Leaf extracts of several taxa ofArgyranthemum (nanophaneroand chamaephytes endemic to the Macaronesian Islands) andChrysanthemum s. str. (therophytes of the West Mediterranean region) were subjected to polyacrylamide disc gel electrophoresis and analysis of peroxidase isoenzymes. The differences between the two genera are small; this points to their close affinity. The taxonomic subdivision ofChrysanthemum s. str. is confirmed. ForArgyranthemum the inclusion ofA. callichrysum andA. ochroleucum into sect.Ismelia is suggested.A. foeniculaceum may fit better into the otherwise isolated sect.Monoptera but somewhat approaches sect.Ismelia. Isoenzyme patterns inA. frutescens s. l. seem to reflect a geographical differentiation of the group on 5 Canary Islands.

The effects of the addition and withdrawal of gibberellic acid (GA3) and Ca2+ on enzyme synthesis and secretion by barley (Hordeum vulgare L. cv. Himalaya) aleurone layers were studied. Incubation of layers in GA3 plus Ca2+ affects the total amount of secreted α-amylase (EC 3.2.1.1) and acid phosphatase (EC 3.1.3.2) by promoting the appearance of different isoenzymic forms of these enzymes. The release of α-amylase isoenzymes 1–4 in response to GA3 plus Ca2+ has a lag of 6 h. When layers are incubated in GA3 alone for 6 h prior to the addition of Ca2+, isoenzymes 1–4 appear in the medium after only 30 min. When the addition of Ca2+ to layers pretreated in GA3 is delayed beyond 12 h, its effectiveness in stimulating the synthesis and release of isoenzymes 3 and 4 is diminished. After 35 h of preincubation in GA3, addition of Ca2+ will not stimulate synthesis of α-amylase isoenzymes 3 and 4. Aleurone layers preincubated for 6 h in GA3 will respond to Ca2+ when the GA3 is withdrawn from the incubation medium by producing α-amylase isoenzymes 1–4. The converse is not the case, however, since layers preincubated in Ca2+ for 6 h will not produce all isoenzymes of α-amylase when subsequently incubated in GA3. The Ca2+-stimulated release of α-amylase from GA3 pre-treated layers is dependent on the time of incubation in Ca2+ and the concentration of the ion. The response to Ca2+ is temperature-dependent, and other divalent cations such as Mg2+ cannot substitute for Ca2+. We conclude that Ca2+ influences α-amylase release by influencing events at the biochemical level.  相似文献

Changes in the subcellular distribution of hexokinase activity from three brain regions and heart were studied during alloxan induced diabetes. There was an overall decrease in the particulate hexokinase with an increase in the soluble form, after different time intervals of the onset of diabetes. Administration of insulin to the diabetic rats showed a partial counteraction of the enzyme changes. A possible regulation of brain hexokinase by metabolite changes is proposed  相似文献

This report describes the characterization and partial purification of rat liver 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) synthase activity. A preliminary characterization of Chinese hamster ovary (CHO) cell HMG CoA synthase activity is also presented. Ion-exchange chromatography of ammonium sulfate precipitates of rat liver cytosol indicate the existence of two isoenzymes of HMG CoA synthase. These isoenzymes are physically, catalytically, and immunologically distinct. One of these isoenzymes, peak 1, resembles mitochondrial HMG-CoA synthase activity as evidenced by similarities in elution upon ion-exchange chromatography, inhibition by MgCl2, and cross reactivity with an antibody prepared against the mitochondrial enzyme. As peak 1 activity is unstable, further purification studies were performed on peak 2 activity. Peak 2 can be further resolved into two activities (peaks 2A and 2B) by gel filtration. In contrast, CHO-K1 cells (a permanent fibroblast line) possess only peak 2 type HMG CoA synthase activity.  相似文献

An investigation into isoenzymic analysis using four different buffer/gel systems was carried out. Fourteen different isoenzyme systems were surveyed on each buffer/gel system. It is shown that comparable results are not obtained between different systems. Previous workers in Cepaea nemoralis have used different buffer/gel systems from one another. Alkaline phosphatase and acid phosphatase and certain lactate and malate dehydrogenases are shown to behave similarly in three different systems. It is tentatively suggested that these isoenzymes may be genetically identical. Nothing dehydrogenase activity is demonstrated in gels stained for other dehydrogenases. It is suggested that nothing dehydrogenase activity may be attributable to glutamate or malate dehydrogenase.This work was supported by the Science Research Council.  相似文献

