Fourier-transform infrared (FTIR) spectroscopy was carried out on single colonies of Pediastrum duplex present in air-dried preparations of mixed phytoplankton samples isolated from a eutrophic freshwater lake. FTIR absorption spectra had 12 distinct bands over the wavenumber range 3300–900?cm?1 which were tentatively assigned to a range of chemical groups, including –OH (residual water, wavenumber 3299?cm?1), –CH2 (lipid, 2924), –C=O (cellulose, 1739), amide (protein, 1650 and 1542), >P=O (nucleic acid, 1077) and –C–O (starch, 1151 and 1077). Measurement of band areas identified residual water, protein and starch as the major detectable constituents. Areas of single bands and combined bands of –CH2, –C–O and >P=O species normalized to protein (to correct for differences in specimen hydration and thickness) showed wide variation between colonies, indicating environmental heterogeneity. Correlation analysis demonstrated close statistical associations between different molecular species. Particularly high levels of correlation between bands 3/4 (CH2), 6/7 (amide) and 8/9 (–CH3) was consistent with their joint origin from the same molecular species. The isolation of bands 11 and 12 in the correlation pattern was confirmed by factor analysis, suggesting that variation in the level of starch is statistically unrelated to other macromolecules being monitored. The use of FTIR spectroscopy to characterize an algal micro-population within mixed phytoplankton has potential for future studies on biodiversity and environmental interactions at the species level. 相似文献

In contrast to the many investigations on possible relationships between climate and geographical distributions in macroalgae, there are almost no similar studies regarding microalgae. In this study we consider the potential influence of temperature on patterns of distribution of six Cosmarium strains isolated from various climate zones that have been cultured long-term (> 15 years) in a relatively low temperature–low light regime. Growth and photosynthetic parameters, obtained from PAM fluorometry, were used to estimate the physiological characteristics of the strains during and after various temperature treatments. Acclimation to constant temperature and light conditions tended to affect photosynthetic parameters more than algal growth characteristics. However, all of the Cosmarium strains demonstrated physiological responses that were consistent with their source location under both low and high temperature conditions, confirming that such responses are genetically preserved. The Cosmarium strains displayed photosynthetic capacities and levels of the onset of saturation that repeatedly exceeded values recorded for other microalgae and seaweeds, indicating that these desmid strains are adapted to high light. This observation, as well as the relatively high growth temperature optima for all of the Cosmarium strains, provides some support for Coesel's hypothesis on the origin of desmids in the tropical zone. Interestingly, the Cosmarium strains used in this study demonstrated not only adaptive characteristics in accordance with the temperature prevailing at their sampling sites, but also with regard to their evolutionary origin. 相似文献

Chromera velia is a recently discovered, photosynthetic, free-living alveolate that is the closest free-living relative to non-photosynthetic apicomplexan parasites. Most plastids, regardless of their origin, have membranes composed chiefly of two galactolipids, mono- and digalactosyldiacylglycerol (MGDG and DGDG, respectively). Because of the hypothesized shared red algal origin between the plastids of C. velia and dinoflagellates, our primary objectives were to examine how growth temperature affects MGDG and DGDG composition via positive-ion electrospray/mass spectrometry (ESI/MS) and positive ion/electrospray/mass spectrometry/mass spectrometry (ESI/MS/MS), and to examine galactolipid biosynthetic genes to determine if shared ancestry translates into shared MGDG and DGDG composition. When growing at 20°C, C. velia produces eicosapentaenoic acid-rich 20:5(n-3)/20:5(n-3) (sn-1/sn-2) MGDG and 20:5(n-3)/20:5(n-3) DGDG as its primary galactolipids, with relative percentage compositions of approximately 35 and 60%, respectively. At 30°C these are lessened by approximately 5 and 8%, respectively, by the corresponding production of 20:5/20:4 forms of these lipids. The presence of 20:5 at the sn-1 position is similar to what has been observed previously in a cluster of peridinin-containing dinoflagellates, but the presence of 20:5(n-3) at the sn-2 position is extremely rare. Thus, the forms of MGDG and DGDG in C. velia displayed similarities and differences to what has been observed in peridinin-containing dinoflagellates, such as Lingulodinium polyedrum, which produces 20:5/18:5 and 20:5/18:4 as the major forms of MGDG and DGDG. We develop conceptual models from the galactolipids observed and galactolipid-relevant gene annotations to explain the presence of polyunsaturated fatty acid-containing MGDG and DGDG in both L. polyedrum and C. velia. 相似文献

