To study the effect of special brain area regional cerebral blood flow (rCBF) abnormal perfusion on learning and memory function
and its molecular mechanism, 64 adult male healthy Sprague-Dawley (SD) rats were randomly divided into two groups, the false
operation group (control group) and the operation group (model group). After surgical operation, the operation group undertook
bilateral common carotid artery permanent ligation, while the other group did not. Learning and memory function were measured
by Y-maze at 4 h, 8 h, 24 h and 3 d after surgical operation, respectively. The rCBF of the right frontal lobe and hippocampus
was also detected by the PerifluxPF model laser Doppler flowmetry, and the expressions of c-fos or c-jun or Bcl-2 and Bax were also measured by immune histochemistry S-P method accordingly. Results showed that the rCBF of the right frontal lobe
and hippocampus in the operation group was significantly lower than that in the false operation group (P < 0.05). The learning indexes, error number (EN), day of reach standard and total reaction time (TRT) in the operation group,
were significantly higher than that in the false operation group (P < 0.05). However, the initiative evasion rate in the operation group was significantly lower than that in the false operation
group. The study also found that the rCBF was relatively more, the indexes (EN, the day of reach standard and TRT) relatively
fewer, but the initiative evasion rate and the memory keeping rate were relatively more. The positive expression and the average
absorbency of Fos and Jun in the operation group were significantly higher than that in the false operation group (P < 0.05). Furthermore, Bax and Bcl-2 positive cells were all increased over time in the operation group, and the expression
ratio of Bax/Bcl-2 in the operation group was significantly higher than that in the false operation group (P < 0.01). In conclusion, rCBF decrease can impair the learning and memory function in rats, which may be related to the increase
of the expression ratio of c-fos or c-jun or Bcl-2 or Bax in the frontal cortex and hippocampus. 相似文献

The functional relationship between correct response probability and response time is investigated in data sets from Rubin,
Hinton and Wenzel, J Exp Psychol Learn Mem Cogn 25:1161–1176, 1999 and Anderson, J Exp Psychol [Hum Learn] 7:326–343, 1981.
The two measures are linearly related through stimulus presentation lags from 0 to 594 s in the former experiment and for
repeated learning of words in the latter. The Tagging/Retagging interpretation of short term memory is introduced to explain
this linear relationship. At stimulus presentation the words are tagged. This tagging level drops slowly with time. When a
probe word is reintroduced the tagging level has to increase for the word to be properly identified leading to a delay in
response time. The tagging time is related to the meaningfulness of the words used—the more meaningful the word the longer
the tagging time. After stimulus presentation the tagging level drops in a logarithmic fashion to 50% after 10 s and to 20%
after 240 s. The incorrect recall and recognition times saturate in the Rubin et al. data set (they are not linear for large
time lags), suggesting a limited time to search the short term memory structure: the search time for recall of unusual words
is 1.7 s. For recognition of nonsense words the corresponding time is about 0.4 s, similar to the 0.243 s found in Cavanagh

A spatial learning task was used to examine cognitive function in 36 cats (1.0–15.1 years of age), with a control for motor function. No significant age-related decline in cognitive function was found. Initial selection of 75 cats showed no significant age differences between young (0–3 years), adult (3.1–8 years), senior (8.1–12 years) and geriatric cats (12.1–15.1 years) locating food rewards in a holeboard box. Consequently, the senior and geriatric groups were combined because of a concern that age effects may not be seen in the 8–12-year-old cats. Differences between these three age groups of cats to learn the position of three food rewards in an array of 30 otherwise empty (food-scented) positions were non-significant. Differences between age groups remained non-significant for retrieval of rewards from these three positions even when masked with a tissue paper cover. There was no significant effect of age on speed of reaching each criterion. Although the total number of errors was similar, there was a significantly increased proportion of working memory errors (when the cat returned to a previously baited position) and decreased proportion of reference memory errors (when the cat put a nose or paw in an empty position) with increasing age. There was no effect of age on motor function, with success in a plank-crossing test related only to plank width. These findings support previous work suggesting that the cat differs from other species in the way in which it demonstrates age-related declines in cognitive function. Further studies using different testing methodologies are required to assess other cognitive functions that may decline with age. 相似文献

A total of 1347 weaned lambs from eight genotypes were tested over five consecutive years: Romanov (ROM) and Lacaune (LAC) pure breeds, the two F1 crossbreeds (RL and LR) and the offspring of ewes from these four genotypes sired with Berrichon-du-Cher rams (BCF). The lambs were individually exposed to three challenging tests involving novelty, human contact and social isolation. Ten synthetic variables were used to express social reactivity (i.e., active vs. passive strategy), exploratory activity and reactivity to humans. BCF crossbreds were more active (i.e., high bleats, locomotion and attempts to escape) than purebreds and F1. In contrast, ROM expressed more passive responses (i.e., low bleats and vigilance postures) than LAC and BCF crossbreds. In addition, ROM approached a motionless human less and had longer flight distances to an approaching human than did LAC and BCF crossbreds. When restrained, ROM, and to a lesser extent B×ROM and B×LR, avoided human contact more than did LAC, RL and B×LAC. Most of these differences were explained by direct additive genetic effects while maternal influences or heterosis effects were rarely significant. Summary A continuous harvest effort can lead a population to extinction. How an “unconscious” immune system would perpetrate such
an effort in order to eliminate a self-replicating antigen (a pathogen) becomes an intriguing problem if the system responses
are functions of the pathogen population: the responses cannot be a continuous effort as the pathogen vanishes. On theoretical
grounds, we show some qualities an immune response must have to support pathogen elimination. Then, three specific mechanisms
are addressed: a pathogen-independent positive feedback loop among the responding cells of the system (e.g., B-lymphocyte
and T-helper); the persistence of antigen bound to presenting cells; and the programmed expansion/contraction of a pool of
responding cells. The maintenance of responding cells due to these mechanisms is the essential feature to the effective clearance
of self-replicating agents. Thus, evolutionarily, the primary function of a helper lymphocyte would be to amplify a response

Objective: The aim of the study was to assess the uptake of dental services by the old and very old population within the scope of the Berlin Aging Study (Berliner Altersstudie BASE ). Design: A multi‐disciplinary structured interview was performed on 928 subjects, aged from 70 to 103 years of whom 516 persons volunteered to take part in a 14‐session intensive protocol. Six representative study groups were matched for age and gender. Subjects were asked to recall the timing of their most recent dental visit. Data were validated by sending for dental records and compared with all study participants from the multi‐disciplinary intake assessment. Data were related to age group, dental state, dementia and education. Results: Reported last contact with dental services ranged from 2 weeks to 52 years (median 18 months) with a higher time lapse in the study groups aged 85 and older. Dentate subjects had seen their dentist more recently than edentate subjects. Higher education correlated with an increased dental utilisation. Subjective memory on the time lapse since the last dental appointment coincided in 13% of the subjects with available dental records (n=84), was misjudged between one and six months in 55%, and by more than six months in the remainder. Moderately or severely demented subjects who remembered their last dental appointment (n=48 of 70) showed no consistently different utilisation to healthy or mildly demented studs participants. Conclusion: Edentate old and very old subjects show the least frequent utilisation of dental services. Data on motivation and barriers to care are needed to develop strategies to improve the use of dental services and thus promote oral health in late life. 相似文献
