 Gene sequences are rapidly accumulating for many commercially and scientifically important plants. These resources create the basis for developing sequence-based markers for mapping and tracking known (candidate) genes, thereby increasing the utility of genetic maps. Members of most of the gene families underlying the synthesis of seed oil fatty acids have been cloned from the medium-chain oilseed Cuphea. Allele-specific-PCR (AS-PCR) and single-strand conformational polymorphism (SSCP) markers were developed for 22 fatty acid synthesis genes belonging to seven gene families of Cuphea using homologous and heterologous DNA sequences. Markers were developed for 4 fatty-acyl-acyl carrier protein thioesterase, 2 β-ketoacyl-acyl carrier protein synthase I, 4 β-ketoacyl-acyl carrier protein synthase II, 3 β-ketoacyl-acyl carrier protein synthase III, 3 acyl carrier protein, 2 β-ketoacyl-acyl carrier protein reductase, and 4 enoyl-acyl carrier protein reductase loci. Eighty-eight percent (14 of 16) of the SSCP loci were polymorphic, whereas only 9% (2 of 22) of the AS-PCR loci were polymorphic. These markers were mapped using a Cuphea viscosissima×C. lanceolata F2 population and produced linkage groups of 10, 3, and 2 loci (3 loci segregated independently). The 10-locus linkage group had every gene but one necessary for the synthesis of 2- to 16-carbon fatty acids from acetyl-CoA and malonyl-ACP (the missing gene family was not mapped). The present study shows that copper oxidation of LDL is a tightly-ordered process which can be finely controlled by appropriate selection of duration of oxidation and of concentrations of LDL and copper. Oxidation of LDL (0.1-2.0 mg LDL protein/ml) was carried out by copper catalysis (in the ratio of 2.5 μM Cu2+ to 0.1 mg LDL protein/ml) in phosphate-buffered saline, and was monitored by agarose gel electro-phoresis, gas chromatography (GC), anion exchange fast protein liquid chromatography (FPLC), fluorescence spectroscopy and dynamic light scattering. Analysis of the data showed strong cross correlations between many of the parameters of oxidation. Oxidation was more rapid for lower concentrations than for higher concentrations of LDL, despite the same ratio of copper to LDL being employed. Chemical kinetics analysis of the GC data suggested that 7β-hydroxycholesterol formation occurred as a first order (or pseudo first order) consecutive reaction to the oxidation of linoleate. The first order rate constants for decomposition of lioleate and production of 7β-hydroxycholesterol correlated closely with the theoretically-calculated times between collision of LDL particles. LDL particle diameter, measured by dynamic light scattering, increased by ca. 50% over 24 h oxidation, suggesting unfolding of apo B-100.

Prolonged oxidation of LDL at low concentration suggested that the radical chain reaction was able to propagate, albeit slowly, on cholesterol after all the polyunsaturated fatty acid was consumed. For higher concentrations of LDL, prolonged oxidation resulted in partial aggregation. These findings are applicable to preparing oxidised LDL with different degrees of oxidation, under controlled conditions, for studying its biological properties.  相似文献

The Vmax of rat muscle mitochondrial CPT I toward the coenzyme A derivatives of 16:0, 16:1n-7, 18:1n-9, and 22:6n-3 were far lower than those recorded previously for this enzyme in rat liver at the same temperature (37°C). However, the Vmax of 7.0 nmol · min−1 · mg mitochondrial protein−1 for linoleoyl CoA (18:2n-6), which was the greatest recorded for the five acyl CoAs examined in muscle, was similar to that in liver. These comparisons presumably reflect a difference in the essential fatty acid requirements of these two rat tissues. Although the Vmax values for CPT I in the musculature of a lower vertebrate (larval lamprey) at 20°C were similar to those exhibited toward the coenzyme A derivatives of 16:0, 16:1n-7, 18:1n-9, and 22:6n-3 by the CPT I of rat musculature at 37°C, the corresponding Vmax toward 18:2n-6 (3.2 nmol · min−1 · mg mitochondrial protein−1) was lower. The latter relatively low activity may spare from oxidation this essential fatty acid, which is in low abundance in the diet of larval lampreys. Although the Vmax values toward the four nonessential fatty acids in larval lamprey muscle were similar to those in rat muscle, the corresponding K0.5 values were lower, thus indicating that the musculature of larval lampreys has a high capacity for energy generation through β-oxidation.  相似文献

