采用休止细胞梯度生长法, 对工业糖蜜酿酒酵母(Saccharomyces cerevisiae)菌株进行高浓度酒精、高温和高渗透压, 以及糠醛毒性、苯酚毒性、乙酸毒性和抗生素G418毒性的耐受性分析。结果表明, 所测定的工业酵母菌株对这些逆境条件的耐受性有明显的差别; 其中AS2.1189和AS2.1190对测定的胁迫条件均表现出相对较好的耐受性; 396对乙酸毒性和G418毒性具有很好的耐受性; 2610对高温表现出较强的耐受性。  相似文献

The investigation presented here describes a protocol designed to perform high-throughput metabolic profiling analysis on human blood plasma by ultra-performance liquid chromatography/mass spectrometry (UPLC/MS). To address whether a previous extraction protocol for gas chromatography (GC)/MS-based metabolic profiling of plasma could be used for UPLC/MS-based analysis, the original protocol was compared with similar methods for extraction of low-molecular-weight compounds from plasma via protein precipitation. Differences between extraction methods could be observed, but the previously published extraction method was considered the best. UPLC columns with three different stationary phases (C8, C18, and phenyl) were used in identical experimental runs consisting of a total of 60 injections of extracted male and female plasma samples. The C8 column was determined to be the best for metabolic profiling analysis on plasma. The acquired UPLC/MS data of extracted male and female plasma samples was subjected to principal component analysis (PCA) and orthogonal projections to latent structures discriminant analysis (OPLS-DA). Furthermore, a strategy for compound identification was applied here, demonstrating the strength of high-mass-accuracy time-of-flight (TOF)/MS analysis in metabolic profiling.  相似文献

荒漠对气候变化具有高度敏感性, 深刻认识和量化非生物因子对荒漠生态系统土壤呼吸的影响具有重要意义。采用自动CO2通量系统(Li-8100)监测了梭梭(Haloxylon ammodendron)、假木贼(Anabasis aphylla)和盐穗木(Halostachys caspica)群落生长季土壤呼吸及温度、土壤含水量等, 深入分析了水热因子对土壤呼吸的影响。土壤呼吸具有不对称的日格局, 最小值出现在8:00, 最大值在12:00~14:00。土壤呼吸的季节格局与气温变化基本同步, 最小值在生长季末期(10月), 最大值在生长季中期(6~7月)。梭梭、假木贼和盐穗木群落生长季平均土壤呼吸速率分别为0.76、0.52和0.46 μmol CO2·m-2·s-1。气温对假木贼(51%)和盐穗木群落(65%)土壤呼吸季节变化的解释率高于梭梭(35%)。梭梭、假木贼和盐穗木群落土壤呼吸温度敏感性(Q10)逐渐增大, 基础呼吸速率(R10)逐渐减小。剔除温度影响后, 梭梭、假木贼群落土壤呼吸与土壤含水量呈显著的幂二次方函数关系, 盐穗木群落两者关系却明显减弱, 未达到显著水平。气温、土壤含水量的二元方程均能解释群落土壤呼吸大部分的时间变异: 梭梭群落71%~93%、假木贼群落79%~82%、盐穗木群落70%~80%。人工模拟降水后土壤呼吸速率表现出降水后10 min减小、180 min时明显增加、达到最大值后再次衰减的现象。5和2.5 mm降水处理下的土壤呼吸速率最大值和其后的递减值高于对照处理, 土壤呼吸增加、达到峰值和其后递减过程与5 cm土壤温度变化基本同步。  相似文献

燃料乙醇作为一种优良的可再生液体燃料,其开发利用受到了人们的广泛关注。微藻是一种高光合、高产生物量的生物质资源,很多的藻体细胞中含有大量的淀粉、纤维素(Iα型)等多糖物质,是制备燃料乙醇的优良原料。发展利用微藻制备燃料乙醇技术工艺,对于缓解我国目前日益短缺的能源问题,减少温室气体排放和环境污染等具有很好的应用前景。综述了国内外利用微藻生物质制备燃料乙醇中所用到的关键技术、存在的问题以及今后的发展前景等。  相似文献

Changes in the total phenolics, condensed tannins (CT), protein-precipitable phenolics content and protein precipitation capacity were determined on a series of mangrove leaves from two true viviparous mangrove species (Kandelia candel and Bruguiera gymnorrhiza) at various stages of development and decomposition in the Jiulong River Estuary, Fujian, China. Similar measurements were also done for the propagules at different developmental stages. The results showed that the total phenolics, extractable condensed tannins, total condensed tannins, protein-precipitable phenolics content and protein precipitation capacity in young leaves were higher than those in mature and senescent leaves. Tannin dynamics during leaf decomposition varied with species, and the rapid loss of phenolics observed during decomposition can be ascribed to leaching and degradation. Protein-bound CT and fibre-bound CT tended to increase with leaf decomposition, with CT binding more strongly to protein than to fibre. Protein-bound CT was higher than fibre-bound CT with the exception of mature leaves. Total phenolics, extractable CT and protein-precipitable phenolics contents in flower tissues were relatively lower than those in hypocotyls at different developmental stages. Protein precipitation capacity fluctuated with the development of propagules. Increases in nitrogen in decaying litter, and declines in contents of total phenolics and total condensed tannins of detritus support the general conclusion that decomposing mangrove detritus can be a more palatable heterotrophic substrate than living leaves.  相似文献

