生长速率显著上升。因此,VdGut2促进了大丽轮枝菌分生孢子的形成、碳源的利用以及对寄主植物的致病过程,VdGpd抑制了大丽轮枝菌分生孢子的形成和对寄主植物的致病力,却促进了甘油的代谢。 相似文献