A bacterium isolated from a dry soil sample collected from McCalla, AL, USA, converted linoleic acid to a novel compound, 12,13,17-trihydroxy-9 (Z)-octadecenoic acid (THOA). The organism is a Gram-positive, non-motile rod (0.5 μ m × 2 μ m). It was identified as a species of Clavibacter ALA2. The product was purified by high pressure liquid chromatography, and its structure was determined by 1H and 13C nuclear magnetic resonance and Fourier transform infrared spectroscopies, and by mass spectrometer. Maximum production of THOA with 25% conversion of the substrate was reached after 5–6 days of reaction. THOA was not further metabolized by strain ALA2. This is the first report of a 12,13,17-trihydroxy unsaturated fatty acid and its production by microbial transformation. Some dihydroxy intermediates were also detected. Tetreutreptia pomquetensis McLachlan, Fritz et Seguel, a quadriflagellated euglenoid isolated from a shallow-water embayment of the southeastern Gulf of St. Lawrence, is a common winter species where habitats are ice covered for 4 to 5 months. Growth in culture, which was similar to that of many euryhaline species, was limited at normal seawater salinities to temperatures between ∼0° and 7° C, and the alga could not be maintained at 10° C; thus, it is a strict psychrophile. The maximum growth rate (μ≅ 0.35·days−1) was around 5° C and, given its size (∼100 μm length), high productivities are indicated. The major fatty acids (FAs) identified were 18:4ω3, 16:4ω3, 20:5ω3, and 16:0. The overall FA spectrum does not align closely to other euglenoids but may reflect more the low growth temperature. The presence of 18:5ω3 is unusual, as this FA had been known previously only from dinoflagellates and haptophytes. The two most abundant sulfoquinovosyl diacylglycerols (SQDGs) had pairings of [18:4/16:0] and [18:4/18:4] and together constituted 80% of the total SQDGs. The latter pairing is unique, not having been previously recognized. The FA data support the hypothesis that the degree of unsaturation is indicative of low temperatures and membrane integrity.  相似文献

The fatty acid and sterol compositions of five species of marine dinoflagellates (Scrippsiella sp. Symbiodinium microadriaticum Freud, Gymnodinium sp., Gymnodinium sanguineum Hirasaki, and Fragilidium sp.) are reported. All contained the major fatty acids that are considered common in dinoflagellates, but the proportions were quite variable, and some species contained low contents of some polyunsaturated fatty acids. Concentration ranges for the major fatty acids were: 16:0 (9.0%–24.8%), 18:4(n-3) (2.5%–11.5%), 18:5(n-3) (7.0%–43.1%), 20:5(n-3) (EPA) (1.8%–20.9%), and 22:6(n-3) (DHA) (9.9%– 26.3%). Small amounts of novel very-long-chain highly unsaturated C28 fatty acids occurred in all species. Each dinoflagellate contained a complex mixture of 4-methyl sterols and 4-desmethyl sterols. Four species contained cholesterol, although the amounts were highly variable (from 0.2% of total sterols in Scrippsiella sp. to 45.6% in Fragilidium sp.). All but G. sanguineum contained the 4-methyl sterol dinosterol, and all species contained sterols lacking a double bond in the ring system (i.e. stanols); in Scrippsiella sp. cholestanol composed 24.3% of the total sterols. Other common features of the 4-methylsterol profiles were the presence of 23,24-dimethyl alkylation and unsaturation at Δ22 in the side chain. In Scrippsiella sp., four steroidal ketones were identified: cholestanone, dinosterone, 4α,23,24-trimethyl-5α-cholest-8(14)-en-3-one, and dinostanone. The structures of these corresponded to the major sterols in this species, suggesting that the sterols and steroidal ketones are biosynthetically linked. Steroidal ketones were not detected in the other species. Although fatty acid profiles can be used to distinguish among algal classes, they were not useful for differentiating among dinoflagellate species. In contrast, whereas some taxonomic groupings of dinoflagellates display similar sterol patterns, others, such as the gymnodinoids studied here, clearly do not. The combination of fatty acid, sterol, and steroidal ketone profiles may be useful complementary chemotaxonomic tools for distinguishing morphologically similar species. The identification of steroidal ketones supports earlier suggestions that certain dinoflagellates might be a significant source of such components in marine environments.  相似文献

