The central cell characterizes the angiosperm female gametophyte (embryo sac or megagametophyte) in that it directly participates in “double fertilization” to initiate endosperm development, a feature distinguishing angiosperm from all other plant taxa. Polygonum‐type central cell is a binucleate cell that, upon fertilization with one of the two sperm cells, forms triploid endosperm to nourish embryo development. Although the formation and the structure of central cell have well been elucidated, the molecular mechanisms for its specification and development remain largely unknown. The central cell plays a critical role in pollen tube guidance during pollination and in endosperm initiation after fertilization. Recently, a group of mutants affecting specific steps of central cell development and function have been identified, providing some clues in understanding these questions. Several studies suggest that females mate multiply so that they can preferentially fertilize eggs with the sperm of genetically more compatible males. Unrelated males are expected to be genetically more compatible with a female than her close relatives. We tested whether black field crickets, Teleogryllus commodus, can bias sperm usage toward unrelated males by comparing egg hatching success of females mated to two of their siblings (SS), two sibling males unrelated to the female (NN) or to one unrelated male and a sibling male (NS or SN). Egg hatching success was highly repeatable. Hatching success varied significantly among females of the three mating types (P = 0.011, n = 245 females). The estimated mean hatching success of 36.8% for SS females was significantly less that the 43.4% of NN females, indicating an effect of inbreeding on hatching success. If females preferentially use the sperm of a less closely related male, the hatching success of NS/SN females should be closer to 43.4% than 36.8%. It was, in fact, only 34.9%. This does not differ significantly from the value expected if the two males contributed an equal amount of sperm that was then used randomly. Although polyandry may confer indirect genetic benefits, our results provide no evidence that female T. commodus gain these benefits by biasing paternity toward genetically more compatible males through postcopulatory mechanisms. 相似文献

A growing number of studies highlight the nontransitive properties of ejaculates when they are in competition to fertilize a female's eggs. Increasingly, these studies suggest that postcopulatory processes act as a filter against sperm from closely related males or those with similar genotypes, limiting the deleterious effects of inbreeding on offspring fitness. We investigated the potential for such postcopulatory mechanisms of inbreeding avoidance in the guppy (Poecilia reticulata), a promiscuous livebearing fish. We used artificial insemination as a method of delivering to a female the combined ejaculates from a first cousin (relatedness coefficient r = 0.125) and an unrelated male. This method of sperm delivery controls behavioral processes of pre- and postcopulatory female choice, which can bias paternity toward unrelated males. Our genetic analysis revealed no effect of parental relatedness on paternity outcomes. The observed mean paternity share for related males (0.47) and associated variance did not differ significantly from an expected binomial distribution that assumes no biased use of sperm with respect to relatedness (0.5). Although our data provide no evidence for postcopulatory mechanisms of inbreeding avoidance, the ability of female guppies to influence ejaculate transfer and retention offers an alternative and easily testable mechanism of inbreeding avoidance in this species. 相似文献

Biofilm development in urinary tract catheters is an often underestimated problem. However, this form of infection leads to high mortality rates and causes significant costs in health care. Therefore, it is important to analyze these biofilms and establish avoiding strategies. In this study a continuous flow-through system for the cultivation of biofilms under catheter-associated urinary tract infection conditions was established and validated. The in vitro urinary tract catheter system implies the composition of urine (artificial urine medium), the mean volume of urine of adults (1 mL min-1), the frequently used silicone catheter (foley silicon catheter) as well as the infection with uropathogenic microorganisms like Pseudomonas aeruginosa. Three clinical isolates from urine of catheterized patients were chosen due to their ability to form biofilms, their mobility and their cell surface hydrophobicity. As reference strain P. aeruginosa PA14 has been used. Characteristic parameters as biofilm thickness, specific biofilm growth rate and substrate consumption were observed. Biofilm thicknesses varied from 105 ± 16 μm up to 246 ± 67 μm for the different isolates. The specific biofilm growth rate could be determined with a non invasive optical biomass sensor. This sensor allows online monitoring of the biofilm growth in the progress of the cultivation. 相似文献

