植物经济谱能够阐述维管植物在资源获取和储存之间的权衡策略, 为理解生态位分化和物种共存机制等提供科学依据。该研究通过对武夷山49种木本植物的单叶面积(ILA)、比叶面积(SLA)、叶碳含量(LCC)、叶氮含量(LNC)和叶磷含量(LPC)等5个叶片性状以及根组织密度(RTD)、比根长(SRL)、比根面积(SRA)、根碳含量(RCC)、根氮含量(RNC)和根磷含量(RPC)等6个细根性状进行测定, 探讨木本植物叶片与细根经济谱是否存在以及常绿和落叶物种间的植物经济谱差异。结果表明: 沿着性状贡献率相对较大的PC1轴, 能够定义出叶经济谱(LES)、根经济谱(RES)和整株植物经济谱(WPES)。大部分常绿物种分布在经济谱保守的一侧, 而大部分落叶物种聚集在获取的一侧。此外, 叶片PC1、细根PC1和整株植物PC1的两两得分之间均存在显著正相关关系, 常绿和落叶物种具有共同的异速指数, 但不存在共同的异速常数。这些结果揭示了亚热带物种叶片与细根的策略遵循着WPES的协调整合, 表明叶片、细根以及整株植物之间是采取协同变化的资源策略, 而分布于经济谱两端的常绿和落叶物种则是通过不同的方式来构建WPES。 相似文献

Root respiration is a critical physiological trait involved in root resource acquisition strategies, yet it is less represented in root trait syndrome. Here we compiled a large dataset of root respiration associated with root chemical and morphological traits from 245 plant species. Our results demonstrated that root respiration correlated positively with root nitrogen concentration (RNC) and negatively with root tissue density (RTD) across and within woody and non‐woody species. However, the relationships between root respiration and specific root length (SRL) and root diameter (RD) were weak or even insignificant. Such root respiration–traits relationships were not completely in line with predictions by the root economics spectrum (RES). Furthermore, the principal component analysis showed that root trait syndrome was multidimensional. Root respiration was associated more strongly with the RNC‐RTD axis (the classical RES) than with the orthogonal SRL‐RD axis for woody species, but not for non‐woody species. Collectively, the linkages of root physiological, chemical, and morphological traits provide a better understanding of root trait covariation and root resource acquisition strategies. 相似文献

中国仓鼠卵巢细胞(CHO)流加培养生产单克隆抗体是目前主流培养方式,其中环境参数(pH和温度)和营养成分均影响细胞生长、碳氮源代谢和外源蛋白表达,是培养过程中关键的控制参数。采用实验设计(design of experiment,DOE)方法研究培养参数(温度、pH)对CHO细胞生长和抗CD20抗体表达的影响,建立营养限制型氨基酸流加策略,实现抗CD20抗体的高表达。结果表明,温度是影响蛋白质表达的显著因素,35℃有助于提高细胞密度和目标抗CD20抗体表达,而pH对抗CD20表达影响不显著,且温度和pH无交互作用,经DOE预测分析最佳培养条件是温度35℃和pH7.0。在该最佳培养条件下,在培养后期酪氨酸和半胱氨酸的浓度都低于0.1mmol/L。在培养的第2天通过补加1.5mmol/L酪氨酸和1mmol/L半胱氨酸避免营养限制,抗CD20抗体表达水平提高了24.1%,且对蛋白糖型无影响。 相似文献

摘要 目的:探讨不同比例输血策略对创伤-失血性休克大鼠的凝血指标、血浆代谢指标影响及作用机制。方法:选择Sprague- Dawley大鼠24只,制备创伤-失血性休克动物模型,采用血气分析仪检测凝血指标(PT及ATPP)血气分析指标(包括pH、PO2、PCO2、乳酸),采用比色法检测常见代谢酶活性,采用PCR法检测mRNA含量,记录并对比目标血压为80 mmHg时B、C、D三组大鼠的输血量。结果:与A组大鼠相比,B、D组大鼠的ATPP、PT水平明显较高(P<0.05),A组与C组间对比无统计学意义(P>0.05);与B组大鼠相比,D组大鼠的ATPP、PT水平明显较低(P<0.05)。与A组相比,B、C、D组大鼠的pH、PO2水平明显降低、PCO2及乳酸水平明显升高对比(P>0.05);与B组大鼠相比,C、D组大鼠的pH、PO2明较高,PCO2、乳酸明显较低(P<0.05);C、D组间对比无统计学意义(P>0.05)。四组大鼠的血浆肌酸激酶、天门冬氨酸基转移酶、碱性磷酸酶、丙氨酸氨基转移酶活动水平对比无统计学意义(P>0.05)。与A组相比,B、D组大鼠的血浆miR-24水平明显较高,肝脏FX mRNA水平明显较低(P<0.05),C组大鼠的血浆miR-24与肝脏FX mRNA与A组对比无统计学意义(P>0.05);与B组相比,D组大鼠的血浆miR-24水平明显较低,肝脏FX mRNA水平明显较高(P<0.05)。达到目标血压为80 mmHg时,输血量B组>C组>D组(F=133.139,P<0.001)。结论:新鲜冰冻血浆:红细胞比为1:1的输血策略较1:2.5的输血策略对创伤-失血性休克大鼠的复苏效果更好,可能与其降低大鼠的循环miR-24水平,提高肝脏FX mRNA水平表达有关。 相似文献

