This study assessed the feeding preference of larvae of Triplectides sp. (Trichoptera, Leptoceridae) exposed to leaves of native (Hoffmannia dusenii Standley, 1931) and exotic (Eucalyptus globulus Labillardiere, 1799) trees. We hypothesized that, regardless of the origin of the leaves, larval preference is determined mainly by leaf anatomy and quality. Leaves from both species were conditioned with and without nutrient enrichment (NPK), and the four food items were offered in paired combinations to 162 larvae. Larval preference varied according to leaf combinations. In treatments containing both species, larvae preferred to feed on H. dusenii because of softer tissues and anatomical structure. The only exception was the treatment containing discs of enriched E. globulus and non-enriched H. dusenii where enhanced microbial activity on enriched leaves provided a softer resource to shredders. Our results corroborate the initial hypothesis and suggest that introduction of exotic leaves and changes in nutrient availability may affect shredder activity in streams and, consequently, organic-matter processing and ecosystem functioning.  相似文献

The unicellular green alga Chlamydomonas reinhardtii has a haplontic life cycle, and forms diploid zygotes for reproduction. The zygospore, a sporulating zygote, begins germination in response to light signals, generating haploid progenies and inducing several cell-biological events; e.g., DNA synthesis and meiotic division, successively. Their regulatory mechanisms remain largely unknown, so we focused on the early stages of germination and analyzed the dynamics of gene expression associated with the germination process. The gene expression levels of zygospores at 1 and 6 h after light exposure were analyzed by a next-generation sequencing platform, the 454 GS Junior. At 6 h, the photosynthesis pathway, including its antenna proteins and two methionine metabolism-related genes (methionine synthase and sulfite reductase), were up-regulated compared to 1 h after light exposure. Meanwhile, three uncharacterized genes that contained an antibiotic biosynthesis monooxygenase domain and an HSP20/alpha crystallin family protein were specifically expressed at 1 h after light exposure. These gene expressions were also verified by quantitative real-time PCR analysis. These results suggest that the photosynthesis and methionine synthesis pathways, both of which occur in the chloroplast, are activated in zygospores at around 6 h after light exposure, and that some polyketides and/or a small heat shock protein may be related to the initiation of zygospore germination.  相似文献

The purpose of this study was to develop a silica nanoparticle-based immunosensor with laser-induced fluorescence (LIF) as a detection system. The proposed device was applied to quantify the immunoreactive trypsin (IRT) in cystic fibrosis (CF) newborn screening. A new ultrasonic procedure was used to extract the IRT from blood spot samples collected on filter papers. After extraction, the IRT reacted immunologically with anti-IRT monoclonal antibodies immobilized on a microfluidic glass chip modified with 3-aminopropyl functionalized silica nanoparticles (APSN–APTES-modified glass chips). The bounded IRT was quantified by horseradish peroxidase (HRP)-conjugated anti-IRT antibody (anti-IRT–Ab) using 10-acetyl-3,7-dihydroxyphenoxazine (ADHP) as enzymatic mediator. The HRP catalyzed the oxidation of nonfluorescent ADHP to highly fluorescent resorufin, which was measured by LIF detector, using excitation lambda at 561 nm and emission at 585 nm. The detection limits (LODs) calculated for LIF detection and for a commercial enzyme-linked immunosorbent assay (ELISA) test kit were 0.87 and 4.2 ng ml−1, respectively. The within- and between-assay variation coefficients for the LIF detection procedure were below 6.5%. The blood spot samples collected on filter papers were analyzed with the proposed method, and the results were compared with those of the reference ELISA method, demonstrating a potential usefulness for the clinical assessment of IRT during the early neonatal period.  相似文献

This study investigated whether shelter (presence or absence) affected the frequency of fin damage in Atlantic salmon Salmo salar exposed to feed restrictions (0·73 or 0·33% of body mass day?1). The presence of shelter had a positive effect on the pectoral fins at both feed ration levels and on the dorsal fin at the higher ration level but it had a negative effect on survival. The reduced feed rations resulted in fish of the same size and nutritional status as wild fish. The provision of shelter has potential to mitigate the negative effects of feed restrictions on fin quality, but the optimal shelter design requires some additional investigation.  相似文献

