A batch test and continuous operation were performed to identify the effect of lactate on hydrogen production at pH 4.5. When the initial lactic acid concentration was increased from 0 to 8 g/L in the batch test, the hydrogen yield also increased from 1.41 to 1.72 mol-H2/mol-glucose. The system exhibited a long lag time and an insignificant hydrogen yield with 16 g-lactic acid/L. A continuous stirred tank reactor (CSTR) was operated at different organic loading rates (OLRs: 10, 15, 20 and 40 g/L/day) and hydraulic retention times (HRTs: 6, 12 and 24 h). At an OLR of 20 g-glucose/L/day and 12 h of HRT, the hydrogen yield was 1.2 mol-H2/mol-glucose. The yield decreased with a 24 h HRT. Even though lactate was one of the major constituents of volatile fatty acids (VFAs), hydrogen production was feasible throughout the operation. Clostridium sp. was the dominant hydrogen-producing bacteria in the system.  相似文献

Pore water dynamics in the sediment of a shallow and hypertrophic lake   总被引:1,自引:3,他引:1  
Seasonal variations in pore water with main stress on pH and phosphate were investigated in the sediment of the shallow and hypertrophic Lake Søbygaard, Denmark. The purpose was to evaluate factors affecting the internal phosphorus loading. Pore water was obtained by in situ incubation of ceramic cups, sampled anaerobicaly from a fixed position in the sediment. The method is evaluated. During summer, pH and phosphate concentrations increased in the upper 8–10 cm of the sediment. Increased pH was most pronounced in the upper 5 cm, where pH increased to between 9 and 10. This is believed to be caused by the photosynthetically elevated pH in the above lake water. Phosphate concentrations increased with depth, from 0–2 mg P 1–1 in the upper 5 cm to 3–6 mg P 1–1 in 6–10 cm depth. Average phosphate gradient in the upper 6–8 cm was 1.0 mg P 1–1 cm –1 in the summer decreasing to 0.2 mg P 1–1 cm t1 in the autumn/winter. In spite of low redox potential, Fe(II) was not present in the upper 20 cm. The seasonal variation in pore water phosphate is believed mainly to be due to the variations in pore water pH inducing a substitution of phosphate ions with hydroxyl ions on ironhydroxides during summer. A considerable sedimentation of organic bound phosphorus and decomposition in the sediment is also considered important. Phosphorus release from the sediment is facilitated by bio- and gas turbation and by the frequent occurrence of resuspension caused by windaction. Net release rate is highly variable during the season. The summer average is 40 mg P m–2 d–1.  相似文献

Summary Translocation of 14C assimilates was studied on four different transport systems of Picea abies branches after induced activation in January. 14CO2 assimilation of terminal shoots for 48 h at 25° C resulted in phloem loading and basipetal transport of 14C photosynthate into the following, older shoot generations. 14C import was enhanced, when these older shoot generations were kept in the dark. Microautoradiographs of the labelled terminal shoots showed that 14C assimilates were exported from needles via sieve elements of the leaf traces and loaded into the latest increment of the axial secondary phloem. No 14C label appeared in the obliterated sieve cells or in the tracheids. In addition, 14C photosynthate accumulated densely in the chlorophyllous cells of the cortex and in cells of the resin ducts, indicating certain sink activity. In the darkened 2-year-old shoot, imported 14C photosynthate was concentrated in the functional secondary phloem, while some 14C label was unloaded into the latest xylem increment. When 6-year-old shoots were exposed to 14CO2 for 48 h in the light, 14C assimilates accumulated in the phloem of the leaf trace and in the latest increment of the axial secondary phloem. However, a substantial amount of radioactivity was unloaded into ray cells and phloem parenchyma cells. Thus, the presence of functioning phloem in needles and twigs of P. abies during winter allows long-distance translocation and radial distribution of assimilates according to existing source-sink relations.  相似文献

This study investigated the potential use of static osmotic loading as a cartilage tissue engineering strategy for growing clinically relevant grafts from either synovium-derived stem cells (SDSCs) or chondrocytes. Bovine SDSCs and chondrocytes were individually encapsulated in 2% w/v agarose and divided into chondrogenic media of osmolarities 300 (hypotonic), 330 (isotonic), and 400 (hypertonic, physiologic) mOsM for up to 7 weeks. The application of hypertonic media to constructs comprised of SDSCs or chondrocytes led to increased mechanical properties as compared to hypotonic (300 mOsM) or isotonic (330 mOsM) media (p<0.05). Constant exposure of SDSC-seeded constructs to 400 mOsM media from day 0 to day 49 yielded a Young's modulus of 513±89 kPa and GAG content of 7.39±0.52%ww on day 49, well within the range of values of native, immature bovine cartilage. Primary chondrocyte-seeded constructs achieved almost as high a Young's modulus, reaching 487±187 kPa and 6.77±0.54%ww (GAG) for the 400 mOsM condition (day 42). These findings suggest hypertonic loading as a straightforward strategy for 3D cultivation with significant benefits for cartilage tissue engineering strategies. In an effort to understand potential mechanisms responsible for the observed response, cell volume measurements in response to varying osmotic conditions were evaluated in relation to the Boyle–van't Hoff (BVH) law. Results confirmed that chondrocytes behave as perfect osmometers; however SDSCs deviated from the BVH relation.  相似文献

