The influence of host odour, windspeed, position of the sun, and temperature on flight behaviour of the cabbage seedpod weevil (Ceutorhynchus assimilis Paykull) were studied. This weevil showed a positive anemotaxis (upwind flight) inside the odour plume of a host crop (Brassica napus L.). Outside the odour plume the weevil showed a pronounced phototaxis at windspeeds below 1.5 m/s. At higher windspeeds, the seedpod weevils flew downwind. The cabbage seedpod weevil flies most readily at low windspeeds (less than 0.5 m/s) and at temperatures above 22 °C.  相似文献

A method for the on-line calculation of conversion rates and yield coefficients under dynamic process conditions was developed. The method is based on cumulated mass balances using a moving average method. Elemental balances were used to test the measured cumulated quantities for gross errors and inappropriate stoichiometry definition followed by data reconciliation and estimation of non-measured conversion rates, using a bioprocess set-up including multiple on-line analysis techniques. The quantitative potential of the proposed method is demonstrated by executing transient experiments in aerobic cultures of Saccharomyces cerevisiae on glucose. Rates and yield coefficients could be consistently quantified in shift-up, shift-down, and accelerostat experiments. The method shows the capability to describe quantitatively transient changes in metabolism including uncoupling of catabolism and anabolism, also for the case when multiple components of metabolism are not measured. The validity of the experiment can be evaluated on-line. Additionally, the method detects with high sensitivity inappropriate stoichiometry definition, such as a change in state of metabolism. It was shown that concentration values can be misleading for the identification of the metabolic state. In contrast, the proposed method provides a clear picture of the metabolic state and new physiological regulations could be revealed. Hence, the novelty of the proposed method is the on-line availability of consistent stoichiometric coefficients allowing a significant speed up in strain characterization and bioprocess development using minimal knowledge of the metabolism. Additionally, it opens up the use of transient experiments for physiological studies.  相似文献

Transgenic melon and squash containing the coat protein (CP) gene of the aphid transmissible strain WL of cucumber mosaic cucumovirus (CMV) were grown under field conditions to determine if they would assist the spread of the aphid non-transmissible strain C of CMV, possibly through heterologous encapsidation and recombination. Transgenic melon were susceptible to CMV strain C whereas transgenic squash were resistant although the latter occasionally developed chlorotic blotches on lower leaves. Transgenic squash line ZW-20, one of the parents of commercialized cultivar Freedom II, which expresses the CP genes of the aphid transmissible strains FL of zucchini yellow mosaic (ZYMV) and watermelon mosaic virus 2 (WMV 2) potyviruses was also tested. Line ZW-20 is resistant to ZYMV and WMV 2 but is susceptible to CMV. Field experiments conducted over two consecutive years showed that aphid-vectored spread of CMV strain C did not occur from any of the CMV strain C-challenge inoculated transgenic plants to any of the uninoculated CMV-susceptible non- transgenic plants. Although CMV was detected in 3% (22/764) of the uninoculated plants, several assays including ELISA, RT- PCR-RFLP, identification of CP amino acid at position 168, and aphid transmission tests demonstrated that these CMV isolates were distinct from strain C. Instead, they were non-targeted CMV isolates that came from outside the field plots. This is the first report on field experiments designed to determine the potential of transgenic plants expressing CP genes for triggering changes in virus-vector specificity. Our results indicate that transgenic plants expressing CP genes of aphid transmissible strains of CMV, ZYMV, and WMV 2 are unlikely to mediate the spread of aphid non-transmissible strains of CMV. This finding is of practical relevance because transgenic crops expressing the three CP genes are targeted for commercial release, and because CMV is economically important, has a wide host range, and is widespread worldwide.  相似文献

Forests of the future need to cope with adverse climatic conditions, in particular drought, to ensure forest stability and productivity. Given the usually long rotation period of forests, forest managers need to select appropriate, i.e. productive and climate-change resilient tree species and/or provenances, to lower tree-mortality risks and sustain current wood production rates at the end of the 21st century. A frequent means of assessing which provenances of a given species are adapted to anticipated climate conditions is common garden experiments, where trees from different provenances are planted under similar climate conditions. However, in this context soil conditions also play an important role, since they govern how climate translates into plant-available water and hence plant’s ability to cope with extreme drought events. Here, we examine the effects of soil conditions on pedunculate oak (Quercus robur L.), by studying 10 different Dutch oak provenances that were planted in three provenance trials on different soil types in 1988 in the Netherlands. Using dendroecological methods, we quantified provenance-specific productivity and assessed provenance- and site-specific growth patterns. Our results indicated clear differences in productivity among provenances as well as soil-type specific growth patterns. Consequently, our study highlights the importance of incorporating soil characteristics when evaluating the growth performance of provenances within common garden experiments.  相似文献
