Summary Homonuclear 3D1H NOESY-TOCSY and 3D1H ROESY-TOCSY experiments were used to resolve and assign nuclear Overhauser effect (NOE) cross peaks between the water signal and individual polypeptide proton resonances in H2O solutions of the basic pancreatic trypsin inhibitor. Combined with a novel, robust water-suppression technique, positive and negative intermolecular NOEs were detected at 4°C. The observation of positive NOEs between water protons and protein protons enables more precise estimates of the very short residence times of the water molecules in the hydration sites on the protein surface. 相似文献
Background: Deforestation processes and species extinction on islands have made them the focus of substantial efforts to create and strengthen local environmental organisations, establish new protected areas and improve natural resource management. In Madagascar, despite promising new policies, the flora and fauna are under serious threat. More than 80% of the island shows a significant loss of natural plant cover. Current levels of diversity reflect natural disturbance regimes, with fire representing one of the more important factors.
Aims: We compared two new protected areas (Ibity and Itremo) with different environmental and management contexts to identify the roles and impacts of the environmental conditions and the different types of management applied by their respective local communities on vegetation. Both areas have extensive stands of tapia (Uapaca bojeri) woodland.
Methods: Herbaceous vegetation surveys were carried out enumerating 16 1-m² quadrats in each of 10 sites on Ibity and seven on Itremo to characterise plant community composition and structure, complemented by tree and shrub surveys within three 40 × 40 m quadrats at each site.
Results: Floristic richness was higher at Ibity. Populations of U. bojeri were better conserved at Itremo.
Conclusions: While the total exclusion of fire is impracticable, careful management should enable the maintenance of natural conditions and at least lead to a reduction in the impacts on the structure of tapia woodland vegetation. 相似文献
Severe, stand-replacing fires affect large areas of northern temperate and boreal forests, potentially modifying ecosystem function for decades after their occurrence. Because these fires occur over large extents, and in areas where plant production is limited by nitrogen (N) availability, the effect of fire on N cycling may be important for long-term ecosystem productivity. In this article, we review what is known about postfire N cycling in northern temperate and boreal forests experiencing stand-replacing fires. We then build upon existing literature to identify the most important mechanisms that control postfire N availability in systems experiencing severe, stand-replacing fires compared with fires of lower severity. These mechanisms include changes in abiotic conditions caused by the opening of the canopy (for example, decreased LAI, increased solar radiation), changes in ground layer quantity and quality (for example, nutrient release, permafrost levels), and postfire plant and microbial adaptations affecting N fixation and N uptake (for example, serotiny, germination cues). Based on the available literature, these mechanisms appear to affect N inputs, internal N cycling, and N outputs in various ways, indicating that severe fire systems are variable across time and space as a result of complex interactions between postfire abiotic and biotic factors. Future experimental work should be focused on understanding these mechanisms and their variability across the landscape. 相似文献
This case study evaluates the effects of a 4.7 mg deslorelin acetate implant on one male olive baboon (Papio anubis). The effective use of prescribed fire in biodiversity conservation is currently inhibited by a limited understanding of fire effects on ecosystem processes such as pollination. Orchids inhabiting fire‐prone landscapes are likely to be particularly sensitive because they often exhibit highly specialized pollination systems and provide no reward to pollinators, making them dependent on co‐flowering heterospecifics to attract and support pollinators. We investigated the hypothesis that fire‐driven changes in the local abundance of rewarding heterospecific flowers influence pollination in two rewardless Australian orchid species, Diuris maculata sensu lato and Caladenia tentaculata. Diuris maculata s.l. is thought to achieve pollination by mimicking papilionoid Fabaceae flowers. Caladenia tentaculata attracts male thynnine wasps through sexual deceit, and these wasps forage on the open‐access flowers of other taxa. We used a space‐for‐time substitution design with sites in different stages of post‐fire succession where we recorded capsule set in D. maculata s.l., pollinator visitation to C. tentaculata, the floral abundance of rewarding heterospecifics and abiotic conditions. Many rewarding taxa responded to fire age, but there was only weak evidence that capsule set in D. maculata s.l. was positively related to the local floral abundance of rewarding species. There was evidence of an overriding effect of rainfall on capsule set that may have obscured effects of the floral community. Visitation to C. tentaculata was not positively associated with any rewarding heterospecifics, and was negatively associated with rewarding Burchardia umbellata. Our preliminary findings highlight the need to account for multiple factors when trying to detect fire effects on pollination. 相似文献
