The cytosolic free calcium was measured with Fura-2 in single prothoracic gland cells of Galleria larvae. During the last two larval instars calcium concentration correlated with ecdysone secretion by the glands. Addition of prothoracicotropic hormone (PTTH) from brains of Galleria larvae to prothoracic glands in vitro induced a significant increase in calcium in the gland cells. This effect of PTTH was abolished by removal of extracellular calcium, or by the addition of lanthanum or of the calcium channel antagonists nicardipine and verapamil. The calcium channel agonist Bay K 8644 evoked an increase in intracellular calcium. TMB-8, an inhibitor of intracellular calcium mobilization, did not block the PTTH-stimulated rise in calcium concentration or ecdysone production, indicating that intracellular calcium stores are not involved in the calcium-mediated ecdysone synthesis. Moreover, PTTH seems to exert its action by influencing dihydropyridine-sensitive calcium channels in the plasma membrane. © 1996 Wiley-Liss, Inc.  相似文献

In the penultimate-larval instar, the total volume of the prothoracic gland and the activities of some oxidative mitochondrial enzymes (cytochrome oxidase, NADH: cytochrome c oxidoreductase, succinate: cytochrome c oxidoreductase) undergo cyclic variations associated with larval growth. These specifically larval-larval growth cycles are absent in the prothoracic glands of normal last-instar larvae. Here the cycles can be induced artificially by implantation of brain or corpora cardiaca-allata complexes or, by exogenous application of juvenile hormone. The smallest size of the prothoracic gland in relation to the size of the body, as well as the minimal activity of all the three mitochondrial enzymes in the gland, have been found exactly at the moment of the pre-pupal peak of ecdysteroid in the body. The possibility that the prothoracic glands alone can synthetize ecdysteroid during the peak is questioned.  相似文献

The evolutionary process leading to the emergence of viviparity in Squamata consists of lengthening the period of egg retention in utero coupled with marked reduction in the thickness of the eggshell. We used light microscopy and scanning electron microscopy to study uterine structure during the reproductive cycle of oviparous and viviparous females of the reproductively bimodal Lacerta vivipara. We compared the structure of the uterine shell glands, which secrete components of the eggshell, during preovulatory and early gestation phases of the reproductive cycle and also compared histochemistry of the eggshells. The uterine glands of both reproductive forms undergo considerable growth within a period of a few weeks during folliculogenesis and vitellogenesis preceding ovulation. The majority of the proteinaceous fibers of the shell membrane are secreted early in embryonic development and the uterine glands regress shortly thereafter. This supports previous observations indicating that, in Squamata, secretion of the shell membrane occurs very rapidly after ovulation. The most striking differences between reproductive modes were larger uterine glands at late vitellogenesis in oviparous females, 101 microm compared to 60 microm in viviparous females, and greater thickness of the shell membrane during early gestation in oviparous females (52-73 microm) compared to viviparous females (4-8 microm). Our intraspecific comparison supports the conclusions of previous studies that, prior to ovulation, the uterine glandular layer is less developed in viviparous than in oviparous species, and that this is the main factor accounting for differences in the thickness of the shell membrane of the two reproductive forms of squamates.  相似文献

Glucocorticoid (GC) hormones rise in response to stressors, including natural events including weather or predator presence, and human activities, such as hunting, scientific research or recreational visits. However, because blood sampling itself causes stress and is dangerous or even impossible in some wildlife species, feedback-free methods for GC determination are needed to assess stress in these animals. Faecal GC analyses have thus gained interest. Here, we validate a non-invasive method to estimate the physiological stress in the Upland goose Chloephaga picta. An adrenocorticotropin hormone (ACTH) challenge was conducted in captive adults (female and male), and droppings were collected before, during and after the experiment. Corticosterone metabolite (CM) secretion in response to the ACTH challenge was measured with several enzyme immunoassays (EIA) to find the most appropriate test. We used CM levels during the periods before and after the experiment as control data. An EIA for 11-oxoetiocholanolone achieved the highest response to the ACTH challenge and also reflected a stress response to unfamiliar environment. Furthermore, CM concentrations of dry samples were highly correlated with the corresponding non-dried (frozen) samples. The data suggest that this method is appropriate to measure the stress in Upland geese, and that samples can be stored either frozen or dry form.  相似文献

Summary A comparative study was undertaken on the innervation of mucous and granular glands in frog skin. Results obtained by the Falck-Hillarp fluorescence technique and cholinesterase staining indicated that both types of glands receive exclusively adrenergic innervation. Electron microscopy was used to investigate the innervation pattern at the ultrastructural level. The distribution of nerve terminals was found to differ in the two types of glands. In the mucous gland, terminals were found at a distance of about 0.5 m from the basement membrane but never within the gland parenchyma. In the granular gland, the terminals were located between smooth muscle cells and also in direct contact with the secretory epithelium but never outside the basement membrane.This work was carried out in part at King Gustaf V:s Forskningsinstitut, Stockholm, and was supported by a grant from Karolinska Institutet  相似文献