Polarella glacialis (Montresor et al.) was identified in Davis Station sea ice by morphological and DNA sequence comparison of cultures with those of the authentic strain P. glacialis CCMP 1383 isolated from McMurdo Sound. Cells and cysts of the Davis isolate (FL1B) were morphologically indistinguishable from P. glacialis, and comparison of the large subunit rDNA of both cultures demonstrated only 0.2% sequence divergence over 1366 base pairs. The photosynthetic pigments of P. glacialis (strains FL1B and CCMP 1383) were typical of dinoflagellates, with peridinin (contributing up to 31%) as the major accessory pigment. Extremely high levels of polyunsaturated fatty acids (PUFA, up to 76.3%) were characteristic of P. glacialis isolate FL1B. The high PUFA concentration of this species is thought to be an adaptation to survive the cold temperatures of the upper fast ice. The sterol profile of FL1B was atypical of dinoflagellates, with 4‐desmethylsterols (up to 79%) in greater abundance than 4α‐methyl sterols (up to 24%). 27‐Nor‐24‐methylcholest‐5,22E‐dien‐3β‐ol was identified as the principle sterol in P. glacialis, contributing up to 64% of the total sterol composition.  相似文献

Divergent natural selection is often believed to be the driving force behind phenotypic differentiation in characters related to resource acquisition, leading to trophic polymorphism in fishes. Here we use variation in the fatty acid composition of adipose and muscle tissues to look at differences in resource use by two recently described sympatric morphs of Percichthys trucha , a common freshwater fish of the Andean and Patagonian regions of South America. Because dietary fatty acids are often stored in carnivorous animals with litde modification after consumption, they can be used to infer information about dietary habits of individuals. We found that the two morphs differed in the overall composition of fatty acids in both adipose and muscle tissue, but that there were some differences in how die morphs differed in lakes from the northern vs southern part of the range. Furthermore, we found that certain fatty acids were correlated with diet as determined by gut content analysis. Consumption of anisopteran larvae was highly correlated widi 14:0 in adipose and muscle tissue; and higher levels of longer chain unsaturated fatty acids (i.e. 20 and 22 carbons) were correlated with die presence offish and also amphipods in die diets. Taken togetiier, die results suggest that diere are marked differences in die foraging ecology of die two morphs of P. trucha inhabiting southern Andean lakes.  相似文献

The phospholipid fatty acid composition of Porphyridium purpureum on a solid medium was studied in the presence of sodium dodecyl sulphate (SDS) and cetyl trimethylammonium bromide (CTAB). The most common fatty acids in phosphatidyl choline (PC) and phosphatidyl ethanolamine (PE) were palmitic (16:0), stearic (18: 0), linoleic (18:2ω 6), arachidonic (20:4ω 6) and eicosapentaenoic (20:5ω 3) acids, 20:4ω 6 being very abundant. In phosphatidyl glycerol (PG) the most common acids were 16:0, trans-hexadecenoic acid (tr 16:1ω 13), oleic acid (18:1) and 20:4ω 6. Both detergents increased the saturation grade of PC and PE by decreasing the relative amount of the polyunsaturated acids, especially 20:4ω 6. A corresponding increase in the amounts of saturated acids was observed in PC and PE. The changes in PG fatty acid composition were not very significant: a slight increase was observed in the amounts of 16:0 and tr 16:1ω 13 , with a corresponding decrease in the amounts of 20:4ω 6 and 20:5ω 3. Both detergents decreased the PC/PE and the (PC + PE)/PG ratios very markedly, most probably as a result of increases in the amounts of PE and PG. In the presence of CTAB the cells seemed to contain much more phospholipids than in the presence of SDS, perhaps as a result of the mucilage-precipitating effect of CTAB. Fatty acids in the leaves of the citrus hybrid B5-9-68, a BC1 progeny [(Citrus paradisi x Poncirus trifoliata) x C. sinensis], were compared with those in P. trifoliata and C. sinensis. Total lipid and triacylglycerol fatty acid profiles of the cold hardened hybrid were similar to the profiles of the hardy P. trifoliata and different from the profiles of the less hardy C. sinensis. When subjected to a freeze regime, the unhardened hybrid lost 22% leaf fatty acids during the freeze stage and 13% during the thaw stage. Linolenic acid accounted for 98% of the fatty acid decrease. Degradation was greatest in phosphatidylcholine (89%), phosphatidylglycerol (83%), monogalactosyldiglyceride (79%) and digalactosyldiglyceride (50%). Phosphatidic acid increased 4-fold over the two stress stages. Total leaf fatty acids during the freeze-thaw regime increased 12% in the cold hardened hybrid. Three molecular species of triacylglycerol which were rich in linolenic acid increased in the hardened hybrid during the freeze-thaw regime. The increase in highly unsaturated triacylglycerol species under freeze-thaw stress suggests that triacylglycerol has a role in maintaining the fluidity of biomembranes during freezing conditions.  相似文献