Question: Do case studies from silvicultural and restoration studies and applied conservation management in second‐growth Pinus palustris stands provide unique insights for conservation models? Methods: A review of management paradigms that conserve the high biological diversity and rare species, drawn from characteristics in both second‐growth and old‐growth stands, is presented for fire‐maintained Pinus palustris (longleaf pine) forests. Results: A common assumption that old‐growth stands provide the primary information for the development of conservation management strategies de‐emphasizes lessons learned from second‐growth and restoration studies. Primary conservation management goals for the Pinus palustris ecosystem include the perpetual regeneration of the fire‐maintained forest and conservation of the characteristically high biological diversity and rare species. Several attributes, such as a sustained population of Picoides borealis (red‐cockaded woodpecker), Aristida stricta (wiregrass)‐dominated ground cover, and undisturbed upland‐wetland ecotones, can predict a diverse and ecologically functional ecosystem. Such indicators are linked to critical structural and functional features of the system and reflect previous land management histories that suggest sustainable approaches. Conclusions: A traditional definition of ‘old‐growth’ relying on overstorey may be limited in describing important features of healthy, diverse Pinus palustris ecosystems. Some characteristics are significantly more important for maintenance of diversity than age of the trees and these features may be present in old‐ or second‐growth forests. We advocate that the management history, structural characteristics and landscape context of stands that harbour desirable conservation attributes (red‐cockaded woodpeckers, wiregrass, gopher tortoises and undisturbed upland‐wetland ecotones) can be used as indicators to identify important conservation and forest management principles. 相似文献
1. Predator interactions are often driven by size differences, with larger predators consuming smaller ones. Cannibalism is widespread among predators, and can be more intense than antagonistic interactions between different predator species. 2. In a series of greenhouse experiments and a field experiment, we investigated the role of size structure in shaping the impacts of a top predator (the wandering spider Cheiracanthium mildei L. Koch) on an insect herbivore in vineyards of Napa County, California. 3. In greenhouse cages, survival of small C. mildei was lower when large C. mildei were present, and the combined effects of small and large C. mildei on leafhopper numbers and grape leaf damage were non‐additive. The frequency of cannibalism in C. mildei did not change when the abundance of leafhoppers was manipulated. However, in the field experiment small and large C. mildei combined additively to reduce leafhopper numbers. 4. Results show that cannibalism has the potential to dampen the cascading effects of C. mildei in a vineyard food web, but that it may be less important than other mortality factors in the field. Nonetheless, C. mildei substantially impacted leafhoppers in both the greenhouse and the field, suggesting that mortality caused by cannibalism or other factors may not compromise its overall predatory impacts. 相似文献
Mesenchymal stem cells(MSCs),derived from adult tissues,are multipotent progenitor cells,which hold greatpromise for regenerative medicine.Recent studies have shown that MSCs are immunosuppressive in vivo and in vitro inboth animals and humans.However,the mechanisms that govern these immune modulatory functions of MSCs remainlargely elusive.Some studies with bulk populations of MSCs indicated that soluble factors such as PGE2 and TGFβ areimportant,while others support a role for cell-cell contact.In this study,we intended to clarify these issues by examin-ing immunosuppressive effects of cloned MSCs.We derived MSC clones from mouse bone marrow and showed thatthe majority of these clones were able to differentiate into adipocytes and osteoblast-like cells.Importantly,cells fromthese clones exhibited strong inhibitory effects on TCR activation-induced T cell proliferation in vitro,and injection ofa small number of these cells promoted the survival of allogeneic skin grafts in mice.Conditioned medium from MSCcultures showed some inhibitory effect on anti-CD3 induced lymphocyte proliferation independent of PGE2 and TGFβ.In comparison,direct co-culture of MSCs with stimulated lymphocytes resulted in much stronger immunosuppressiveeffect.Interestingly,the suppression was bi-directional,as MSC proliferation was also reduced in the presence of lym-phocytes.Taking together,our findings with cloned MSCs demonstrate that these cells exert their immunosuppressiveeffects through both soluble factor(s)and cell-cell contact,and that lymphocytes and MSCs are mutually inhibitory ontheir respective proliferation. 相似文献
Species responses to climate change may be influenced by changes in available habitat, as well as population processes, species interactions and interactions between demographic and landscape dynamics. Current methods for assessing these responses fail to provide an integrated view of these influences because they deal with habitat change or population dynamics, but rarely both. In this study, we linked a time series of habitat suitability models with spatially explicit stochastic population models to explore factors that influence the viability of plant species populations under stable and changing climate scenarios in South African fynbos, a global biodiversity hot spot. Results indicate that complex interactions between life history, disturbance regime and distribution pattern mediate species extinction risks under climate change. Our novel mechanistic approach allows more complete and direct appraisal of future biotic responses than do static bioclimatic habitat modelling approaches, and will ultimately support development of more effective conservation strategies to mitigate biodiversity losses due to climate change. 相似文献
