编码天麻抗真菌蛋白cDNA的分子克隆

用重组ＤＮＡ 技术研究了编码天麻抗真菌蛋白（ＧＡＦＰ）的基因,从天麻（Ｇａｓｔｒｏｄｉａ ｅｌａｔａＢｌ．）块茎中提取Ｐｏｌｙ（Ａ） ｍ ＲＮＡ 后合成ｃＤＮＡ,构建成表达型ｃＤＮＡ 文库,用纯化的蛋白质探针通过免疫筛选找出对应的ｃＤＮＡ 克隆。在进一步证明所选用的ｃＤＮＡ 克隆含有重组的λ－ｐｈａｇｅ ＤＮＡ 后,提取和纯化含有插入片段重组子的ＤＮＡ,用Ｅｃｏ ＲＩ酶切分析可见插入片段。已分离出编码天麻抗真菌蛋白的基因     

A serotype-specific epitope of dengue virus 1 identified by phage displayed random peptide library

Abstract From a panel of monoclonal antibodies of dengue viruses, a serotype-specific epitope of dengue virus 1 was screened from a random peptide library displayed on phage. The epitope was the determinant reactive with monoclonal antibody 15F3-1 that was specific to dengue 1. The screening was monitored by a dot blotting procedure, and after three rounds of screening a consensus motif, HRYSWK, was found. This sequence matches the sequence HKYSWK, corresponding to the amino acid residues 885–890 of polyprotein or residues 111–116 of the non-structural protein 1 of dengue virus serotype 1. The linear epitope was confirmed by testing the antigenicity of chemically synthesized 8-branched peptide.     

从cDNA文库中筛选分析阳性克隆的简便方法

cDNA文库中阳性克隆的传统筛选分析法既费时又费力.利用微波炉加热的方法,简化了原位杂交中噬菌斑裂解、DNA变性与固定的程序;进一步运用PCR扩增技术,特异扩增克隆载体中插入的cDNA片段,加快了阳性克隆分析的进程.     

The human genome contains multiple sequences of varying homology to mouse mammary tumour virus DNA

Sequences related to the mouse mammary tumour virus (MuMTV) DNA were isolated from a genomic library of human DNA by screening under conditions of relaxed stringency. It is estimated that there are in the order of 50 MuMTV-like sequences per haploid genome and that the homology between the different human sequences and MuMTV varies by 15%.     

Stable integration of foreign DNA into the chromosome of the cyanobacterium Synechococcus R2

The blue-green alga, Synechococcus R2, is transformed to antibiotic resistance by chimeric DNA molecules consisting of Synechococcus R2 chromosomal DNA linked to antibiotic-resistance genes from Escherichia coli. Chimeric DNA integrates into the Synechococcus R2 chromosome by homologous recombination. The efficiency of transformation, as well as the stability of integrated foreign DNA, depends on the position of the foreign genes relative to Synechococcus R2 DNA in the chimeric molecule. When the Synechococcus R2 DNA fragment is interrupted by foreign DNA, integration occurs through replacement of chromosomal DNA by homologous chimeric DNA containing the foreign insert; transformation is efficient and the foreign gene is stable. Mutagenesis in some cases attends integration, depending on the site of insertion. Foreign DNA linked to the ends of Synechococcus R2 DNA in a circular molecule, however, integrates less efficiently. Integration results in duplicate copies of Synechococcus R2 DNA flanking the foreign gene and the foreign DNA is unstable. Transformation in Synechococcus R2 can be exploited to modify precisely and extensively the genome of this photosynthetic microorganism.     

大鼠肾脏组织cDNA文库的制备和葡萄糖转运蛋白cDNA克隆的筛选

 大鼠的肾脏组织经匀浆后,提取纯化总RNA和mRNA合成cDNA,进行甲基化,加人工接头,最后连入载体(λgt11)和外壳蛋白包装步骤制成肾脏组织的cDNA文库。插入载体的cDNA长度为0.5kb到8kb之间。经反复稀释,测定出该文库的效价为1.5×10~7pfu/mL。     

A rapid procedure for the construction of PCR cDNA libraries from small amounts of plant tissue

We describe a general method for the preparation of λZAP II cDNA libraries from very small amounts (<50 mg) of plant tissue. We have achieved this by combining an efficient method for RNA extraction with a modified PCR protocol for the synthesis and amplification of cDNA. Using this protocol we have found it possible to generate cDNA libraries containing more than 10⁶ clones from as little as 1 μg of total RNA.     

鲑鱼生长激素cDNA的分子克隆和序列分析

从太平洋切奴克鲑鱼(Pacific Chinook Salmon,Oncorthychus tschawytscha)垂体poly(A)~+ RNA构建cDNA文库。按照鲑鱼生长激素(sGH)部分氨基酸序列合成两个寡聚脱氧核苷酸探针,它们分别与编码第1—7和第166—172氨基酸序列互补。用探针筛查cDNA文库,得到了完整的sGH cDNA克隆。cDNA序列已测定,包括编码210个氨基酸的编码序列。其中含有22个氨基酸的信号肽序列和188个氨基酸的成熟GH序列。该克隆还包括了5'端和3'端非翻译区,分别为72个和438个碱基对长。与Chum鲑鱼比较表明,核酸序列和氨基酸序列的同源性分别为97%和99%。     

粪产碱菌(Alcaligenes faecalix)基因文库的构建和nifH基因同源顺序的克隆

采用λ噬菌体置换型载体EMBL4,构建了Alcaligenes faecalis A-15 H1菌株总DNA的基因文库。用Sau3AⅠ限制酶完成部分酶切,取13—20kb大小的片段进行克隆。载体DNA经BamHⅠ和SaiⅠ完全双酶切,左右臂“退火”形成左右臂载体分子后再与外源片段连接。左右臂载体分子与外源片段按照1:1的分子比进行体外连接。用E.coli BHB2688和E.coli BHB2690制备的包装抽提物进行体外包装,所得基因文库效价测定为1.2×10~6 pfu,远远超过理论上所需的库容量。以nif H基因作为探针,经3轮噬菌斑原位杂交,从基因文库中筛选出含有其同源顺序的克隆,并得到了梯度点杂交的验证。对所得重组噬菌体克隆之一进行Southern转移杂交,结果证实,其3.5kb的EcoRⅠ酶切片段为nif H阳性杂交条带。将其克隆到质粒pUC19 DNA上后,转入受体菌JM101中。再次经Southern转移杂交,证明所得重组质粒克隆(pAFH)含有粪产碱菌中的与nifH基因有同源顺序的片段。