The majority of ticks in the family Ixodidae secrete a substance anchoring their mouthparts to the host skin. This substance is termed cement. It has adhesive properties and seals the lesion during feeding. The particular chemical composition and the curing process of the cement are unclear. This review summarizes the literature, starting with a historical overview, briefly introducing the different hypotheses on the origin of the adhesive and how the tick salivary glands have been identified as its source. Details on the sequence of cement deposition, the curing process and detachment are provided. Other possible functions of the cement, such as protection from the host immune system and antimicrobial properties, are presented. Histochemical and ultrastructural data of the intracellular granules in the salivary gland cells, as well as the secreted cement, suggest that proteins constitute the main material, with biochemical data revealing glycine to be the dominant amino acid. Applied methods and their restrictions are discussed. Tick cement is compared with adhesives of other animals such as barnacles, mussels and sea urchins. Finally, we address the potential of tick cement for the field of biomaterial research and in particular for medical applications in future.  相似文献

The Cossidae are an economically important family of moths consisting of six subfamilies and more than 700 species globally. Their larvae are often of serious concern to forestry and horticulture in that they are known to bore in the branches and trunks of a wide range of trees and shrubs. Cossulinae is one of the six subfamilies of Cossidae and is restricted to the New World. As a result of this revision, the Costa Rican Cossulinae has been found to consist of five genera and 18 species. The phylogenetic relationships of all Cossulinae genera were analysed using the computer programs WinClada and NONA. Glandular organs new to Cossidae and the Lepidoptera have also been discovered. Four genera, Biocellata gen. nov. , Magulacra gen. nov. , Simplicivalva gen. nov. and Spinulata gen. nov. , and the following nine species are described as new: Biocellata bifida sp. nov. , Biocellata davisorum sp. nov. , Cossula buspina sp. nov. , Cossula duplexata sp. nov. , Cossula longirostrum sp. nov. , Cossula minutiloba sp. nov. , Simplicivalva ampliophilobia sp. nov. , Spinulata oblongata sp. nov. and Spinulata quasivinnea sp. nov. © 2008 The Linnean Society of London, Zoological Journal of the Linnean Society, 2008, 154 , 222–277.  相似文献

The histology and ultrastructure of the salivary glands of male and female H.a. anatolicum ticks have been examined m unfed and feeding ticks with special emphasis on aspects related to the feeding process. The salivary glands of H.a. anatolicum consisted of three types of acinus (acinus I, II and III) in females and an additional type IV acinus in males. The type I acinus was agranular and showed slight morphologic changes during feeding. The presence of cells with ultrastructural features characteristic of epithelia involved in the secretion of hyperosmotic fluids supports the hypothesis that these acini secrete hygroscopic saliva during questing stages to absorb water from an unsaturated atmosphere. There were five granular cell types (a, b, c1c3) in type II acinus, three granular cell types (d, e, and f) in type III acinus, and one granular cell type (g) in type IV acinus. The cells a, d and e secreted most of their granules early in feeding and are considered to be cement precursors. The b and c cells appeared to synthesise and secrete their products throughout feeding and so are likely to secrete anticoagulants, enzymes and other pharmacologically active agents required during feeding. The interstitial cells, which were insignificant in acinar types II, III and IV of unfed ticks, became more distinct during feeding. The type III acinus in females showed remarkable cell transformations, during the course of feeding. The ablumenal interstitial cells of type III acinus, in females formed a basal labyrinth of extraordinary complexity by interdigitating with the basolateral membranes of transformed f cells to form a network of extracellular channels to excrete fluid during feeding. There was an enormous increase in the secretory granules of g cells as the feeding advanced. The secretory granules were released by a process of exocytosis, by direct fusion with the apical membranes and through channels connecting several granules.  相似文献

Synopsis Spermatozoa stored in oviducal glands of sharks sampled off the North American east coast were revealed by viewing stained tissue sections using light microscopy. Of eleven species surveyed, sperm were found in nine:Alopias vulpinus, Lamna nasus, Carcharhinus obscurus, Carcharhinus plumbeus, Galeocerdo cuvieri, Prionace glauca, Rhizoprionodon terraenovae, Sphyrna lewini andSphyrna tiburo. Three insemination patterns are proposed to account for differences noted in these findings: (1) non-storage/immediate insemination for sharks such asLamna nasus; (2) short-term storage/delayed insemination as found in sharks in which ovulation is prolonged over weeks or months such asRhizoprionodon terraenovae, and (3) long-term storage/repeated insemination, a characteristic of nomadic sharks such asPrionace glauca andCarcharhinus obscurus which can store sperm in specialized tubules for months to years.  相似文献