Three young northern temperate forest communities in the north‐central United States were exposed to factorial combinations of elevated carbon dioxide (CO2) and tropospheric ozone (O3) for 11 years. Here, we report results from an extensive sampling of plant biomass and soil conducted at the conclusion of the experiment that enabled us to estimate ecosystem carbon (C) content and cumulative net primary productivity (NPP). Elevated CO2 enhanced ecosystem C content by 11%, whereas elevated O3 decreased ecosystem C content by 9%. There was little variation in treatment effects on C content across communities and no meaningful interactions between CO2 and O3. Treatment effects on ecosystem C content resulted primarily from changes in the near‐surface mineral soil and tree C, particularly differences in woody tissues. Excluding the mineral soil, cumulative NPP was a strong predictor of ecosystem C content (r2 = 0.96). Elevated CO2 enhanced cumulative NPP by 39%, a consequence of a 28% increase in canopy nitrogen (N) content (g N m?2) and a 28% increase in N productivity (NPP/canopy N). In contrast, elevated O3 lowered NPP by 10% because of a 21% decrease in canopy N, but did not impact N productivity. Consequently, as the marginal impact of canopy N on NPP (?NPP/?N) decreased through time with further canopy development, the O3 effect on NPP dissipated. Within the mineral soil, there was less C in the top 0.1 m of soil under elevated O3 and less soil C from 0.1 to 0.2 m in depth under elevated CO2. Overall, these results suggest that elevated CO2 may create a sustained increase in NPP, whereas the long‐term effect of elevated O3 on NPP will be smaller than expected. However, changes in soil C are not well‐understood and limit our ability to predict changes in ecosystem C content.  相似文献

目的:分析心脏瓣膜置换同期行双极射频消融改良迷宫术治疗房颤的术后中远期疗效及其影响因素。方法:选取2007年7月至2010年12月于我院行心脏瓣膜置换同期行双极射频消融改良迷宫术治疗房颤的72例患者,术后随访3至6年,获得患者术后十二导联心电图、24 h动态心电图和心脏彩超结果,并且记录术后药物应用、相关并发症、心功能恢复情况及生活质量是否改善等情况。根据心电图结果,将患者分为窦性心律组与非窦性心律组,应用统计学方法比较两组患者术前相关因素是否有差异。结果:截止至随访终点,有效随访的68例患者中窦性心律维持率为63.2%。单因素分析显示术前房颤病程、术前左室射血分数、是否合并三尖瓣成形或置换是影响手术效果的影响因素,多因素分析显示术前房颤病程、合并三尖瓣成形或置换是影响手术效果的危险因素。结论:心脏瓣膜置换同期行双极射频消融改良迷宫术治疗房颤的中晚期疗效好,术后并发症少,能够有效防止血栓栓塞,术前房颤病程长、合并三尖瓣成形或置换的患者术后中远期效果相对较差。  相似文献

It has been postulated that implementing appropriate cognitive challenges in the housing of captive animals allows them to use their cognitive skills, and to learn to control certain aspects of their environment. This cognitive activity is thought to be a major source of positive emotions and a prerequisite for psychological well-being of the animal. We studied the use of an automated learning device (LD) by group-housed dwarf goats. To determine if the animals seek cognitive challenges, we investigated whether the goats continued to operate the LD to get a reward when the same amount of reward was available concurrently without additional cognitive effort. In a first training period (W), a group of 10 dwarf goats were trained to press a button at a waterer (WA) to get 35 ml of drinking water. In two following periods (P1 and P2), the goats learned to discriminate two different sets of visual shapes at the LD to get the same amount of water as a reward. The WA was closed in these periods. Finally, in two experimental periods (P2r and P3), water was concurrently available at the WA and the LD for five days. We analysed the use of the LD when the WA was accessible: (a) while the goats were presented with the discrimination problem that they had learned previously (P2r); and (b) while they were given a new problem to learn (P3). The total number of button presses that triggers the delivery of water (presses at the WA and/or successful presses at the LD) was stable throughout the study, whether water was delivered only at the waterer (WA) or, after successful shape discrimination, at the LD, or if it was concurrently available at both devices. When water was concurrently available at the WA and the LD (P2r, P3), the goats constantly directed 1/3 of all daily button presses to the LD. By doing so, they gained 23.0% (P2r) and 13.8% (P3) of daily drinking water at the LD because of differing learning success. Goats showed large individual differences in the number of button presses at the LD in P2r and P3. We found a strong correlation between individual learning success and the use of the LD in P2r (rs = 0.75; P = 0.025) and P3 (rs = 0.76; P = 0.036). Results indicate that cognitive challenges, and especially successful coping with the challenge, could have at least moderate intrinsic reinforcing properties beyond the reward.  相似文献