实验显示,一种氨基酸混合液(含异亮氨酸、甲硫氨酸和苯丙氨酸,添加浓度分别为1.0、0.5和2.0g/L)能显著提高自絮凝酵母——粟酒裂殖酵母和酿酒酵母融合株SPSC的耐酒精能力。实验将菌体分别培养于添加(试验组)和未添加(对照组)该氨基酸混合液的条件下,然后收集菌体进行酒精(20%,V/V)冲击试验(30℃,9h),结果,试验组的菌体尚有一半以上的存活细胞,而对照组的菌体全部死亡。通过对试验组和对照组的菌体细胞膜蛋白质氨基酸组成分析发现,试验组的菌体耐酒精能力提高与所添加氨基酸组入菌体的细胞膜密切相关。以DPH为荧光探针的细胞膜流动性测定分析进一步揭示,氨基酸组入菌体的细胞膜后,细胞膜能有效抵抗高浓度酒精冲击诱发的膜流动性的提高,从而维持膜的稳定。因此,实验首次揭示膜蛋白氨基酸组成可通过改变膜流动性而影响酵母菌的耐酒精能力。  相似文献

1. Historical nutrient changes in Grasmere were investigated using a 300‐year record derived from six sediment cores. One core was investigated at high resolution for diatoms, total sedimentary phosphorus, and loss‐on‐ignition (LOI), and was dated using 210Pb and 137Cs. Six other cores were scanned for magnetic susceptibility, diatoms and LOI to confirm the stratigraphic integrity of the primary record. 2. A rise in nutrient levels occurred after 1855 AD. This event was marked by a shift away from benthic diatom assemblages and a rise in Asterionella formosa. The onset of eutrophication from 1855 corresponds to the expansion of the local and tourist population in the area. 3. The replacement of A. formosa with Cyclotella spp. ca 1945–65 indicates reduced nutrient loads, possibly because of enhanced flushing brought about by the seasonal rainfall distribution. 4. After 1965 a step‐wise increase in both absolute and relative amounts of Asterionella was found. High sedimentary P and diatom inferred TP confirmed the high nutrient loading of the lake. Nutrient increase is attributable to problems with the Grasmere village sewage system and the installation of a wastewater treatment works (WwTW) on the River Rothay in 1971. Modifications to the WwTW in 1982 caused an initial improvement, but have not led to a full recovery to pre‐1965 ecological conditions. 5. The diatom record indicates a further improvement after 1990 by a return toward Achnanthes minutissima. 6. The sedimentary archive of sensitive sites provides important benchmarks against which to judge the attainment of water quality targets.  相似文献

通过对混凝剂(PAS)用量、酸度、温度对垃圾处理过程中渗滤液混凝效果影响的研究,结果表明:用聚合硫酸铝进行混凝处理时,对渗滤液的氨氮处理效果不佳,对CODcr和浊度处理效果甚佳.根据三因素三水平的正交实验,得出处理过程中三个因素对CODcr去除率影响的主次关系为:pH>聚合硫酸铝的用量>温度.验证试验结果表明:混凝剂加入量15mL(10%)、pH为6.7、温度为50℃时为最优化工艺条件,CODcr去除率达到76.2%.  相似文献

The extent of impairment to some Appalachian watersheds from acid precipitation is so extreme that watershed scale analytical tools are needed to help guide cost‐effective management decisions. The objective of this study was to develop a measure of the functional value of streams as potential areas for juvenile Brook trout recruitment. This measure, which we term “weighted potential recruitment area” (WPRA), is a function of the expected Brook trout spawning intensity and juvenile survivorship. Estimates of WPRA for each stream segment were then used to identify restoration priorities and optimal restoration programs in the upper Shavers Fork watershed in West Virginia, U.S.A. Using this approach, we determined that the watershed has lost nearly 80% of its historic juvenile recruitment potential as a result of acid precipitation. We also determined that of the 145 stream segments in the watershed, eight critical stream segments account for nearly 20% of the loss. The costs and ecological benefits of a series of five alternative restoration programs were then assessed using an ArcGIS model (Environmental Systems Research Institute, Redlands, CA, U.S.A.). This approach identified two “optimal” alternatives: (1) a low‐cost, moderate‐benefit approach that would use existing rail access to treat acidification in three critical headwater locations and (2) a high‐cost, high‐benefit approach that would use aerial limestone application to treat numerous acidic tributaries near their source. The measure of stream ecological value that we developed was effective in identifying critical restoration priorities and optimal restoration strategies in this watershed. A similar procedure could be used to guide watershed restoration decisions throughout the Appalachian region.  相似文献

 利用分布于我国不同气候区的7个气象台站1951~1995年的月降水资料,比较分析了标准化降水指标(Standardized precipitation index, SPI)和在我国已成熟应用的Z指数。结果表明,SPI计算简单,资料容易获取,而且计算结果与Z指数有极好的一致性。同时,由于SPI是通过概率密度函数求解累积概率,再将累积概率标准化而得,具有稳定的计算特性,消除了降水的时空分布差异,在各个区域和各个时段均能有效地反映旱涝状况,优于在我国广泛应用的Z指数。此外,SPI还可以计算不同时间尺度的指标值,能够满足不同水资源状况分析的要求。以北京为例,探讨了应用4种时间尺度的SPI值反映1951~1995年期间的旱涝事件,发现SPI能准确地反映北京45年间的旱涝趋势,对于旱涝灾害有着良好的预测作用,可用于我国的水资源评估以及不同时间尺度的干旱监测。  相似文献
