Summary In human red cells homozygous for hemoglobin C (CC), cell swelling and acid pH increase K efflux and net K loss in the presence of ouabain (0.1mm) and bumetanide. We report herein, that K influx is also dependent on cell volume in CC cells: cell swelling induces a marked increase in the maximal rate (from 6 to 18 mmol/liter cell × hr) and in the affinity for external K (from 77±16mm to 28±3mm) of K influx. When the external K concentration is varied from 0 to 140mm, K efflux from CC and normal control cells is unaffected. Thus, K/K exchange is not a major component of this K movement. K transport through the pathway of CC cells is dependent on the presence of chloride or bromide; substitution with nitrate, acetate or thiocyanate inhibits the volume- and pH-dependent K efflux. When CC cells are separated according to density, a sizable volume-dependent component of K efflux can be identified in all the fractions and is the most active in the least dense fraction. N-ethylmaleimide (NEM) markedly stimulates K efflux from CC cells in chloride but not in nitrate media, and this effect is present in all the fractions of CC cells separated according to density. The persistence of this transport system in denser CC cells suggests that not only cell age, but also the presence of the positively charged C hemoglobin is an important determinant of the activity of this system. These data also indicate that the K transport pathway of CC cells is not an electrodiffusional process and is coupled to chloride.  相似文献

The larvae of the dipteran insect, Chironomus ramosus, found in Shillong, India, contain eleven (11) hemoglobin (Hb) components of which three are monomers (CI, CIV and CVI) and seven are dimers (CIII, CV, CVII, CVIII, CIX, CX and CXI), while one (CII) exists in both monomeric and dimeric states.Four monomeric components were isolated, purified and partially characterized. The N-terminal amino acids were determined and showed glycine for CI and leucine for the other components (CII, CIV and CVI).Three hemoglobin components were found to be present in all stages of larval development, except the first instar larvae. Some Hb components were synthesized in a particular instar, as revealed by electrophoretic appearance.Electrophoretic mobilities of seven components and N-terminal amino acid residues of two components of Hb were similar in both Chironomus ramosus and Chironomus thummi thummi.  相似文献

Hemoglobin concentrations ofChironomus cf.plumosus larvae were measured in two different habitats of the same pond. Larger larvae have higher hemoglobin concentrations than small larvae. There is strong indication that the animals of poorly oxygenated deep water, have higher hemoglobin concentrations than the animals from the well-oxygenated littoral zone.  相似文献

A recombinant double mutant of hemoglobin (Hb), E6V/L88A(beta), was constructed to study the strength of the primary hydrophobic interaction in the gelation of sickle Hb, i.e., that between the mutant Val-6(beta) of one tetramer and the hydrophobic region between Phe-85(beta) and Leu-88(beta) on an adjacent tetramer. Thus, a construct encoding the donor Val-6(beta) of the expressed recombinant HbS and a second mutation encoding an Ala in place of Leu-88(beta) was assembled. The doubly mutated beta-globin gene was expressed in yeast together with the normal human alpha-chain, which is on the same plasmid, to produce a soluble Hb tetramer. Characterizations of the Hb double mutant by mass spectrometry, by HPLC, and by peptide mapping of tryptic digests of the mutant beta-chain were consistent with the desired mutations. The absorption spectra in the visible and the ultraviolet regions were practically superimposable for the recombinant Hb and the natural Hb purified from human red cells. Circular dichroism studies on the overall structure of the recombinant Hb double mutant and the recombinant single mutant, HbS, showed that both were correctly folded. Functional studies on the recombinant double mutant indicated that it was fully cooperative. However, its gelation concentration was significantly higher than that of either recombinant or natural sickle Hb, indicating that the strength of the interaction in this important donor-acceptor region in sickle Hb was considerably reduced even with such a conservative hydrophobic mutation.  相似文献

Numerous studies have demonstrated that several diseases and stress conditions are associated with changes in the levels of zinc in the blood plasma and cellular elements. In this research the association between serum zinc concentrations and other hematic parameters of diagnostic interest has been evaluated. Quantitative determinations of zinc, total plasmatic proteins, albumin, hemoglobin and calculation of mean corpuscular volume were performed on blood samples from 58 males aged 20–61 years. Concentrations measured in our sample are comparable with reference values. Statistically significant correlation coefficients were found between age and albumin (r = - 0.562, P < 0.001), serum zinc and albumin (r = 0.328, P < 0.05), serum zinc and hemoglobin (r = 0.291, P < 0.05), and total plasmatic proteins and albumin (r = 0.463, P < 0.001). These correlation coefficients were significant even after adjustment for age effect. The determination of serum zinc concentration may be useful in the assessment of clinical scenarios. Particularly, it may provide additional information for the diagnosis of specific pathologies, such as hepatic malfunctions. It could also be useful in the identification of different stages of anemia.  相似文献