目的探讨早产儿肠道微生态变化及其与胎龄、出生体质量的关系。方法选取2018年5月至2020年5月我院收治的80例早产儿作为早产组,同期收治的80例足月新生儿作为对照组。收集入组新生儿出生后3 d、3周的粪便,比较2组研究对象粪便标本中细菌丰富度和Shannon Wiener指数;根据早产组胎龄和出生体质量不同分组,分析胎龄、出生体质量与肠道微生态变化的关系。结果(1)出生3 d,早产组新生儿粪便标本的DGGE图谱条带数、Shannon Wiener指数均显著低于对照组(t=3.179、3.521,均P<0.05);(2)出生3 d,胎龄34+1~36+6周组新生儿粪便标本的DGGE图谱条带数、Shannon Wiener指数显著高于28~30周组、30+1~32周组和32+1~34周组(q=9.653、6.476、4.848和8.796、8.008、6.277,均P<0.05);(3)出生3 d,体质量>2 000 g组新生儿粪便标本的的DGGE图谱条带数、Shannon Wiener指数显著高于≤1 500 g组(q=5.601和4.593,均P<0.05);(4)不同出生体质量、胎龄的早产儿,出生3周粪便标本的DGGE图谱条带数、Shannon Wiener指数相比差异无统计学意义(F=1.577和2.326,均P>0.05)。结论与足月新生儿相比,早产儿出生后细菌定植出现延迟且多样性差,胎龄越小、出生体质量越低的早产儿的肠道菌群的多样性更低、定植延迟的可能性更大,出生3周后其肠道微生态逐渐恢复正常。 相似文献

This study was designed to localize transforming growth factor alpha (TGF-) and epidermal growth factor receptor (EGFR) expression in the developing human gastrointestinal tract and pancreas. Immunohistochemical techniques using specific antibodies against human TGF- and EGFR were performed on digestive tissues of fetuses from 9 to 10 to 24 weeks of gestation, children and adults. In fetuses, TGF- and EGFR proteins were expressed in all epithelial tissues studied with a good correlation and from an age as early as 9 to 10 weeks of gestation, except for TGF- in the esophagus. The strongest TGF- immunostaining was noted in the stomach and the proximal colon. Unexpectedly, immunoreactive gut endocrine cells were observed with the two antibodies used. Relatively numerous in fetuses, they decreased in number with age and were rare in adults particularly along the colon. Enteroglucagon-secreting cells were shown to express TGF- while some gastrin, somatostatin and pancreatic glucagon cells were immunostained with EGFR antibodies. The presence of TGF- and of its recetor in digestive tract epithelium and pancreatic tissues early in fetal life suggests a functional role for TGF- during the developmental process of the digestive system. Summary The retinal projections to the brain were studied in three species of European Salamandridae using anterograde transport of horseradish peroxidase and autoradiography. The results obtained were basically identical for all species and confirmed earlier findings on the fiber supply to the preoptic nucleus and the basal optic neuropil. In the anterior thalamus projections to three distinct terminal fields are clearly visible: (i) the diffusely stained corpus geniculatum thalamicum, (ii) the neuropil of Bellonci, pars lateralis, and (iii) a dorsomedial terminal field, the neuropil of Bellonci, pars medialis. Caudal to these terminal fields is an almost terminal-free region, the lateral neuropil. In the posterior thalamus a medial terminal field, the uncinate field, and a laterally located terminal field, the posterior thalamic neuropil, are distinguishable. The tectum opticum displays as many as four dense layers of retinofugal fibers and terminals in the rostral part and, in addition, a more densely stained strip of neuropil running from rostral to caudal over the tectum. The extent of ipsilateral fibers is greater than previously reported in other urodele species. They supply the medial and the lateral parts of the neuropil of Bellonci, the uncinate field, and reach the tectum opticum via the medial optic tract. Further, they form terminals in the innermost optic fiber layer throughout the rostral half of the ipsilateral tectum. A small proportion of ipsilateral fibers contributes very sparsely to all other thalamic terminal fields, leaving only the caudal part of the tectum and several layers of the rostral tectum completely free of a direct retinofugal fiber supply. 相似文献