目的:研究抗HIV融合多肽CP32M的制备工艺,为其临床前试验奠定基础。方法:采用固-液相混合策略规模合成目标肽,用离子交换色谱、反相高效液相色谱对粗肽进行纯化。结果:获得了利于提高合成及片段缩合效率的3条优选片段,CP32M粗品经DEAE离子交换色谱及C18反相纯化后纯度高于98%。结论:CP32M按3条片段(Ac-1-9-OH、Fmoc-10-21-OH、H-22-32-NH2)划分,可显著提高片段合成及缩合效率,DEAE阴离子交换色谱能除去大部分杂质,提高了第二步C18反相色谱纯化效率。 相似文献

In this study, we numerically investigated to what extent introducing resting-egg dynamics would stabilize simple Daphnia–algae consumer–resource models. In the models, the density of viable resting eggs was explicitly expressed, and we assumed
that zooplankton produced resting eggs seasonally or in response to food deficiency and that resting eggs hatched seasonally.
The models predicted that, although the paradox of enrichment was not completely resolved (i.e., the system was destabilized
by eutrophication), we found the following conditions under which the stabilizing effects of resting eggs would be significantly
large: (1) resting eggs are produced seasonally (rather than in response to food deficiency), (2) the annual average allocation
ratio to resting eggs is large, and (3) the annual average hatching rate of resting eggs is low. The results suggest that
resting-egg dynamics can significantly reduce the paradox of enrichment within the biologically meaningful parameter space

The relationship between adult and offspring size is an important aspect of reproductive strategy. Although this filial relationship has been extensively examined in plants and animals, we currently lack comparable data for protists, whose strategies may differ due to the distinct ecological and physiological constraints on single‐celled organisms. Here, we report measurements of adult and offspring sizes in 3888 species and subspecies of foraminifera, a class of large marine protists. Foraminifera exhibit a wide range of reproductive strategies; species of similar adult size may have offspring whose sizes vary 100‐fold. Yet, a robust pattern emerges. The minimum (5th percentile), median, and maximum (95th percentile) offspring sizes exhibit a consistent pattern of increase with adult size independent of environmental change and taxonomic variation over the past 400 million years. The consistency of this pattern may arise from evolutionary optimization of the offspring size‐fecundity trade‐off and/or from cell‐biological constraints that limit the range of reproductive strategies available to single‐celled organisms. When compared with plants and animals, foraminifera extend the evidence that offspring size covaries with adult size across an additional five orders of magnitude in organism size. 相似文献

Defective human leukocyte antigen (HLA) class I expression in malignant cells facilitates their escape from destruction by CD8+ cytotoxic T lymphocytes. In this study, a post-translational mechanism of HLA class I abnormality that does not involve defects in the HLA subunits and antigen processing machinery components was identified and characterized. The marked HLA class I downregulation phenotype of a metastatic carcinoma cell line can be readily reversed by trypsin, suggesting a masking effect by serine protease-sensitive HLA class I-interacting factors. Co-immunoprecipitation, combined with LC–tandem mass spectrometry and immunoblotting identified these factors as cytokeratin (CK) 8 and its heterodimeric partners CK18 and CK19. Ectopic CK8/18 or CK8/19 expression in HEK293 cells resulted in surface CK8 expression with an HLA class I downregulation phenotype, while redirecting CK8/18 and CK8/19 to the endoplasmic reticulum (ER) had no such effect. This observation and the failure to constrain CK8/18 and CK8/19 membrane trafficking by an ER-Golgi transport inhibitor suggested an ER-independent route for CK8 access to HLA class I molecules. Monoclonal antibody mapping revealed a potential CK8 blockade of HLA class I-CD8 and -TCR contacts. These findings, along with the emerging role of cell surface CK8 in cancer metastasis, may imply a dual strategy for tumor cell survival in the host. 相似文献