We have used [2-13C]d-glucose and carbon-13 nuclear magnetic resonance (NMR) spectroscopy to investigate metabolic fluxes through the major pathways of glucose metabolism in intact human erythrocytes and to determine the interactions among these pathways under conditions that perturb metabolism. Using the method described, we have been able to measure fluxes through the pentose phosphate pathway, phosphofructokinase, the 2,3-diphosphoglycerate bypass, and phosphoglycerate kinase, as well as glucose uptake, concurrently and in a single experiment. We have measured these fluxes in normal human erythrocytes under the following conditions: (1) fully oxygenated; (2) treated with methylene blue; and (3) deoxygenated. This method makes it possible to monitor various metabolic effects of stresses in normal and pathological states. Not only has 13C-NMR spectroscopy proved to be a useful method for measuring in vivo flux through the pentose phosphate pathway, but it has also provided additional information about the cycling of metabolites through the non-oxidative portion of the pentose phosphate pathway. Our evidence from experiments with [1-13C]-, [2-13C]-, and [3-13C]d-glucoses indicates that there is an observable reverse flux of fructose 6-phosphate through the reactions catalyzed by transketolase and transaldolase, even in the presence of a net flux through the pentose phosphate pathway.  相似文献

摘要 目的:探讨血糖控制水平与2型糖尿病合并根尖周病的相关性。方法:回顾性选择2018年9月~2022年9月我院收治的300例2型糖尿病患者临床资料,根据是否发生根尖周病将患者分为根尖周病组(45例)和对照组(255例),根据糖化血红蛋白(HbA1c)分为血糖控制不佳组(HbA1c≥6.5%,105例),血糖控制良好组(HbA1c<6.5%,195例)。多因素Logistic回归分析2型糖尿病合并根尖周病的相关因素,受试者工作特征曲线(ROC)分析HbA1c诊断2型糖尿病合并根尖周病的价值。结果:血糖控制不佳组根尖周病发病率(28.57% vs 7.69%)、PAI评分(2.66±0.41分 vs1.24±0.26分)均高于血糖控制良好组(P<0.001)。根尖周病组吸烟史比例、牙周疾病比例、血糖控制不佳比例、FPG高于对照组(P<0.001),2型糖尿病病程长于对照组(P<0.001),服用二甲双胍比例低于对照组(P<0.01)。多因素Logistic回归结果显示血糖控制不佳、吸烟史是2型糖尿病患者合并根尖周病的危险因素(P<0.001,P<0.01),服用二甲双胍是保护因素(P<0.001)。HbA1c诊断2型糖尿病患者合并根尖周病的曲线下面积为0.659(95%CI:0.650-0.756, P<0.01),灵敏度为73.33%,特异度为62.75%,约登指数为0.3608。结论:血糖控制不佳可能与2型糖尿病患者根尖周病的发生有关,临床应积极控制血糖水平以阻止根尖周病的发生。  相似文献

本文报道49例Hb G Coushatta在我国西北部的分布情况。新疆和甘肃的发生率为0.57‰和0.43‰,高于陕西的发生率0.07‰。维吾尔、哈萨克、柯尔克孜和回族的发生率为0.66‰—1.74‰,高于汉族的发生率0.45‰。Hb G Coushatta基因流呈由北向南和向西漂移和特点,结合国内外有关资料进行分析,这种变异体起源于东北亚地区,为古代游牧民族的遗传标志。  相似文献

A true hemoglobin (Hb) was shown to be present in the root nodules of Alnus glutinosa L. After purification by gel filtration and ion exchange, the Hb formed a stable complex with oxygen. This oxygen complex could then be converted to carboxyhemoglobin by treatment with CO. Optical absorption spectra typical of Hb were observed. The molecular weight was estimated to be 15 100 by gel filtration, and 18 300 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The Hb was largely insoluble when the initial homogenization was done in the absence of a detergent. Under these conditions much of the Hb appears to be associated with clusters of Frankia , the nitrogen-fixing actinomycete that infects plant cells within the nodules. The exact localization of the Hb in vivo is uncertain. The relatively low average concentration of Hb in Alnus nodules suggests that it is either confined to a relatively small fraction of total nodule volume, or has a function other than facilitation of O2 transport.  相似文献

On-line NAD(P)H fluorescence and culture redox potential (CRP) measurements were utilized to investigate the role of Vitreoscilla hemoglobin (VHb) in perturbing oxygen metabolism of microaerobic Escherichia coli Batch cultures of a VHb-synthesizing E. coli strain and the iso-genic control under fully aerated conditions were subject to several high/low oxygen transitions, and the NAD(P)H fluorescence and CRP were monitored during these passages. The presence of VHb decreased the rate of net NAD(P)H generation by 2.4-fold under diminishing oxygen tension. In the absence of aeration, the strain producing VHb maintained a steady NAD(P)H level 1.8-fold less than that of the control, indicating that the presence of VHb keeps E. coli in a more oxidized state under oxygen-limited conditions. Estimated from CRP, the oxygen uptake rates near anoxia were 25% higher for cells with VHb than those without. These results suggest that VHb-expressing cells have a higher microaerobic electron transport chain turnover rate. To examine how NAD(P)H utilization of VHb-expressing cells responds to rapidly changing oxygen tension, which is common in large-scale fermentations, we pulsed air intermittently into a cell suspension and recorded the fluorescence response to the imposed dissolved oxygen (DO) fluctuation. Relative to the control, cells containing VHb had a sluggish fluorescence response to sudden changes of oxygen tension, suggesting that VHb buffers intracellular redox perturbations caused by extracellular DO fluctuations.(c) John Wiley & Sons, Inc.  相似文献