A simplified procedure is described for the extraction and derivatization of low molecular weight carbohydrates from intact, freeze-dried samples of marine brown, green and red algae prior to their estimation by gas-liquid chromatography. This technique, which involves direct extraction into hot dimethyl sulphoxide (DMSO) (100°C, 18 h), proved to be at least as effective as the widely used method employing 80% (v/v) ethanol-water as an extractive solvent and has the dual advantage of being considerably faster and involving fewer preparative steps. 相似文献

For the first time, a comprehensive assessment of Mesophyllum species diversity and their distribution in Atlantic Europe and the Mediterranean Sea is presented based on molecular (COI-5P, psbA) and morphological data. The distribution ranges were redefined for the four species collected in this study: M. alternans, M. expansum, M. macroblastum and M. sphaericum. Mesophyllum sphaericum, which was previously known only from a single maerl bed in Galicia (NW Spain), is reported from the Mediterranean Sea. The known range of M. expansum (Mediterranean and Macaronesia) was extended to the Atlantic Iberian Peninsula. The occurrence of M. alternans was confirmed along the Atlantic French coast south to Algarve (southern Portugal). Mesophyllum lichenoides was only recorded from the Atlantic, whereas M. macroblastum appears to be restricted to the Mediterranean Sea. A positive correlation was observed between maximum Sea Surface Temperature (SSTmax) and the depth at which M. expansum was collected, suggesting that this species may compensate for higher SST by growing in deeper habitats where the temperature is lower. The latter indicates that geographic shifts in the distribution of coastal species as a result of global warming can possibly be mitigated by changes in the depth profile at which these species occur. Mesophyllum expansum, an important builder of Mediterranean coralligenous habitats, may be a good target species to assess its response to climate change. 相似文献

Using transmission electron microscopy, the process of cell wall development following cytokinesis is described. Three distinct components appear to be localized in the wall and these are discussed in relation to previous work. In older cells the walls are stratified and the origin of the layers is described. Algae biomass is a potential raw material for the production of biofuels and other chemicals. In this study, biomass of the marine algae, Ulva lactuca, Gelidium amansii,Laminaria japonica, and Sargassum fulvellum, was treated with acid and commercially available hydrolytic enzymes. The hydrolysates contained glucose, mannose, galactose, and mannitol, among other sugars, at different ratios. The Laminaria japonica hydrolysate contained up to 30.5% mannitol and 6.98% glucose in the hydrolysate solids. Ethanogenic recombinant Escherichia coli KO11 was able to utilize both mannitol and glucose and produced 0.4 g ethanol per g of carbohydrate when cultured in L. japonica hydrolysate supplemented with Luria-Bertani medium and hydrolytic enzymes. The strategy of acid hydrolysis followed by simultaneous enzyme treatment and inoculation with E. coli KO11 could be a viable strategy to produce ethanol from marine alga biomass. 相似文献

Larger benthic Foraminifera (LBF) are ecologically important coral reef protists that harbour a large diversity of symbionts from at least four algal phyla. In this study the photosynthetic plasticity of different endosymbiontic algae found within LBF was investigated using pulse amplitude modulated (PAM) fluorometry. Maximum quantum efficiencies of photosystem II (Fv/Fm) obtained from foraminiferal specimens directly after field collection indicated several pronounced differences between species containing endosymbionts from different algal phyla and, to a lesser extent, also varied between species that contain the same phyla of endosymbiontic algae. Foraminiferal species retaining functional chloroplasts and rhodophyte-bearing species had distinctly lower Fv/Fm, than LBF with dinoflagellates, diatoms or chlorophytes. A laboratory experiment was conducted over 48 h exposing species occurring in high- (photophilic), medium- and low-light (sciaphilic) environments to three manipulated light levels. Photophysiological responses were monitored by measuring Fv/Fm at regular intervals and rapid light curves (RLCs) at the end of the experiment. This experiment demonstrated oscillation of maximum quantum efficiencies according to the light-dark cycle. Changes in the shape of the RLCs (e.g., higher α and lower Ek under low light conditions) indicated that photosynthetic plasticity allows LBF to acclimatise rapidly (< 48 h) to different light conditions. 相似文献