Red cell acid phosphatase phenotypes of 207 captive animals of the genera Lemur, Hapalemur, and Propithecus were determined by starch gel electrophoresis and phosphatase-specific staining. In Lemur fulvus, three phenotypes, designated A, B, and AB, were observed. In each of the species L. catta, L. macaco, L. mongoz, and L. variegatus, a single phenotype was observed. In Hapalemur griseus, three phenotypes were found: A, B, and AB. In Propithecus verreauxi, a single phenotype was found. Examination of breeding records in conjunction with the results of the electrophoretic analyses supports the conclusion that the erythrocytic acid phosphatases in this group of nonhuman primates are the products of at least two codominant autosomal alleles. There is a wide range of specific activities of the acid phosphatases as determined by colorimetric assays. The values range from 60.6 moles of p-nitrophenol released per gram of hemoglobin per 30 min in Lemur catta to 429.1 moles in Propithecus verreauxi. The enzymes of L. fulvus and P. verreauxi were purified approximately 400-fold, and Michaelis-Menten constants were determined on the purified preparations. For L. fulvus phenotype A, K m =0.8 mM; for L. fulvus phenotype B, K m =0.8 mM; and for P. verreauxi, K m =0.6 mM; the substrate in each case was p-nitrophenylphosphate.The work reported here was taken in part from a doctoral dissertation submitted by G.A.M. to the Graduate School of Duke University in partial fulfillment of the requirements for the Doctor of Philosophy degree. (NH4)2SO4 fractionation followed by Sephadex G-200 chromatography of sugar cane juice gave an acid invertase with MW of 380 000 and 23.5% carbohydrate and a neutral invertase with MW of 66 000 and 22% carbohydrate. For acid invertase, Km is 2.8 mM and Vmax is 2.7 μmol sucrose hydrolysed/hr/mg protein. For neutral invertase, Km and Vmax are 0.32 mM and 2.8 μmol hydrolysed/hr/mg protein, respectively. Inhibition of both invertases by either lauryl sulfate or metasilicate is not competitive.  相似文献

Nitrate-grown cells of Stichococcus bacillaris Naeg. (UTEX 314) contained much higher activities of glutamine synthetase (GS) and NADPH-glutamate dehydrogenase (GDH) than ammonium-grown cells. Methylamine, a non-metabolizable ammonium analog, caused a decrease in GS activity in nitrate-grown cells suggesting that GS is regulated by the size of the endogenous ammonium pool. The decrease in GS observed in methylammonium-loaded nitrate-grown cells was accompanied by an increase in NADPH-GDH activity. Stichococcus bacillaris can be grown in the presence of methionine sulfoximine (MSX), a potent inhibitor of GS. However, only a fraction of a control cell population showed a requirement for glutamine or arginine for growth following MSX addition. Fully adapted MSX-grown cells were indistinguishable from control cells in their ability to photosynthesize and utilize amino acids as nitrogen sources. Alanine, arginine, asparagine, glutamine, glycine and proline were good nitrogen sources, and maximum capacity for amino acid transport was developed in cells grown on these amino acids. Compared to nitrate-grown cells the activity of GS in ammo acid-grown cells was low, whereas NADPH-GDH was very active. The activity of NADH-GDH in amino acid-grown cells was highest under heterotrophic conditions.  相似文献

The white-rot fungus Trametes versicolor grown in submerged culture produced two laccase isoenzymes, LacI and LacII. Addition of insoluble lignocellulosic materials into the culture medium increased the total laccase activity. The proportion of laccase isoenzymes also changed depending on the lignocellulosic material employed, with ratios of activity LacII/LacI from 0.9 (barley straw) to 4.4 (grape stalks). Conclusions With the wide availability of insect cell culture media, it can generally be considered a routine process to develop new cell lines. Exceptions to this statement do exist, of course. Difficulties may arise when attempting to culture a specific cell type. For example, while there are a few cell lines from insect fat body and at least one from the midgut, it may not be possible to obtain cell lines from these tissues from all insect species due to terminal differentiation and other factors. Also, researchers have desired cell lines from certain species, such as the honey bee, for which no success has been obtained. As in the early days of tissue culture, it is difficult to discern why negative results occur. However, as more is learned about the physiology and nutrition of various insects and tissues, we may get clues which will help solve these questions.The remaining chapters in this book will provide the reader with exciting uses for insect cell culture.