Gap junction intercellular communication (GJIC) consists of intercellular exchange of low molecular weight molecules. Chemically induced alterations of this communication have been suggested to result in abnormal cell growth and tumour promotion. Several in vitro assays have been developed to determine the effect of chemicals on gap junction communication in cultured cells. The scrape loading dye transfer technique is based on studying the transfer of the fluorescent dye Lucifer Yellow in cells where the dye is loaded through a cut in the cell monolayer. This technique is rapid and relatively uncomplicated, but has only been used to qualitatively demonstrate communication, due to lack of an appropriate method for quantification of the dye spreading. We show here that analysis of digital fluorescence images of cells scrape loaded with Lucifer Yellow can be used for quantitative determination of GJIC. We have analysed the images both by means of distance of diffusion of the dye in the cell monolayer, as well as by area of dye-coupled cells. The results are consistent with that obtained using microinjection of Lucifer Yellow and the method offers a simple way for quantitative determination of GJIC.  相似文献

目的:评定低温环境下持续性军事职业活动对抑制控制能力的影响,为精准军事运动训练提供依据。方法:20名健康男性青年学员,平均年龄:(23.32±1.62)岁;身高:(175.34±4.14) cm;体重:(68.19±3.12) kg,分别在常温(16℃~23℃)和低温(-3℃~-1℃)环境下,持续36 h时间内完成多项军事课目,记录全程RR间期和核心温度,活动前后进行斯特鲁普色词测试、闪光融合临界频率测试。结果:与安静状态相比,两次军事职业活动中的心率、运动后的过氧消耗和运动冲量值均显著增加(P<0.05),但两次活动之间并无显著差异(P>0.05);与安静值相比,两次活动平均核心温度和最大核心温度均显著增加(P<0.05),与常温环境相比,低温环境下核心温度的平均值显著增加(P<0.05),但最大核心温度无显著差异(P>0.05);与安静状态相比,两次活动后抑制控制能力均显著降低(P<0.05),且低温环境下显著低于常温环境(P<0.05)。另外,与安静状态相比,两次活动均导致明显的精神疲劳(P<0.05),低温环境下更为严重(P<...  相似文献

A sample of 32 obstructive sleep apnea patients (27 males, 5 females) was assessed with overnight polysomnography and the Multiple Sleep Latency Test (MSLT), an objective measure of daytime sleepiness. Patients also participated in a maximal exercise test, which served as an objective indicator of physical fatigue. The Fatigue Severity Scale (FSS) was used as a subjective measure of fatigue. Subjective fatigue ratings were significantly correlated with percent of predicted maximum heart rate achieved during exercise testing, suggesting that self-reported fatigue in apnea patients may refer to reduced physical fitness. FSS scores and exercise testing results were not significantly correlated with the MSLT, indicating that daytime fatigue and daytime sleepiness are independent problems in apnea patients. Participants self-reported a high level of fatigue, and exercise testing revealed decreased physical work capacity among apnea patients, but objective and subjective indicators of fatigue were not significantly correlated with apnea severity. This study focused on an investigation of a high drug-loaded solid dispersion system consisting of drug, carrier, and surfactant. Solid dispersions of a water-insoluble ofloxacin (OFX) with polyethylene glycol (PEG) of different molecular weights, namely binary solid dispersion systems, were prepared at drug to carrier not less than 5∶5. Polysorbate 80, a nonionic surfactant, was incorporated into the binary solid dispersion systems as the third component to obtain the ternary solid dispersion systems. The powder x-ray diffraction and differential scanning calorimetric studies indicated that crystalline OFX existed in the solid dispersions with high drug loading. However, a decreased crystallinity of the solid dispersions obtained revealed that a portion of OFX was in an amorphous state. The results indicated a remarkably improved dissolution of drug from the ternary solid dispersion systems when compared with the binary solid dispersion systems. This was because of polysorbate 80, which improved wettability and solubilized the non-molecularly dispersed or crystalline fraction of OFX.  相似文献

The possibility of shifting a methanogenic process for hydrogen production by changing the process parameters viz., organic loading rate (OLR) and hydraulic retention time (HRT) was evaluated. At first, two parallel semi-continuously fed continuously stirred tank reactors (CSTR) were operated as methanogenic reactors (M1 and M2) for 78 days. Results showed that a methane yield of 198-218 L/kg volatile solids fed (VS(fed)) was obtained when fed with grass silage at an OLR of 2 kgVS/m3/d and HRT of 30 days. After 78 days of operation, hydrogen production was induced in M2 by increasing the OLR from 2 to 10 kgVS/m3/d and shortening the HRT from 30 to 6 days. The highest H? yield of 42 L/kgVS(fed) was obtained with a maximum H? content of 24%. The present results thus demonstrate that methanogenic process can be shifted towards hydrogen production by increasing the OLR and decreasing HRT.  相似文献
